  • The death of Rasputin—A forensic evaluation

      Abstract: Abstract Grigori Yefimovich Rasputin, a confidant of Tsar Nicholas and his wife, was murdered by Prince Yussupov and his co-conspirators in the cellar of the prince’s Moika Palace in St Petersburg, Russia, on the evening of December 30th, 1916 (December 17th in the Russian calendar). The narrative of his death is largely based on Prince Yussupov’s published memoirs and has Rasputin being poisoned with cyanide, shot, bludgeoned, and finally drowned. A review of the available forensic material, however, shows a photograph with a contact gunshot wound to Rasputin’s forehead. This would indicate that he was dead prior to being dropped into the Little Nevka River. His distaste for sweet foods and the absence of poison at autopsy would also suggest that the story of cyanide toxicity was fabricated. Yussupov’s description of Purishkevich firing at Rasputin from a distance as he ran across the courtyard in an attempt to escape would also not be consistent with the post mortem photograph. The simplest version of the events would be that Rasputin was executed by a contact gunshot wound to the forehead when he visited the Yussupov Palace. While it appears that the events of that fateful evening have been embellished, it is certainly not uncommon for perpetrators of homicides to provide histories that are later shown to be at odds with the truth. Re-evaluation of historic cases may provide compelling evidence for alternative interpretations to the popular historic record.

  • A comparison between the enzymatic oxidation method and headspace gas
           chromatography with a flame ionization detector in the determination of
           postmortem blood ethanol

      Abstract: Abstract Ethanol is the most commonly encountered substance in forensic toxicology. Determining blood alcohol concentration (BAC) in autopsies accounts for the majority of work in forensic diagnosis. The most common method to assess BAC is the enzymatic oxidation method because of its low cost, easy operation, and high throughput. Still, the elevated lactate and lactate dehydrogenase (LDH) levels in postmortem blood may affect accuracy. This study uses headspace gas chromatography with a flame ionization detector (HS-GC/FID) to assess the interference of lactate and LDH levels on BAC in 110 autopsied blood samples determined by the enzymatic oxidation method. The results showed that lactate and LDH levels in postmortem blood were higher than in normal blood. There was a weak correlation between the lactate levels and BAC difference (r = 0.23, p < 0.05) and a strong correlation between LDH levels and BAC difference (r = 0.67, p < 0.001). The differentiation of BAC between the enzymatic oxidation method and HS-GC/FID was significant (p < 0.001), confirming the interference significantly. All postmortem blood samples with lactate and LDH levels higher than regular lead to a positive error in determining BAC by enzymatic oxidation method. The study results suggest that the HS-GC/FID method should be used to determine BAC in postmortem blood samples instead of the enzymatic oxidation method to avoid mistakes in forensic diagnosis.

  • Identification of the efficacy of parentage testing based on bi-allelic
           autosomal single nucleotide polymorphism markers in Taiwanese population

      Abstract: Abstract Parentage testing is crucial for forensic DNA analysis, using short tandem repeats (STRs). Single nucleotide polymorphisms (SNPs) with high minor allele frequency (MAF) are promising for human identification. This study aimed to develop SNP markers for parentage testing in the Taiwanese population and compare their accuracy with STRs. The TPMv1 SNP microarray (714,457 SNPs) was used to screen 180,000 Taiwanese individuals and analyze the SNP data using PLINK. After quality control, allelic distribution, and MAF considerations, a set of SNPs with significant inheritance information was selected. Parentage testing was conducted on 355 single parent-child pairs using both STRs and SNPs, employing three kinship algorithms: identity by descent, kinship-based inference for genome-wide association studies, and the combined paternity index/probability of paternity (CPI/PP). An Affymetrix signature probe for kinship testing (ASP) was also used. Based on the quality control and selection criteria, 176 SNPs with MAF > 0.4995 were selected from the Taiwanese population. The CPI/PP results calculated using SNPs were consistent with the STR results. The accuracy of the SNPs used in the single-parent-child parentage testing was > 99.99%. The set of 176 SNPs had a higher identification rate in the single parent-child parentage test than in the ASP. The CPI/PP value calculated using 176 SNPs was also more accurate than that calculated using ASP. Our findings suggest that these 176 SNPs could be used for single-parent-child parentage identification in the Taiwanese population.

