Pawar Pages: 1 - 20 Abstract: The main aim of present investigation was to develop and study mucoadhesive buccal tablets of sumatriptan succinate using different bioadhesive polymer. Nine formulations were prepared by using direct compression method with varying concentration of polymer as HPMC (Metolose SR 90SH-15000), xanthan gum (XTGM) and sodium carboxy methyl cellulose (sodium CMC). Fourier transform infrared spectroscopy was used to identify drug excipient interaction. Micromeritic properties of lubricated blend, physical properties and chemical evaluation of compressed tablet was performed and found satisfactory. The optimized formulation F8 containing HPMC (Metolose SR 90SH-15000) and xanthan gum (1:2) and in vitro drug release study showed sustained drug released for 9 Hours. Optimization study was performed by using design of experiment (DoE). In vitro drug release study data revealed that combination of Metolose SR 90SH-15000 (30 mg) and Xanthan Gum (60 mg) along with diluent dibasic calcium phosphate (41 mg) shown desired drug release profile with sustained manner. The approach describes the efficiency of bioadhesive polymer to adhere the mucous lining and prolong the drug release. Backing layer of ethyl cellulose (EC) was used to provide unidirectional drug release. The in vitro release kinetics reveals that formulation follows Korsmeyer–Peppas model. This study has taken with the antiulcer activity and preliminary phytochemical screening on the Hemigraphis colorata. In the present study, qualitative study of the phytochemicals present in the sample was determined, antiulcer were carried out. On the premise of the prevailing study, the outcomes eventually concluded that the ethanolic extract of Hemigraphis colorata own antiulcer pastime. Methods: Chromatographic separations were performed on an Inspire (4.6 x 150mm, 5m) column with a mobile phase composition of 0.1 percent OPA buffer and Acetonitrile (30:70) delivered at a flow rate of 1 ml/min, and detection was performed on a waters HPLC auto sampler, separation module 2695 with PDA detector 2996 at wavelength 260 nm. Results: The retention time for Gallic acid and curcumin were 2.119 and 2.730 minute respectively. The linearity correlation coefficients for Gallic acid and curcumin were 0.999 and the concentration ranges were 500-2500 g/ml and 5-25 g/ml, respectively. For accuracy the total recovery was found to be 100.58% and 100.54% for Gallic acid and curcumin. LOD and LOQ for gallic acid 3.05 and 10.07. LOD and LOQ for Curcumin 2.28 and 9.98. Conclusion: The study's findings revealed that the suggested RPHPLC method is a simple, accurate, precise, rugged, robust, rapid, and repeatable method for routinely estimating Gallic acid and curcumin in pharmaceutical dose form.
