  • Synthesis and Biological Activity of
           2-(3,3-Dimethyl-6,7-Diethoxy-3,4-Dihydroisoquinolin-1-yl)Carboxylic Acid
           Hydrochlorides and Their Benzo[f]-Analogs

      Abstract: Ritter reactions of 1-(3,4-diethoxyphenyl)-2-methylpropanol-2 with 2-alkylcyanoacetamides (alkyl = Et, n-Pr, n-Bu, n-Am, i-Am) were used to synthesize 2-(3,3-dimethyl-6,7-diethoxy-3,4-dihydroisoquinolin-1-yl)carboxylic acid amides. Similarly, reactions of 1-(2-naphthyl)-2-methylpropanol-2 with these same nitriles were used to obtain 2-(2,2-dimethyl-1,2-dihydrobenzo[f]isoquinolin-1-yl)carboxylic acid amides. Hydrochlorides of the synthesized compounds were tested for antiarrhythmic activity, which was not observed. With respect to the effect on blood clotting, nine compounds proved to be hemostatics; one compound, an anticoagulant. Three compounds of the benzo[f]isoquinoline series were the most active and increased blood coagulation by 31.7, 29.1, and 28.9%. All 10 compounds showed anthelmintic activity exceeding that of pyrantel. Two amides had anthelmintic activity exceeding that of levamisole and reduced the lifespan of worms by 11.4 and 11.6 min (vs. 20.2 min for levamisole). Nine compounds had larvicidal action exceeding that of imidacloprid. Two of them reduced the larval lifespan to 10.2 and 13.8 min, exceeding the effect of diazinon (17.0 min).

  • Effect of Euphorbia hirta on Urinary Flow in Albino Male Rats

      Abstract: The aim of this work was to evaluate diuretic activity of the ethanol extract of Euphorbia hirta leaves and elucidate the possible mechanism of its action. The diuretic activity was studied on male albino rats in comparison to furosemide as a standard drug. Two doses of E. hirta extract (300 and 600 mg/kg) were administered and the urinary volume and electrolyte (Na+, K+) concentrations were measured. Results showed that administration of the ethanol extract of E. hirta led to significantly increased urinary volume and excretion of urinary electrolytes such as sodium, potassium and chloride in 24 h urine compared to that for normal animals. Among the two selected doses, 600 mg/kg body weight exhibited higher diuretic activity level than 300 mg/kg dose. Although both these levels were statistically significant when compared to control in respect of all parameters, these levels were lower compared to the standard drug. Hence, the ethanol extract of E. hirta exhibited a dose dependent diuretic activity. Upon the isolation and identification of active compounds from E. hirta ethanol extract, it was found that lupeol and quercetin were the major constituents responsible for the diuretic activity in rats. The present study confirmed validity of the ethnopharmacological use of E. hirta as a diuretic agent under experimental conditions studied.

  • Protective Effect of Zingiber officinale Rhizomes Against Experimental
           Induced Ulcers in Diabetic Rats

      Abstract: This study was commenced to assess the antiulcer effect of the ethanol extract of Zingiber officinale rhizomes (EEZOR) against pylorus ligation-, aspirin-, and ethanol-induced ulcer models in diabetic rats. Diabetes and ulcers were induced in rats as per reported methods. The ulcer index, free acidity, pH and total acidity in pylorus ligated induced ulcer model in diabetic rats was examined using variable dosage of EEZOR and the ulcer index, % protection, and antioxidant activity parameters were studied. It was found that EEZOR inhibited the acid secretion in diabetic rats (p < 0.001) as compared to control rats with diabetes and ulcers. EEZOR also showed significant mucosal protection and antioxidant properties. It has been concluded that the EEZOR protective effect against different model ulcers in diabetic rats may be due to the presence of flavonoids and polyphenolic compound in ginger.

  • Effect of Tin(II) Chloride Amount on 99mTc-Labeling of One
           6-Hydrazinonicotinamide-Conjugated Peptide in the Presence of
           Tricine/Nicotinic Acid

      Abstract: The use of tin(II) chloride in amount above its required mole ratio as reducing agent can lead to instability of peptide-based radiopharmaceuticals. In this work, we optimize the radiolabeling conditions, especially tin(II) chloride amount for 99mTc-labeling of one 6-hydrazinonicotinamide (HYNIC) conjugated peptide in the presence of tricine and nicotinic acid (NA). Implementation of this procedure leads to 99mTc-labeling of this HYNIC-conjugated peptide in high radiochemical yield (>95%). By comparison, our experimental results can provide an efficacious strategy for 99mTc-labeling of HYNIC-conjugated biomolecules in the presence of tricine/NA. The resulting 99mTc-labeled peptide, without the need for any chemical modification and pretreatment, shows good stability in saline and human serum. This work can provide a new perspective for 99mTc-labeling of HYNIC-conjugated biomolecules.