  • Study of secondary dentine deposition in central incisors as an age

      Abstract: Abstract This work aimed to assess the pulp/tooth area ratio’s utility in the upper central incisors using orthopantomograms. A convenience sample of 801 adult patient orthopantomograms was studied. Image J® software was used to measure the pulp/tooth area ratio, and a regression model was developed. Our results conclude that the methodology assessing upper incisors’ pulp/tooth area ratio using orthopantomograms can lead to age overestimation and statistically significant differences between chronological and estimated age. For those over 50, no correlation between pulp/tooth area ratio and chronological age was found, suggesting that this may be the upper limit of this technique in this population. This methodology may not be suitable for age estimation, particularly in older adults.

  • On the forensic relevance of tattoos: distinguishing black inks with
           energy dispersive spectroscopy and backscattered scanning electron
           microscopy

      Abstract: Purpose This study aimed to analyze black tattoo inks by means of energy dispersive spectroscopy and backscattered scanning electron microscopy. Methods The sample consisted of five types of commercial tattoo pigments of the black colour (Easy Glow™, Electric Ink™, Iron Works™, Master Ink™, and Viper™). An Energy Dispersive Spectroscopy (EDS) detector (Silicon Drift Detector – SDD – type) attached to a Scanning Electron Microscope (SEM) device (Tescan Vega3 LMU, Libusina, Czech Republic) was used. X-ray characteristic signs were detected for each tattoo ink in an interval between 0 and 2.5 keV. The electron acceleration potential in the microscope was 15 keV. Two regions were analyzed for each sample (n = 10). On each region, a micrography of backscattered electrons (BSE) was obtained. Means and standard deviations (SD) of the weight percentages (Wt%) were calculated. Results C and O were predominant, with a mean O/C ratio between 2.69 and 2.74 Wt%. Electric Ink and Master Ink were the most similar pigments, while Easy Glow was the most distinctive — with agglomerates of Al that had a concentration 25 × higher than other specimens. Other compounds detected in the sample were Cl and Cu. Conclusion EDS and SEM were efficient to distinguish black tattoo inks. These are our preliminary outcomes on the use of EDS and SEM to analyze black tattoo inks. Thus, careful interpretation is necessary to avoid rash applications in human identification practice.

  • A rare case of type II tri-allelic inheritance at vWA, SE33, D8S1179, and
           D13S317 loci demonstrated by STR analysis in paternity testing

      Abstract: Abstract Short tandem repeat (STR) typing has been regularly used in paternity disputes and forensic human identification linked caseworks. Occasionally, forensic scientists come across aberrant allele patterns during STR typing because of mutations, genetic variations, and other abnormalities. The tri-allelic pattern of STR is rare, particularly, the case where this pattern exists at 4 loci. Here, we report the type II tri-allelic patterns observed at vWA, SE33, D8S1179, and D13S317 loci in the product of conception (POC) sample during the course of our regular paternity case investigation. The DNA extracted from the blood samples and tissue of POC were subjected to STR typing for autosomal and sex STR loci using the commercial QIAGEN’s Investigator® IDplex Plus Kit and QIAGEN’s Investigator® 24plex QS Kit. Capillary electrophoresis was carried out in 3500 and 3500xL Genetic Analyzer Applied Biosystems and genotyped using GeneMapper ID-X Software v1.5 and v1.6. In this case of paternity inclusion, the POC sample displayed type II tri-allelic patterns at vWA (16, 19, 20), SE33 (19, 28.2, 29.2), D13S317 (16, 19, 20), and D8S1179 (10, 13, 17) loci. In addition, the POC displayed an abnormal genotype with a heterozygous peak imbalance (type II-B) of (1:2) pattern at D3S1358, D21S11, and D16S539 loci, of (2:1) pattern at D1S1656, D12S391, D10S1248, D2S1338, D2S441, D18S317, FGA, CSF1PO, and D5S818 loci, and type II-C allelic pattern (one single peak with triplicate height) at D19S433 and DS7820 loci. Understanding of such anomalous genotypes improves the knowledge about tri-allelic pattern of CODIS loci and helps in the appropriate interpretation of the results in STR typing.

