  • Survival of Azotobacter and other microbial group in dry soil by

    • Abstract: Resumen En suelos secos la supervivencia del género y especies de Azotobacter, es paralela a la de otros grupos microbianos nativos como actinomicetos y hongos de ese ambiente, dependiente de la capacidad de adaptación genética de cada uno, en combinación con algunas de las propiedades fisicoquímicas del suelo, y de las condiciones ambientales de almacén. Los objetivos de este trabajo fueron: i) analizar la supervivencia de Azotobacter en suelo recién colectado y almacenado por 11 y 30 años ii) caracterizar bioquímicamente Azotobacter vinelandii aislado estos suelos iii) dinámica de supervivencia de actinomicetos, bacterias y hongos en los suelos. Para cual cada uno de los suelos se conservó por distinto periodo de tiempo en contenedores de vidrio estériles a temperatura ambiente, de ahí diluyeron para cuantificar cada grupo en: agar nutritivo para bacterias, agar de caseína almidón para actinomicetos, agar Rosa de Bengala para hongos y agar Burk para Azotobacter. Los resultados mostraron que el número de Azotobacter supervivientes en el suelo almacenado por 30 años fue de 12 x 106 UFC/g, de 52 x 106 UFC/g en suelo seco por 11 años y de 300 x 106 UFC/g de suelo seco recién colectado. En el suelo de 30, 11 años se detectó una densidad de Azotobacter spp., inferior al suelo seco recién colectado. Se encontró diferencia en los perfiles bioquímicos de A. vinelandii de suelo almacenado por 30 años, en comparación el mismo género y especie en el recién colectado. Lo que anterior indica que las propiedades fisicoquímicas del suelo, el periodo de desecación fue crítico en la supervivencia de Azotobacter y de los otros grupos de microorganismos nativos de ese ambiente.Abstract In dry soils, the survival of the genus and species of Azotobacter is parallel to that of other native microbial groups such as actinomycetes and fungi of that environment, depending on the genetic adaptation capacity of each one, in combination with some of the physicochemical properties of the soil. soil, and environmental storage conditions. The objectives of this work were: i) to analyze the survival of Azotobacter in soil recently collected and stored for 11 and 30 years ii) to biochemically characterize Azotobacter vinelandii isolated from these soils iii) survival dynamics of actinomycetes, bacteria and fungi in soils. For which each of the soils was kept for a different period of time in sterile glass containers at room temperature, from there they were diluted to quantify each group in: nutrient agar for bacteria, casein starch agar for actinomycetes, Rose Bengal agar for fungi and Burk agar for Azotobacter. The results showed that the number of surviving Azotobacter in soil stored for 30 years was 12 x106 CFU/g, 52 x 106 CFU/g in dry soil for 11 years, and 300 x106 CFU/g in freshly collected dry soil. In the soil of 30, 11 years, a density of Azotobacter was detected, lower than the dry soil recently collected. A difference was found in the biochemical profiles of A. vinelandii from soil stored for 30 years, compared to the same genus and species in the newly collected soil. The above indicates that the physicochemical properties of the soil, the drying period was critical in the survival of Azotobacter and other groups of microorganisms native to that environment.
  • Detection of Aspergillus fumigatus in Hordeum vulgare marked at Morelia,
           Mich, México with potential to synthetize ochratoxin A

    • Abstract: Resumen Los granos de Hordeum vulgare o Hv (cebada) comercializados en la ciudad de Morelia, Michoacán, México como alimentos o bebida (cerveza artesanal) de humanos y animales, pueden contaminarse con propágulos de Aspergillus cuando Hv se cultiva mediante el sistema agrícola intensivo, basado en la aplicación de fertilizantes químicos y plaguicidas. Los granos de Hv transportados, almacenados y comercializados en condiciones inadecuadas en Morelia, permiten el crecimiento de géneros y especies de Aspergillus que potencialmente sintetizan micotoxinas como la ocratoxina A. Los objetivos de esta investigación fueron i) analizar la densidad de Aspergillus sp en granos de Hv, comercializado en Morelia, Mich, México, ii) Demostrar la capacidad potencial de Aspergillus aislados de Hv en agar dextrosa Sabouraud (ADS) para sintetizar ocratoxina A. Por lo cual, granos de Hv colectados en Morelia, Mich, se usaron para determinar la densidad y diversidad especies de Aspergillus. Como la capacidad potencial de la principal especie encontrada: A. fumigatus en ADS para la síntesis de ocratoxina A. Todos los datos experimentales se analizaron por ANOVA/Tukey. Los resultados indican un relativo alto número de propágulos de A. fumigatus que contaminan los granos de Hv comercializados en Morelia, donde el 67 % de los aislamientos de A. fumigatus mostraron potencial para sintetizar ocratoxina A en ADS, dado que la composición química del ADS suplió la demanda nutricional de A. fumigatus. Se concluye que existe un evidente riesgo para los humanos y animales que consumen cebada contaminada por A. fumigatus que potencialmente generan ocratoxina A, por lo que se requiere atención sanitaria en el manejo y comercialización de Hv en la ciudad de Morelia.Abstract The grains of Hordeum vulgare or Hv (barley) marketed in the city of Morelia, Michoacán, Mexico as food or beverage (craft beer) for humans and animals, can be contaminated with propagules of Aspergillus. When Hv is grown through an intensive agricultural system, based on the application of chemical fertilizers and pesticides. The Hv grains transported, stored and marketed under inappropriate conditions in Morelia, allow the growth of Aspergillus genera and species that potentially synthesize mycotoxins such as ochratoxin A. The objectives of this research were i) to analyze the density of Aspergillus in grains of Hv, commercialized in Morelia, Mich, Mexico, ii) Demonstrate the potential capacity of A. fumigatus isolated from Hv in Sabouraud dextrose agar (SDA) to synthesize ochratoxin A. Therefore, Hv grains collected in Morelia, Mich was used to determine the density and diversity of Aspergillus species. As the potential capacity of the main species isolated: A. fumigatus in SDA for the synthesis of ochratoxin A was analyzed without no problem since chemical composition of SDA was enough for nutritional demand for A. fumigatus. All experimental data were analyzed by ANOVA/Tukey. The results indicate a relatively high number of propagules of A. fumigatus that contaminate the grains of Hv commercialized in Morelia, where 67 % of the isolates of A. fumigatus showed the potential to synthesize ochratoxin A in SDA. It is concluded that there is an evident risk for humans and animals that consume barley (Hv) contaminated by A. fumigatus that potentially generate ochratoxin A, for which health care is required in the handling and commercialization of Hv, in Morelia City.
  • Saccharomyces exiguus uses kerosene as a source of carbon and