  • Simultaneous Analysis of Atorvastatin and Rosuvastatin by LC-MS:
           Development, Validation, and Application of the Proposed Method to
           Analysis of Atorvastatin Pharmacokinetics in Pakistani Population

      Abstract: The present work has been done as part of optimization procedures aimed at developing a sensitive HPLC-MS method with good precision and accuracy for the quantification of atorvastatin and rosuvastatin in pharmaceutical and clinical samples. Amixture of methanol and water in a 70:30 ratio by volume was selected as the mobile phase and its pH was set at 3. The oven temperature was 25°C and the sample injection volume was set at 1 μL. ESI-MS interface was operated in the positive ionization mode at block temp 400°C, interface temp 350°C, DL temp 250, CID gas Ar 270 kpa, Neb gas N2 flow at 2.0 L/min. drying gas N2 flow at 10 L/min, heating gas Zero Air flow at 10.0 L/min. The analytical method validation was carried out as prescribed by the USP and ICH guidelines to provide reproducible results. Pharmacokinetics of atorvastatin from conventional (40 mg) tablets was evaluated in patients aged between 24 and 32 years. Results showed that the proposed method was rapid, both drugs were separated within 10 min. Linearity was evaluated at 0 – 100 ng/mL and showed the response to be linear for both drugs. Thus, the method proved to be rapid, specific, precise, and sensitive for simultaneous analysis of atorvastatin and rosuvastatin in biological and pharmaceutical samples.

  • Comparison of the Independent and Combined Effects of Aerobic Training and
           Ozone Therapy on Selected Heart Variables in Rats with Osteoarthritis

      Abstract: There are shreds of evidence that osteoarthritis is a risk factor for cardiovascular disease. Regular exercise and ozone therapy independently can prevent such diseases; however, their combined effect was not investigated until now. The current study was aimed to compare the independent and combined effects of aerobic training and ozone therapy on mitochondrial fission 1 protein (FIS1) and mitofusin 2 (MFN2) gene expressions, KI67 protein expression, and percentage collagen deposition in the heart tissue of rats with osteoarthritis. Thirty rats were randomly divided into five groups (six per group), named healthy control, osteoarthritis control, osteoarthritis training, osteoarthritis ozone, and osteoarthritis training + ozone. The interventions were performed according to the group name (aerobic training: run on the treadmill five sessions/week, 30 – 50 min/session for 8 weeks; ozone therapy: 20 μg/mL, once/week, for three weeks). Rat heart tissue was collected 48 h after the last intervention. Data were analyzed using two-way ANOVA and P < 0.05 was considered as significant effect. The results showed that osteoarthritis intervention significantly increased the expression of FIS-1 and MFN-2 genes as well as the percentage of collagen degradation in heart tissue. Osteoarthritis also decreased expression of the heart KI67 protein. Aerobic training and ozone therapy independently positively affect mentioned variables; however, the synergic effect was seen just for the percentage of collagen degradation. In conclusion, the present study confirms effectiveness of the aerobic exercise and ozone therapy concerning research variables and shows that combination of the exercise and ozone therapy had a greater effect than using either alone in improving collagen degradation.

      Abstract: Urinary excretion of 9-(2-diethylaminoethyl)-2-phenylimidazo[1,2-a]benzimidazole dinitrate after intragastric administration at a dose of 10 mg/kg has been studied in laboratory rats. The main metabolite in blood serum of rats was determined after a single intravenous injection at a dose of 10 mg/kg. The metabolite was determined in urine samples using HPLC. Its structure was established using HPLC-MS. According to the results, the drug substance was excreted unchanged in small amounts (~0.01% of the administered dose) because it was metabolized. The main metabolite was found in rat plasma as early as 3 min after intravenous injection at a dose of 10 mg/kg.

  • In-Vitro Antimicrobial, Antioxidant and Enzyme Inhibitory Activities of
           Fixed Oil Extracted from Stem Bark of Tamarix aphylla