  • Genital lichen sclerosus misdiagnosis: Forensic insights

      Abstract: Abstract In this case study, we present an instance of genital lichen sclerosus et atrophicus that was initially misdiagnosed as child sexual abuse. A 6-year-old female presented to the emergency department with complaints of vaginal bleeding accompanied by pruritus. The initial suspicion of child abuse was prompted by the presence of blood. However, a thorough physical examination conducted by the forensic department ruled out any traumatic injuries and instead suggested an underlying pathology, ultimately diagnosed as lichen sclerosus et atrophicus. This case report underscores the vital role of forensics in excluding abuse as a cause in similar presentations, guiding the case towards a more accurate diagnosis. Additionally, it emphasizes the significance of possessing a comprehensive understanding of pathologies that can mimic abusive injuries, thereby preventing the potential consequences of an incorrect abuse diagnosis.

  • Forensically relevant anatomical brain regions cannot be
           sub-differentiated by RNA expression analysis

      Abstract: Abstract The contextualization of biological traces generated by severe head injuries can be beneficial for criminal investigations. Here we aimed to identify and validate mRNA candidates for a robust sub-differentiation of forensically and traumatologically relevant brain regions. To this purpose, massively parallel sequencing of whole transcriptomes in sample material taken from four different areas of the cerebral cortex (frontal, temporal, parietal, occipital lobe) was performed, followed by bioinformatical data analysis, classification, and biostatistical candidate selection. Candidates were evaluated by Multiplex-RT-PCR and capillary electrophoresis. Only a weak relative upregulation and solely for candidates expressed in the parietal lobe was observed. Two candidates with upregulation in the cerebellar region (PVALB and CDR2L) were chosen for further investigation; however, PVALB could not reliably and repeatedly be detected in any lobe whereas CDR2L was detectable in all lobes. Consequently, we suggest that differences in mRNA expression between four regions of the cerebral cortex are too small and less pronounced to be useful for and applicable in forensic RNA analysis. We conclude that sub-differentiation of these brain regions via RNA expression analysis is generally not feasible within a forensic scope.

      Abstract: Abstract The world’s forensic entomologists have much in common. They face similar research challenges, apply the same scientific methodology, study the same kinds of evidence, and access global research databases. Nevertheless, some regional heterogeneity inevitably exists. For most countries, and particularly those that have complex and diverse ecosystems, the current priority is to stimulate use of forensic entomology by establishing open access databases with time series data using standardised protocols for occurrence, abundance, distribution, niche preferences, life cycle, and identification characteristics for the key regional species that may be encountered in forensic entomology cases. Even in countries where forensic entomology is routinely used as a tool in reconstructing the history of corpses found on crime scenes in accordance with principles found in the rapidly developing body of literature, there is still much to learn about forensically useful insects. Examples of regional gaps include the taxonomy of lesser-known carrion insect species, seasonal occurrence of carrion species in the country’s various geographical regions, and rates of development of the local species modeled in the many site situations of forensic interest. The first published study of carrion insects found in Athens, Greece published in this journal is an example of entomologists in a region taking the necessary first step towards establishing baseline data about native and introduced species and hence, physiological and behavioural responses to local environmental conditions, including life cycles and likelihood of occurrence or absence in the region.

