  • OPTIMASI SUHU, pH DAN AMOBILISASI SELULASE DARI KONSORSIUM MIKROBA
           SELULOLITIK (KMS.UU1a) PADA KALSIUM ALGINAT

      Abstract: ABSTRAK: Tujuan penelitian ini adalah untuk mengetahui suhu dan pH optimum selulase dari konsorsium mikroba selulolitik KMS.UU1a dalam kondisi bebas dan terambobilisasi, konsentrasi kalsium alginat optimum untuk amobilisasi selulase, serta keberulangan (reusability) dan efisiensi selulase terambilisasi. Pengukuran aktivitas selulase dilakukan berdasarkan kandungan gula pereduksi sebelum dan sesudah reaksi enzimatis dengan metode Dinitrosalycilic acid (DNS). Aktivitas selulase KMS.UU1a baik dalam kondisi bebas dan teramobilisasi diuji pada suhu 35, 40, 45, 50, 55 dan 60 oC; dan pH 4, 5, 6, 7, 8, dan 9. Konsentrasi sodium alginat yang digunakan dalam proses amobilisasi selulase KMS.UU1a adalah 0,5, 1,0, 1,5, 2,0, 2,5 dan 3,0 % (b/v) dalam CaCl2 0,15 M. Uji keberulangan dilakukan dengan mengukur aktivitas selulase KMS.UU1a amobil pada 4 (empat) kali pemakaian enzim, sedangkan efisiensi diketahui dari uji aktivitas selulase bebas sebelum dan setelah amobilisasi. Konsentrasi kalsium alginat optimum untuk amobilisasi selulase KMS.UU1a adalah 2,0 % (b/v). Suhu optimum selulase KMS.UU1a bebas lebih rendah (40 oC) dibandingkan dengan selulase amobil (45 oC), dengan aktivitas selulase bebas 0,005636 U/mL, lebih tinggi dibandingkan dengan aktivitas selulase amobil 0,003617 U/mL. Selulase KMS.UU1a bebas dan amobil sama-sama memiliki pH optimum 6,0 dengan aktivitas selulase berturut-turut sebesar 0,006444 U/mL dan 0,003771 U/mL. Keberulangan dilihat dari besarnya aktivitas selulase KMS.UU1a amobil pada pemakaian kedua, ketiga, dan keempat terhadap pemakaian pertama berturut-turut sebesar 70,53; 61,66; dan 23,92 %. Efisiensi amobilisasi selulase KMS.UU1a pada kalsium alginate diperoleh sebesar   63,8711 %. ABSTRACT: The purpose of this study was to determine the optimum temperature and pH of cellulase from the KMS.UU1a cellulolytic microbial consortium in free and immobilized conditions, the optimum percentage of calcium alginate used for cellulase immobilization, reusability and efficiency of immobilized cellulase. The enzyme activity was determined by the Dinitrosalycilic acid (DNS) method KMS.UU1a cellulase activity in both free and immobilized conditions was tested at temperatures of 35, 40, 45, 50, 55 and 60 oC; and pH 4, 5, 6, 7, 8, and 9. The concentration of alginate used in the immobilization process of KMS.UU1a cellulase was 0.5, 1.0, 1.5, 2.0, 2.5 and 3.0% (w/v) in 0.15 M CaCl2, was done by measuring the immobilized KMS.UU1a cellulase activity in 4 (four) enzyme usage cycles, while the efficiency was known from the cellulase free activity test before and after immobilization. The optimum concentration of calcium alginate for immobilization of KMS.UU1a cellulase was 2.0% (w/v). The optimum temperature of free KMS.UU1a cellulases was lower (40 oC) compared to immobilized cellulases (45 oC), with free cellulase activity of 0.005636 U / mL, higher than that of immobilized cellulases of 0.003617 U / mL. Both free and immobilized KMS.UU1a cellulases had an optimum pH of 6.0, with cellulase activity of 0.006444 U/mL and 0.003771 U/ mL, respectively. Repetition can be seen from the immobilized KMS.UU1a cellulase activity in the second, third, and fourth cycles of the first cycle respectively of 70.53; 61.66; and 23.92%. The immobilization efficiency of KMS.UU1a cellulase on calcium alginate was 63.8711%.