      Abstract: This research work was aimed to assess the antioxidant, antimicrobial and enzyme inhibitory properties of fixed oil extracted from Tamarix aphylla stem bark. Fixed oil composition was assessed using gas chromatography–mass spectrometry (GC-MS) technique. The plant extract activity was assessed against various bacterial and fungal strains using hole diffusion and macro dilution techniques. Spectrophotometric method was used to measure the lipoxygenase and acetylcholinesterase inhibitory activities while antioxidant activity was assessed using ABTS radical scavenging and FRAP assay. Fixed oils were analyzed with the help of GC-MS, resulting in the identification of 03 compounds. Antibacterial assay showed that Staphylococcus aureus was the most affected bacterial strain with 7 mm zone of inhibition while among fungal strains, Candida glabrata was most significantly inhibited with 11 mm zone of inhibition. The tested samples were very efficient in its inhibitory activities against bacteria when 0.125, 0.5 and 1.0 mg/ml concentration was used. Similarly in case of fungi, 0.5, 1.0, 2.0 and 4.0 mg/ml of tested samples were showing significant results. Lipoxygenase and acetylcholinesterase inhibitory activities showed IC50 values of 90.1 μg/mL and 128.4 μg/mL, respectively. The antioxidant activity was assessed using ABTS radical scavenging and FRAP assays, and maximum inhibition of 87.01% was achieved at 800 μg/mL concentration. These results validate and endorse the centuries old folklore usage of T. aphylla as medicinal plant against various diseases.

  • Synthesis and Anti-Inflammatory Activity of N-aroyl-substituted
           mono(di)haloanthranilic Acid Amides and Hydrazides

      Abstract: A series of new N-aroyl-substituted mono(di)halo-anthranilic acid amides and hydrazides (I-XII) were synthesized via amidation of 6-bromo-, 6-iodo-, 6,8-dichloro-, and 6,8-dibromo-2-substituted-3,1-benzoxazin-4(3H)-ones with the corresponding 4-methylphenyl-, benzyl-, and butylamines and hydrazine. The anti-inflammatory activity of some synthesized compounds was evaluated. Compounds containing 4-methylphenyl, butyl, and benzyl substituents in the amide were active with the percentage inhibition of the carrageenan edema inflammatory reaction ranging from 45.8 to 76.9%. This class of compounds was found to be promising in the search for biologically active substances with anti-inflammatory activity.

      Abstract: Polymer complexes of the homopolymer of 2-acrylamido-2-methylpropanesulfonic acid containing 18.9 – 35.3 wt% of the antituberculosis antibiotic rifampicin were synthesized. The obtained complexes were shown to be stable in EtOH using liquid chromatography. The cytotoxicity of the polymer complexes was found to be 2.4 – 2.8 times less than that of the starting antibiotic while the high antituberculosis activity of rifampicin was retained.

  • Cytotoxic Effects and Antitumor Activity of Polysaccharides Isolated from
           the Fruiting Body of Ganoderma lucidum Basidial Mushroom

      Abstract: The work was devoted to the isolation of polysaccharides from Ganoderma lucidum basidiomycete raw material collected in Uzbekistan and the study of their cytotoxic effect and antitumor activity. Water-soluble polysaccharides were isolated from the basidiomycete raw material in a yield of 15.16% by successive water extraction. The isolated polysaccharides were purified from proteins by the Savage method. The amount of carbohydrates (52.74%) was determined using the phenol-sulfuric acid method. An analysis of the elemental composition showed that the isolated polysaccharide contained 5.27% N, 54.30% C, and 7.79% H. It was established that the isolated polysaccharides did not produce direct cytotoxic effects on CML and HeLa cells in vitro. The antitumor activity of the polysaccharides was studied in vivo. It was found that the isolated polysaccharides at 25 mg/kg/day for 10 d inhibited Ehrlich adenocarcinoma solid tumor growth in mice by 59.34 vol% and 60.21 wt%.

  • From Assessment of Radionuclide Contamination in Herbal Medicine Raw
           Material of Voronezh Region to Issues Concerning Standardization of
           Natural Radionuclides

      Abstract: The research was aimed at studying the accumulation of natural and artificial radionuclides in medicinal plant raw materials (MPMs) and determining approaches to standardization of natural radionuclides in them using plants growing in Voronezh Region as examples. A total of 36 collection sites of soil and MPM located throughout Voronezh Region and diverse in terms of anthropogenic impact were selected. Pharmacopoeial species of MPM were used as research subjects. All MPMs met the requirements of GPM.1.5.3.0001.15 for specific activity of cesium-137 and strontium-90 and for the total indicator of compliance with radiation safety requirements and its determination accuracy. The obtained specific activities of natural radionuclides in most cases exceeded the numerical values for radionuclides of anthropogenic origin. Asupplement to general pharmacopoeia monograph GPM.1.5.3.0001.15 establishing the additional numerical indicator “Effective specific activity of natural radionuclides not greater than 165 Bq/kg” is proposed to monitor contamination of MPM with natural radionuclides. These recommendations are based on an analysis of greater than 360 MPM samples from territories subject to various anthropogenic impacts. However, they are preliminary in nature and require additional research on MPMs from other regions of the Russian Federation.