  • Short-term postmortem interval estimation by detection of
           apoptosis-related protein in skin

      Abstract: Abstract Time-of-death extrapolation has always been one of the most important issues in forensic practice. For a complicated case in which a corpse is destroyed with little evidence, judging the time of death of the deceased is a major challenge, which also enables criminals to escape legal sanctions. To find a method to roughly judge the time of death of a corpse with only a small amount of skin tissue, in this study, we established an early death model by using mice; furthermore, the postmortem interval was estimated by determining the protein and mRNA levels of Bax and Bcl-2 in the skin. In this process, 0 h after death was used as the control group, and the expression levels of Bax and Caspase-3 reached the maximum value at 8–12 h, while Bcl-2, as an inhibitor of apoptosis protein, peaked after 24 h. The mRNA expression levels of related proteins in postmortem skin tissues were also different. The results of these data indicate that the protein and mRNA levels of Bax and Bcl-2 in the skin have potential application in early time-of-death estimation.

  • Death and injuries caused by cattle: A forensic overview

      Abstract: Abstract Lethal episodes arising from interactions with cattle may be the result of a response of the animals to simple handling and herding, or from reactions to deliberate provocation or goading intended to incite aggressive behavior for public entertainment purposes. Deaths may be considered to be unprovoked and unanticipated, or provoked and predictable. Cattle cause significant numbers of deaths globally and are considered the most dangerous large animal in Britain. Behavior may be unpredictable even in apparently docile domesticated animals, and attacks may be by a single animal or a herd and result in injuries from kicking, head/butting/charging, stomping, goring, and crushing. Craniofacial injuries may involve fractures of the spine or skull with cerebral contusions and lacerations associated with subarachnoid, subdural, and extradural hemorrhages. Chest injuries are also characterized by fractures which may be multiple with flail chest, hemo- and pneumothoraces, and organ disruption. Injuries to the abdomen and perineum include intestinal perforations, splenic rupture, perineal and vaginal tears, urethral lacerations and avulsions, and bladder and rectal perforations. Significant vascular injuries include complete and partial transections and lacerations. Males living in rural areas are most at risk of a fatal encounter.

  • Application of the Kvaal method to CBCT reconstructed panoramic images for
           age estimation

      Abstract: Abstract As the teeth are more durable than other parts of the skeleton, they provide valuable data for age estimation. Age estimation from adult teeth is mainly based on secondary dentin production. The present study aimed to devise a regression formula for age estimation specific to the Anatolian population using the Kvaal method on CBCT reconstructed panoramic images. In total, 201 individuals aged between 20 and 69 were divided into two groups: data from the study group (n = 101) were used to create the regression formulae, and data from the control group (n = 100) were used to test the formulae. Pearson’s correlation coefficients and linear regression analyses were performed. Maxillary teeth provided more accurate age estimates than mandibular teeth. The regression formulae derived in this study are found to be statistically applicable and reasonably accurate. However, these results should be interpreted with caution.

      Abstract: Abstract This commentary examines the use of artificial intelligence (AI) in forensic science, highlighting its benefits in enhancing accuracy and efficiency across a number of forensic disciplines, including medicine, anthropology, forensics, and taphonomy. However its use, also raises concerns about privacy, data protection, bias, fairness, and the reliability of AI systems. The commentary emphasises the importance of scrutiny, standardized procedures, and ongoing dialogue to ensure AI is responsibly advanced in forensic science.

  • Anaphylaxis as probable cause of death in a rare case of fatal Vipera
           berus bite

      Abstract: Abstract This case report describes the death of a 52-year-old male who was bitten by a venomous snake, Vipera berus (common European adder), on his left wrist. Despite experiencing symptoms, the patient refused medical care and self-medicated with alcohol instead. He was later found dead in his residence. Autopsy and histological examination revealed evidence of an anaphylactic reaction in response to the snake bite, with additional findings of alcohol intoxication and other underlying medical conditions.

