  • Comparación de ELISA indirecto en suero sanguíneo versus ELISA indirecto
           en leche, para el diagnóstico de paratuberculosis en bovinos de
           tamborealizada en la provincia de Buenos Aires, Argentina

    • Abstract: Resumen: El objetivo de este trabajo fue comparar las características operativas de un ELISA en suero, con un ELISA en leche, para el diagnóstico de paratuberculosis bovina. Utilizando análisis ROC, el área bajo la curva fue de 0,84 para ELISA en suero y 0,88 para ELISA en leche, sin diferencias significativas (p= 0,30) entre ambas AUC; la sensibilidad fue de 91,7% y 87,5%, la especificidad de 82,0% y 86,5% para ELISA en suero y ELISA en leche respectivamente. También se consideraron como variables, los días en lactancia y el número de lactancias. Los días en lactancia se analizaron con correlación y regresión lineal simple, evidenciando una relación positiva (p <0,05), entre ELISA en suero y ELISA en leche. Los números de lactancias se analizaron mediante ANOVA solo en ELISA en leche, con diferencias significativas (p= 0,016) entre el grupo de 1era, con los de 2da y >2da lactancias. Los resultados preliminares obtenidos, demostraron que el ELISA en suero presenta características operativas similares al ELISA en leche, siendo el ELISA en leche una alternativa práctica y menos invasiva al muestreo.Abstract: The aim of this paper was to compare the operational characteristics of serum ELISA with milk ELISA, for the diagnosis of bovine paratuberculosis. Using ROC analysis, the area under the curves were 0.84 in serum ELISA and 0.88 in milk ELISA, without significant differences (p= 0.30) between both AUC; the sensitivities were 91.7% and 87.5%, the specificities 82.0% and 86.5% for serum ELISA and milk ELISA respectively. In addition, days in lactation and lactation numbers were also included as variables. Days in lactation were analyzed with correlation and simple linear regression, showing a positive relationship (p < 0.05) between serum ELISA and milk ELISA. Lactations numbers were analyzed only in milk ELISA with ANOVA, significant differences (p= 0.016) were evident between 1st groups with 2nd and> 2nd lactations. The preliminary results demonstrate that serum ELISA presents similar operational characteristics to milk ELISA, being milk ELISA a practical and less invasive alternative to sampling.
       
  • Diagnóstico molecular de Neospora caninum en fetos abortados
           espontáneamente en bovinos de Uruguay

    • Abstract: Resumen: En Uruguay el diagnóstico de rutina de Neospora caninum se realiza fundamentalmente por estudios histopatológicos de los fetos abortados, complementando esta información con resultados de serología y/o Inmunohistoquímica (IHQ). El objetivo de este estudio fue estandarizar un protocolo de amplificación de secuencias mediante PCR (polymerase chain reaction) para la detección de Neospora caninum en órganos de fetos abortados espontáneamente en bovinos para su incorporación a la rutina diagnóstica. Con este propósito se realizó un estudio retrospectivo utilizando muestras congeladas de archivo, de un periodo comprendido entre los años 2013 y 2016, con diagnóstico positivo o negativo a Neospora caninum. Los resultados mostraron que de 31 muestras positivas a Neospora caninum por las técnicas de rutina, 26 fueron positivas por PCR (84%), demostrando una alta sensibilidad de diagnóstico de esta técnica. Todas las muestras con diagnóstico negativo mediante histopatología (n=20) fueron confirmadas como negativo mediante PCR. La comparación de los resultados de PCR con los de histopatología, mediante el test de Cohen’s Kappa, señalaron un acuerdo muy alto (81%, p<0.001) entre ambos diagnósticos. Dichos resultados sumados a la simplicidad y rapidez de la técnica podrían sugerir a la PCR como alternativo de la IHQ como técnica confirmatoria en el diagnóstico de rutina. También permitiría realizar análisis retrospectivos de muestras archivadas, paso fundamental para futuros estudios genéticos de la población de Neospora caninum presente en Uruguay.Abstract: The diagnosis of Neospora caninum in Uruguay is carried out by histopathological studies of aborted fetuses, complementing this information with serology test and/or immunohistochemistry (IHC) results. The aim of this study was to standardize a protocol of polymerase chain reaction (PCR) to detect of N. caninum in organs of spontaneously aborted bovine fetus. A retrospective study was conducted using frozen samples from a period between 2013 and 2016, with positive or negative diagnosis of N. caninum. Results showed that 26 of 31 samples positives for N. caninum by routine techniques were positive by PCR, demonstrating a high sensitivity of the PCR. All negative samples by Histopathology (n=20) were confirmed as negative by PCR. Cohen's Kappa test indicated a very high agreement (81%, p<0.001) between PCR and Histopathology with serology test and/or immunohistochemistry (IHC) results. The high sensitivity, specificity and simplicity of PCR test suggest this technique as an alternative to IHC as a confirmatory technique in routine diagnosis.
       