  • SINTESIS DAN KARAKTERISASI HIDROKSIAPATIT (HAp) DARI CANGKANG KERANG
           ALE-ALE MENGGUNAKAN METODE PRESIPITASI DOUBLE STIRRING

      Abstract: ABSTRAK: Kerang ale-ale (Meretrix meretrix) merupakan sumber daya alam potensial di Kabupaten Ketapang Provinsi Kalimantan Barat. Penangkapan kerang ale-ale yang berlangsung sepanjang tahun menghasilkan dampak negatif berupa limbah cangkang yang belum dikelola secara maksimal. Cangkang kerang ale-ale memiliki kandungan kalsium tinggi yang berpotensi untuk dimanfaatkan sebagai prekursor kalsium dalam sintesis hidroksiapatit (HAp). Penelitian ini bertujuan memanfaatkan cangkang kerang ale-ale sebagai prekursor Ca untuk menyintesis HAp, melakukan sintesis HAp dari cangkang kerang ale-ale, dan melakukan karakterisasinya. Analisis serbuk cangkang kerang ale-ale menggunakan Spektrofotometer Serapan Atom (AAS) menunjukkan kandungan kalsium dalam cangkang sebesar 68.04%. Hasil sintesis dengan metode presipitasi double stirring menunjukkan HAp yang diperoleh merupakan fasa tunggal Hap dengan derajat kristalinitas sebesar 90.12% dengan ukuran kristal rata-rata sebesar 41,08 nm. Hasil analisa SEM pada hidroksiapatit menunjukkan partikel berbentuk bola dengan ukuran sekitar 65 nm dan ukuran rata-rata pori yang homogen.   ABSTRACT: Ale-ale clamshell (Meretrix meretrix) is a potential natural resource of Ketapang Regency of West Kalimantan. The ale-ale fishing activities in this region take place throughout the year resulting the negative impact in the form of clamshell waste that has not yet been optimally managed. The ale-ale clamshell contains high calcium content which has the potency to be used as a precursor of calcium in synthesis of hydroxyapatite (HAp). This study aimed to utilize the waste of the ale-ale clamshells as a precursor of calcium in synthesizing of HAp, to synthesize of HAp from ale-ale clamshell, and to carry out the characterizations. Analysis of calcium content by Atomic Absorption Spectroscopy (AAS) showed that ale-ale clamshell was 68.04% (by weight). The double-stirring precipitation method synthesized HAp confirmed the formation of single phase of Hap with crystalinity degree and average crystal zise of 90.12% and 41.08 nm, respectively. The SEM data showed that the HAp has spherical in morphology with around 65 nm in size and homogen average porous size.

           paradisiaca L.) TERHADAP BAKTERI Staphylococcus aureus dan Escherichia
           coli

      Abstract: ABSTRAK: Musa x paradisiaca L. merupakan salah satu jenis pisang yang berpotensi sebagai antibakteri. Tujuan penelitian ini adalah untuk mengetahui aktivitas antibakteri ekstrak metanol kulit buah pisang pecah seribu (Musa x paradisiaca L.) terhadap bakteri Staphylococcus aureus dan Escherichia coli serta menentukan kandungan metabolit sekundernya. Ekstraksi dilakukan dengan cara maserasi menggunakan pelarut metanol 95% pengujian aktivitas antibakteri dilakukan dengan metode sumur difusi, dan kandungan metabolit sekuder dalam ekstrak metanol dilakukan dengan uji fitokimia. Hasil uji fitokimia menunjukkan bahwa ekstrak mengandung metabolit sekunder flavonoid, fenol, alkaloid, dan triterpenoid. Hasil uji antibakteri menunjukkan bahwa Konsentrasi Hambat Minimum (KHM) dan Konsentrasi Bunuh Minimum (KBM) ekstrak metanol terhadap Staphylococcus aureus sebesar 4% dan Escherichia coli sebesar 6% dengan daya hambat masing-masing sebesar 6,83 dan 5,25 mm. ABSTRACT: Musa x paradisiaca L. is one type of banana that has potential as an antibacterial. The purpose of this study was to determine the antibacterial activity of the methanol extract of pecah seribu banana peels (Musa x paradisiaca L.) against Staphylococcus aureus and Escherichia coli and determine the content of secondary metabolites. Extraction was carried out by maceration using 95% methanol. Antibacterial activity assay was perfomed by the diffusion well method and the content of secondary metabolites in the methanol extract was carried out by phytochemical tests. The phytochemical test results show that the extract contains secondary metabolites of flavonoid, phenol, alkaloid, and triterpenoid. The antibacterial assay results revealed that the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of methanol extract against Staphylococcus aureus of 4% and Escherichia coli of 6% with inhibition of 6.83 and 5.25 mm, respectively.