  • A retrospective review of methylamphetamine detected in child deaths
           reported to the Victorian Coroner, Australia

      Abstract: Abstract This study investigated methylamphetamine (MA) exposures in the deaths of children (≤ 12 years old) reported to the Coroner in the state of Victoria, Australia, between 2011 and 2020. Demographics, autopsy findings including the cause of death, self-reported prenatal or caregiver drug use, child protection services information, and toxicological findings were summarized by descriptive statistics. Validated methods of liquid chromatography-tandem mass spectrometry were used in the analysis of drugs. There were 50 child deaths with MA detected in blood, urine, and/or hair with 64% (n = 32) identified in 2018–2020. Most children were 1–365 days old (66%, n = 33) and the cause of death was unascertained in 62% (n = 31) of cases. MA was toxicologically confirmed in hair (94%, n = 47) significantly more than blood (18%, n = 9). Prenatal or caregiver drug use was self-reported in 44% (n = 22) and 42% (n = 21) of cases, respectively. Moreover, only 54% (n = 27) of deceased children were a child protection client at their time of death. These findings suggest the number of deceased children exposed to MA has increased over the past 10 years, which is consistent with the greater supply of crystal MA in the Australian community. Hair analysis provided additional means to identify cases that were unknown to child protection services and may have implications for other children in the same drug exposure environment.

  • Modeling of inflicted head injury by shaking trauma in children: what can
           we learn'

      Abstract: Abstract Inflicted shaking trauma can cause injury in infants, but exact injury mechanisms remain unclear. Controversy exists, particularly in courts, whether additional causes such as impact are required to produce injuries found in cases of (suspected) shaking. Publication rates of studies on animal and biomechanical models of inflicted head injury by shaking trauma (IHI-ST) in infants continue rising. Dissention on the topic, combined with its legal relevance, makes maintaining an up-to-date, clear and accessible overview of the current knowledge-base on IHI-ST essential. The current work reviews recent (2017–2023) studies using models of IHI-ST, serving as an update to two previously published reviews. A systematic review was conducted in Scopus and PubMed for articles using animal, physical and mathematical models for IHI-ST. Using the PRISMA methodology, two researchers independently screened the publications. Two, five, and ten publications were included on animal, physical, and mathematical models of IHI-ST, respectively. Both animal model studies used rodents. It is unknown to what degree these can accurately represent IHI-ST. Physical models were used mostly to investigate gross head-kinematics during shaking. Most mathematical models were used to study local effects on the eye and the head’s internal structures. All injury thresholds and material properties used were based on scaled adult or animal data. Shaking motions used as inputs for animal, physical and mathematical models were mostly greatly simplified. Future research should focus on using more accurate shaking inputs for models, and on developing or and validating accurate injury thresholds applicable for shaking.

  • Death due to sickle cell crisis: a case report

      Abstract: Abstract Sickle cell disease (SCD) is the most common hereditary hemoglobinopathy worldwide. It results in characteristic acute and chronic findings on postmortem computed tomography (PMCT), macroscopic and microscopic examinations. While the diagnostic imaging and macroscopic features are not specific for SCD on their own, when coupled with microscopic features such as sickled erythrocytes and evidence of chronic venous congestion (i.e., Gamna-Gandy bodies), these clues can help alert forensic pathologists to the presence of SCD. Despite the prevalence of the disease and the constellation of findings alluded to above, SCD is not often explored in forensic pathology literature. This case demonstrates classic acute and chronic features of SCD on PMCT, macroscopic and microscopic examinations. It explores the pathophysiology leading to sudden and unexpected death in a person with SCD and possible pitfalls in attribution of cause of death.

  • The integration and implications of artificial intelligence in forensic
           science

      Abstract: Abstract This commentary explores the integration of artificial intelligence (AI) in forensic science and its potential implications. The applications of AI in forensic disciplines such as medicine, forensic anthropology, digital forensics, and taphonomy have enhanced the accuracy and efficiency of identification processes and the analysis of digital evidence. However, this rapid advancement prompts critical considerations in privacy, data protection, bias and fairness, and the accuracy and reliability of AI systems. The inherent challenges of the “black box” nature of AI algorithms call for transparency and accountability to maintain trust and uphold the integrity of forensic investigations. Ethical use, legal compliance, interdisciplinary collaboration, education, data integrity, standardization, human oversight, and societal impact, along with sustainability are identified as pivotal areas requiring urgent attention. The discussion underscores the need for rigorous scrutiny, standardized operating procedures, and proactive dialogue to ensure the responsible advancement of AI in forensic science.