  • Proteína de membrana externa CD de Moraxella bovis como antígeno
           potencial contra la queratoconjuntivitis infecciosa bovina

    • Abstract: Abstract: Infectious bovine keratoconjunctivitis (IBK) is a severe ocular disease that affects cattle and has a significant economic impact worldwide. The principal etiological agent of IBK is Moraxella bovis. Another species, Moraxella bovoculi, has also been reported as a possible etiological agent. Due to the health problems caused by IBK, it is necessary to develop effective vaccines to prevent the disease. Outer membrane protein-CD (OmpCD) has exhibited potential as a protective antigen in related species such as Moraxella catarrhalis. The objective of this study was to produce a recombinant M. bovis OmpCD and evaluate its immunogenicity in CD-1 mice. The ompCD gene was amplified in all of the M. bovis and M. bovoculi strains studied, and sequence alignment revealed highly conserved regions. The reactivity of M. bovis recombinant OmpCD was seen when sera and tears of diseased bovines were used in Western blots. CD-1 mice were immunized and the antibody response was analyzed in serum and bronchoalveolar lavage (BAL) fluid. Cytokine levels in the BAL fluid were also determined. The trials in mice allowed us to demonstrate that OmpCD administered systemically induced a significant increase in sera IgG specific antibodies while the intranasal immunization induced a significant increase in IgA and IL-4 levels in BAL. Moreover, the combination of OmpCD and a mucosal adjuvant significantly increased the IgA response in BAL. These results encourage further studies to assess the role of OmpCD as a possible vaccine candidate against IBK, using challenge models in cattle.Resumen: La Queratoconjuntivitis Infecciosa Bovina (QIB) es una enfermedad ocular grave que afecta frecuentemente al ganado y que tiene un impacto económico significativo mundialmente. Los agentes etiológicos asociados a la QIB son principalmente Moraxella bovis y Moraxella bovoculi. El problema sanitario originado por la QIB ha llevado a procurar desarrollar vacunas efectivas para prevenir la enfermedad. La proteína de membrana externa CD (Omp-CD), ha exhibido potencial como antígeno protector en especies relacionadas como Moraxella catharralis. El objetivo de este estudio fue producir una proteína recombinante OmpCD de M. bovis y evaluar su inmunogenicidad en ratones CD-1. El gen ompCD se amplificó en todas las cepas de M. bovis y M. bovoculi estudiadas, y el alineamiento de secuencias reveló regiones altamente conservadas. La reactividad de OmpCD recombinante de M. bovis se observó cuando se utilizaron sueros y lágrimas de bovinos enfermos por Western blot. Se inmunizaron ratones CD-1 y se analizó la respuesta de anticuerpos en suero y líquido de lavado broncoalveolar (BAL). También se determinaron los niveles de citoquinas en el líquido del BAL. Los ensayos en ratones permitieron demostrar que la OmpCD administrada sistémicamente inducía un aumento significativo de anticuerpos específicos IgG en suero mientras que la inmunización intranasal inducía un aumento significativo de los niveles de IgA e IL-4 en el BAL. Además, la combinación de OmpCD y un adyuvante de mucosas aumentaron significativamente la respuesta de IgA en BAL. Estos resultados estimulan la realización de estudios adicionales para evaluar el papel de OmpCD como posible candidato a vacuna contra QIB, utilizando modelos de desafío en bovinos.
       
  • Utilidad del proteinograma electroforético capilar en el diagnóstico de
           la Peritonitis Infecciosa Felina (PIF) no efusiva: estudio de caso

    • Abstract: Resumen: La peritonitis infecciosa felina (PIF) es una de las enfermedades infecciosas con mayor tasa de mortalidad entre la población felina y puede presentarse en forma efusiva o no efusiva. El diagnóstico de PIF en su forma no efusiva es un gran desafío y se basa en la sintomatología clínica, detección viral y en los hallazgos de laboratorio. En este sentido, la relación albúmina/globulina y el aumento en la fracción de γ-globulinas obtenida mediante proteinograma electroforético (PEF) son indicadores importantes y complementarios para el diagnóstico de la PIF. El objetivo del presente trabajo fue utilizar por primera vez el PEF como herramienta diagnóstica de un caso clínico de un felino con diagnóstico clínico de PIF no efusivo, donde la realización un PEF junto a estudios complementarios de rutina permitió arribar a un diagnóstico definitivo. El resultado del PEF evidenció un pico policlonal en las γ globulinas, una concentración de albúmina sérica disminuida y una relación albúmina/globulina de 0,23. Basados en estos resultados, se discute cómo el PEF principalmente en la forma no efusiva de la PIF puede apoyar el diagnostico de esta enfermedad, permitiendo emitir un pronóstico preciso y contribuir con el veterinario clínico en la toma de decisiones terapéuticas.Abstract: Feline infectious peritonitis (PIF) is an infectious disease which has a high mortality rate compared to other infectious diseases among feline population. It clinically manifested under two forms, effusive or non-effusive. PIF’s diagnosis, mainly the non-effusive form, is a clinical challenge based on symptoms, viral detection and laboratory findings. In this regard, the albumin/globulin ratio and the increase on the γ globulin fraction of the capillary zone electrophoresis (PEF) are important and complementary indicators for the antemortem diagnosis of PIF. This job’s objective was to exhibit a feline clinical case clinically diagnosed with non-effusive PIF in which the PEF, along with other complementary routine tests, allowed to reach a definitive diagnosis. PEF’s results showed a polyclonal peak on the γ globulin fraction, a decreased serum albumin concentration and an albumin/globulin ratio of 0,23. Based on these results we discuss how the PEF, mainly on the non-effusive form of PIF, can support this disease diagnosis allowing to dictate an accurate prognosis and helping the clinical veterinarian on therapeutic decisions.
       