      Abstract: ABSTRAK: Teknik ekstraksi pelarut transport aktif (EPTA) merupakan ekstraksi pelarut dengan menambahkan zat lain yang berfungsi ganda sebagai ekstraktan dan zat aktif permukaan. Tujuan penelitian ini adalah menentukan persentase recovery logam paladium (II) yang terekstraksi dari sampel limbah industri pertambangan dengan teknik EPTA pada kondisi optimum dari parameter ekstraksi. Metode penelitian yang digunakan adalah teknik EPTA. Hasil penelitian menunjukkan bahwa kondisi terbaik ekstraksi untuk teknik EPTA menggunakan larutan standar Pd (II) 100 ppm adalah waktu ekstraksi selama 20 menit, laju putaran pengadukan sebesar 1000 rpm, konsentrasi HCl pada fasa air adalah 0,25 M, dan konsentrasi trioktilmetilamonium klorida (TOMAC) pada fasa organik adalah 0,033 M sedangkan pada sampel limbah industri pertambangan dapat mentranspor rata-rata sekitar 61,04 ± 1,88%  yang mengandung Pd (II) sebesar 320 ppm. ABSTRACT: Active transport solvent extraction technique (EPTA) is a solvent extraction by adding other substances that act as extractants and surface active agent. The purpose of this study was to determine the percentage of recovery of palladium (II) extracted from mining industry waste samples with the EPTA technique at the optimum conditions of the extraction parameters. The research method used is the EPTA technique. The results showed that the best extraction conditions for the EPTA technique using Pd (II) standard solution of 100 ppm were extraction time of 20 minutes, stirring rate of 1000 rpm, HCl concentration of 0.25 M in the aqueous phase, and methyltrioctylammonium chloride (TOMAC) concentration of 0.033 M in the organic phase. The extraction process using the optimum conditions can only transport about 61,04 ± 1,88% of Pd (II) content out of 320 ppm from the mining industrial waste sample.

  • PENETAPAN KADAR PARASETAMOL DAN KAFEIN DENGAN METODE HIGH PERFORMANCE

      Abstract: ABSTRAK: Parasetamol dan kafein menjadi kombinasi dalam persediaan obat untuk dapat memberikan efek analgetik. Penggunaan ini menjadi semakin meningkat untuk dapat mencapai efek yang baik dalam pengawasan zat aktif dalam formulasi farmasetik. Penelitian ini bertujuan untuk dapat melakukan preparasi untuk sediaan tablet dengan kandungan parasetamol dan kafein. Desain penelitian adalah eksperimental menggunakan metode HPLC dengan detektor dan direkam dalam bentuk kromatogram dengan kolom reversed phase C18 dengan ukuran 4.6 mm x 10 cm. Temperatur kolom 45 ± 1 oC dan fase gerak melalui membran filter dengan kecepatan 2 mL/menit dengan detector UV 275 nm. Pada penelitian ini dilakukan uji kesesuaian sistem serta penentuan presisi dan akurasi metode analisis. Hasil penelitian ini menunjukkan bahwa uji kesesuaian sistem memenuhi syarat dengan deviasi relatif (RSD). Berdasarkan metode yang dilakukan terdapat validasi metode presisi, akurasi, linieritas, LOD dan LOQ diterima. Nilai RSD larutan uji parasetamol dan kafein berturut-turut adalah 0,4437% dan   2,8959%. Untuk validasi presisi diperoleh rata-rata % recovery larutan uji parasetamol dan kafein berturut-turut adalah 100,01758% dan 97,42951907%. Nilai LOD dan LOQ diperoleh berturut-turut adalah sebesar 26.74885159 ng/10µL dan 89,162838 ng/10µL. Kadar larutan sampel parasetamol 998,226 ng/10µL. Dengan demikian, karena kadar sampel parasetamol melebihi nilai batas deteksi sehingga sampel parasetamol dapat dideteksi namun tidak dapat dikuantifikasi. ABSTRACT: Paracetamol and caffeine become a combination of drug supplies to provide analgesic effects. This use is increasingly being used to achieve a good effect on the control of active substances in pharmaceutical formulations. This study aims to be able to acurately prepare tablets containing paracetamol and caffeine. The research design was experimental using the HPLC method with a detector with a 4.6 mm x 10 cm long packing column C18. The temperature of the reversed-phase column was 45 ± 1 oC and the moving phase through the filter membrane at a rate of 2 mL/minute with a 275 nm UV detector. In this study, the system suitability test was carried out by determining  the precision and accuracy of the analytical method. The results of this study indicate that the system suitability test meets the requirements with relative deviation (RSD). Based on the method used, there are validation methods for precision, accuracy, linearity, LOD, and LOQ are accepted. The RSD value of the paracetamol and caffein test solutions were 0.4437% and 2.8959%, respectively. For precision validation, the average % recovery for the paracetamol test solution was 100.01758% and the average% recovery for the caffeine test solution was 97.43%. Then, for the validation of the method, the LOD and LOQ  were 26.7489 ng / 10µL and 89.1628 ng / 10µL respectively.The paracetamol concentration of sample solution was 998.226 ng / 10µL. Since  the paracetamol sample level exceeds the detection limit value so that the paracetamol sample can be detected but can not be quantified.