  • Spectrophotometric Methods for Simultaneous Determination of Sulfadoxine
           and Pyrimethamine in Bulk and Tablet Forms

      Abstract: Three sensitive and accurate spectrophotometric methods were developed for the quantitative determination of sulfadoxine (SUL) and pyrimethamine (PYR) in bulk and tablet forms. Method I is based on the solvent extraction using separatory funnel, which depends on the partition of SUL and PYR in different solvents and spectrophotometric analysis of the extracts. SUL was extracted using 1M NaOH (upper layer) and PYR was extracted by CHCl3 (lower layer). The swamping method (II) is based on the presence of SUL in high amount compared to PYR (20:1). Method III is based on the simultaneous spectrophotometric determination of both drugs at 274 and 286 nm. The developed methods were validated according to ICH guidelines in terms of linearity, accuracy and precision.

      Abstract: A comparative study of methods for the determination of bacterial endotoxins using recombinant reagents confirmed that the analytical results did not differ from the values obtained using a chromogenic kinetic test with LAL reagent of natural origin. It was experimentally confirmed that recombinant reagents did not respond to the presence of β-glucan impurities, thus excluding the possibility of false-positive reactions.

      Abstract: Organic salts of N1-(2,3,4-trimethoxybenzyl)-N2-{2-[(2,3,4-trimethoxybenzyl)amino]ethyl}-1,2-ethanediamine (I) (succinate ALM-802S and citrate ALM-802C) were obtained. These organic salts were found to retain completely the anti-arrhythmic and anti-ischemic properties of I trihydrochloride (ALM-802) in equimolar doses (2.15 – 2.75 mg/kg, i.v. administration). The new compounds belonged to toxicity class VI (low-toxicity substances). The LD50 values of the organic salts of I recalculated as the base corresponded to that of I trihydrochloride. The pH values of the organic salts of I were significantly higher than that of I trihydrochloride (5.5 – 5.8 vs. 3.5), which eliminated the local irritating action and undesirable organoleptic properties.

      Abstract: A synthetic method for 2-amino-1-(1-indol-3-yl)ethylphosphonic acid and 2-amino-1-(1-methylindol-3-yl)ethylphosphonic acid hydrochlorides, i.e., new indole-containing functionalized aminophosphonic acids, is proposed. The reactions are carried out at room temperature in LiOH solution followed by precipitation of the salts using HCl. The synthesized indolylaminoethylphosphonic acids, the molecules of which contain an indole ring and amine and phosphonic groups, could be considered synthetic precursors of functionalized tryptamine analogs.

      Abstract: The bioavailability of the tablet dosage form of pharmaceutical substance RU-891, i.e., 9-(3,4-dihydroxyphenacyl)-2,3-dihydroimidazo[1,2-a]benzimidazole hydrobromide, was investigated. The main pharmacokinetic parameters were determined. The relative bioavailability index was estimated at 93.98%. No evidence of drug accumulation from an RU-891 270-mg tablet dosage form was detected in rabbits upon threefold administration at a dose of 23 mg/kg.

  • Synthesis and Anti-Inflammatory Activity Evaluation of Some Benzimidazole
           Derivatives

      Abstract: Hyper-inflammation aggravates the symptoms of both communicable and non-communicable diseases. Therefore, anti-inflammatory compounds may have wide therapeutic application. Benzimidazole is a privileged scaffold and its success in drug development is evident from the long list of benzimidazole-based drugs with wide range of applications. This study was undertaken to develop new small molecules with anti-inflammatory properties. Compounds MBPHYD, MBNHYD and MBHYDX were synthesised, purified, characterised and found to be non-toxic both in vitro (in 100 μMconcentration for 24 h vs. 3000 Vero cells/well) and in vivo (at a dose of 100 mg/kg in female Wistar rats with animals observed for 48 h for any mortality). Compounds MBPHYD and MBNHYD were found to possess significant anti-inflammatory properties. Further, in silico analysis suggested their compliance with drug-likeness. While no toxicity was predicted, both compounds were suggested to have good oral bioavailability. Thus, results of this study may encourage further investigation to establish new anti-inflammatory benzimidazoles for application against various disease conditions.

  • New Potentiometric Sensor for Determination of Metformin

      Abstract: A potentiometric metformin-sensitive detector with a plasticized polyvinyl chloride membrane has been developed. The sensor contains an ion associate of metformin with bromophenol blue dye. The response is linear within the range of metformin ion concentrations 1 × 10–-4–1 × 10–-1 M. The slope of the electrode function is characteristic of a singly charged cation. The limit of detection is 8 × 10–-5 M. The sensor can be used for detecting and determining metformin in medicines (RSD = 0.59 – 2.52%).