    • Abstract: Resumen El queroseno es un combustible derivado del petróleo, una mezcla hidrocarburos alifáticos y aromáticos, que eventualmente causan contaminación ambiental. En la naturaleza existen géneros y especies de microorganismos heterotróficos aerobios, nativos de todo ambiente, que tienen la capacidad potencial para degradar queroseno como algunos géneros y especies de levadura, para sintetizar proteína de origen unicelular o bien para bioaumentación del impacto negativo ambiental del queroseno, por lo anterior. El objetivo de este trabajo fue analizar la capacidad de Saccharomyces exiguus para utilizar queroseno como fuente de carbono y energía. Para ello, S. exiguus se aisló de pozos petroleros, se creció en queroseno al 5 % con NH4Cl al 1.2 % y 50 ppm de extracto de levadura. El crecimiento de S. exiguus en queroseno se analizó mediante las variables respuesta: peso seco, cuantificación de proteínas y por cromatografía de gases se demostró la utilización de los componentes del queroseno como fuente de carbono y energía. Los resultados mostraron que S. exiguus puede de utilizar los hidrocarburos alifáticos y aromáticos del queroseno como única fuente de carbono y energía, este potencial es aplicable para sintetizar proteína unicelular o en la recuperación de ambientes impactados por queroseno.Abstract Kerosene is a fuel derived from petroleum, a mixture of aliphatic and aromatic hydrocarbons, which eventually cause environmental pollution. In nature, there are genera and species of aerobic heterotrophic microorganisms, native to all environments, that have the potential capacity to degrade kerosene, such as some genera and species of yeast, to synthesize protein of unicellular origin or to bioaugment the negative environmental impact of kerosene. for the above. The objective of this work was to analyze the ability of Saccharomyces exiguus to use kerosene as a carbon and energy source. For this, S. exiguus was isolated from oil wells, it was grown in 5% kerosene with 1.2% NH4Cl and 50 ppm yeast extract. The growth of S. exiguus in kerosene was analyzed using the response variables: dry weight, protein quantification, and gas chromatography showed the use of kerosene components as a carbon and energy source. The results showed that S. exiguus can use the aliphatic and aromatic hydrocarbons of kerosene as the only source of carbon and energy, this potential is applicable to synthesize unicellular protein or in the recovery of environments impacted by kerosene.
  • Comparison of in vitro anti-Staphylococcus aureus activity of eight
           antibiotics and four dilutions of propolis

    • Abstract: Resumen El objetivo de esta investigación se basó en la comparación de halos de inhibición de una muestra de Staphylococcus aureus, enfrentada in vitro a ocho antibióticos de uso en medicina, así como, al enfrentamiento a cuatro concentraciones alcohólicas de propóleos. Los resultados señalan que la acción antimicrobiana de propóleos, depende de los compuestos bioactivos como: flavonoides, polifenoles, ácidos aromáticos, etc., contenidos en este producto. La actividad antibacteriana de las cuatro diluciones de propóleos, produjeron halos de inhibición entre 10 y 20 mm. De los ocho antibióticos con los que se realizó la comparación, solo la eritromicina resultó ser resistente a S. aureus. La penicilina produjo los halos con menores dimensiones. Aunque los halos derivados por el resto de los antimicrobianos fueron mayores a los del propóleos, muchos de ellos se encontraron dentro del rango originado por dicho producto de la colmena, no se ha demostrado resistencia antibacteriana comparada, con la que a lo largo de los años los antibióticos presentan, al ser usados indiscriminadamente para eliminar o controlar agentes antimicrobianos.Abstract The objective of this research was based on the comparison of inhibition halos of a sample of Staphylococcus aureus, faced in vitro with eight antibiotics used in medicine, as well as four alcoholic concentrations of propolis. The results indicate that the antimicrobial action of propolis, depending on the bioactive compounds such as: flavonoids, polyphenols, aromatic acids, etc., contained in this product. The antibacterial activity of the four propolis dilutions produced inhibition halos between 10 and 20 mm. Of the eight antibiotics with which the comparison was made, only erythromycin was found to be resistant to S. aureus. Penicillin produced halos with smaller dimensions. Although the halos derived from the rest of the antimicrobials were greater than those from propolis, many of them were found within the range originated by said product from the hive. Compared antibacterial resistance has not been demonstrated, with which over the year’s antibiotics present, when used indiscriminately to eliminate or control antimicrobial agents