  • POTENSI ZEOLIT ALAM ENDE SEBAGAI MEDIA ADSORBEN KOMPETITIF PEWARNA BIRU
           METILENA DAN METIL ORANYE

      Abstract: ABSTRAK: Limbah yang berasal dari industri tekstil dianggap sebagai sumber potensial pencemaran lingkungan khususnya air  karena mengandung pewarna yang berbahaya. Dalam penelitian ini zeolit alam digunakan sebagai alternatif adsorben yang efektif dan efisien untuk mengatasi pencemaran akibat pewarna biru metilena dan metil oranye. Aktivasi zeolit alam dilakukan dengan larutan HCl 3 M dan dikarakterisasi menggunakan difraksi sinar-X (XRD) dan Scanning Electron Microscope (SEM). Proses adsorpsi biru metilena dan metil oranye dipelajari pada variasi bobot adsorben, waktu kontak,  dan pH. Konsentrasi zat pewarna yang tertinggal dalam larutan diamati dengan instrumen Spektrofotometer UV-Vis.  Hasil penelitian menunjukkan bahwa kapasitas adsorpsi zeolit alam dalam menjerap biru metilena sebesar 21,189 mg/g dan dan metil oranye sebesar 18,208 mg/g. Kondisi optimum adsorpsi biru metilena dan metil oranye dicapai dengan bobot adsorben berturut-turut 0,3 g dan 0,4 g, waktu kontak berturut-turut 60 menit dan 90 menit, serta pH berturut-turut 6 dan 2. Faktor bobot adsorben, waktu kontak, dan pH memberikan pengaruh terhadap penjerapan biru metilena maupun metil oranye oleh zeolit alam Ende. ABSTRACT: Waste from the textile industry is considered as a potential source of environmental pollution, especially water because it contains dangerous dyes. In this research, natural zeolite was used as an effective and efficient alternative adsorbent to overcome pollution caused by methylene blue and methyl orange dyes. Activation of natural zeolite was carried out with aqueous solution of HCl 3 M and characterized using X-Ray Diffraction (XRD) and Scanning Electron Microscope (SEM). The methylene blue and methyl orange adsorption processes were studied on various weight of the adsorbent, contact time, and pH. The concentration of dyes remaining in the solution was observed with Spectrophotometer UV-Vis. The results showed that the adsorption capacities of activated natural zeolite were 21.189 mg / g and 18.208 mg / g for methylene blue and methyl orange, respectively. The optimum conditions of methylene blue were 0.3 g of adsorbent weight, 60 minutes of contact time, and 6 of pH while for the methyl orange adsorption were achieved at  0,4 g of adsorbent weight, 90 minutes of contact time and 2 of pH.