  • Detección en la conjuntiva y el oído externo de especies potencialmente
           patógenas del género Mycoplasma en ovinos

    • Abstract: Resumen Los micoplasmas son responsables de varias infecciones en pequeños rumiantes. En Brasil se han detectado micoplasmas potencialmente patógenas en el conducto auditivo externo sano de caprinos y bovinos y en asociación con ácaros, pero nunca se han detectado en ovinos. La presente investigación tuvo como objetivo evaluar la conjuntiva y el oído externo como fuente de infección silenciosa de micoplasmas en ovinos. Se obtuvieron muestras de frotis del conducto auditivo externo y ocular de cuarenta y cinco ovinos (machos y hembras). Fueron realizadas las pruebas de Detección molecular (PCR) y aislamiento de Mycoplasma spp. Se determinó la concentración mínima inhibitoria para el aislamiento, utilizando oxitetraciclina, cloranfenicol, tilosina y ácido nalidíxico. Fue aislado Mycoplasma conjunctivae a partir de la conjuntiva ocular de un ovino sano, además de la detección molecular de M. agalactiae y M. mycoides subsp. capri en ambos sitios anatómicos de ovinos del Estado de São Paulo, Brasil. El análisis filogenético reveló proximidad y similitud con un aislado de ovino y de cabra estadounidenses. La conjuntiva ocular y el oído externo de los ovinos son sitios anatómicos que pueden portar micoplasmas patógenos provocando infecciones a ovinos vulnerables.Summary: Mycoplasmas are responsible for several infections in small ruminants. Potentially pathogenic mycoplasmas have already been detected in the external ear canal of Brazilian healthy goats and cattle, and in association with mites, but not in sheep. The present research aimed to evaluate the role of the conjunctiva and external ear as a silent source of micoplasmas in sheep. Ocular and external ear canal swab samples were obtained from forty-five male and female sheep. Molecular detection (PCR) and isolation of Mycoplasma spp. were performed. Minimum inhibitory concentration was performed using oxytetracycline, tylosin, chloramphenicol and nalidixic acid. We describe the isolation of Mycoplasma conjunctivae fromthe ocular conjunctivae of a healthy sheep, besides the molecular detection of M. agalactiae and M. mycoides subsp. capri in both anatomical sites in sheep from the State of São Paulo. Phylogenetic analysis revealed proximity and similarity with an American sheep and goat isolates. The ocular conjunctiva and the external ear of sheep are keepers of potentially pathogenic veterinary mycoplasmas which can infect vulnerable sheep.Resumo: Os micoplasmas são responsáveis por várias infecções em pequenos ruminantes. Micoplasmas potencialmente patogênicos já foram detectados no conduto auditivo externo de caprinos e bovinos brasileiros saudáveis e em associação com ácaros, mas não em ovinos. A presente pesquisa teve como objetivo avaliar o papel da conjuntiva e da orelha externa como fonte silenciosa de micoplasmas em ovinos. Amostras de esfregaço ocular e do canal auditivo externo foram obtidas de 45 ovelhas machos e fêmeas. Detecção molecular (PCR) e isolamento de Mycoplasma spp. foram realizados. A concentração inibitória mínima foi realizada com oxitetraciclina, tilosina, cloranfenicol e ácido nalidíxico. Descrevemos o isolamento de Mycoplasma conjunctivae da conjuntiva ocular de uma ovelha saudável, além da detecção molecular de M. agalactiae e M. mycoides subsp. capri em ambos os sítios anatômicos em ovinos do Estado de São Paulo. A análise filogenética revelou proximidade e similaridade com isolados de ovinos e caprinos americanos. A conjuntiva ocular e a orelha externa das ovelhas são detentoras de micoplasmas veterinários potencialmente patogênicos que podem infectar ovelhas vulneráveis.
