      Abstract: The review summarizes and systematizes the literature data on the use of 5(4)-aminopyrazoles in the synthesis of pyrazolo[3,4-b]- and pyrazolo[4,3-b]pyridines. The synthetic methods are classified on the basis of how well studied the pyrazolopyridine fragment is, i.e from more to less studied. The bibliography of the review includes 91 references published over the last two decades.

  • The effect of substituents on carbon–carbon double bond isomerization in
           heterocyclic hydrazine derivatives

      Abstract: Even though enehydrazide moiety is present in many pharmaceuticals, there is currently no straightforward method available for preparing cyclic enehydrazides, which could be valuable building blocks in anticancer research. Herein, we report how electronic effects and ring size influence the direction and yield of Ru catalytic carbon–carbon double bond isomerization in heterocyclic enehydrazines. Having the knowledge of how variation of these properties affects the equilibrium between double bond isomers enables us to control the outcome when preparing different cyclic enehydrazides. Six enehydrazide heterocycles and five enehydrazine heterocycles were synthesized and characterized with the current method. In addition, cytotoxicity evaluation of the synthesized compounds showed that several heterocycles produced in this study could be used in developing anticancer drugs.

      Abstract: Direct SNH amidation of 6- and 7-nitroquinoline N-oxides in anhydrous DMSO allowed to obtain N-oxides of 2- and 8-aroylaminonitroquinolines, respectively. 5-Nitroquinoline N-oxide was transformed into a mixture of amides derived from the N-oxides of 5-nitroand 5-nitrosoquinolines. 8-Nitroquinoline N-oxide underwent destruction under the same conditions.

  • Synthesis of 1,2,6-trisubstituted indoles from
           6-propargylcyclohex-2-enones and primary amines

      Abstract: The synthesis of 1,2,6-trisubstituted indoles from accessible 6-propargylcyclohex-2-enones and primary amines is reported. Simple acidcatalyzed amination–cyclization reaction followed by air oxidation of the corresponding intermediated 4,5-dihydroindoles was found to be suitable to obtain the desired indoles. The reaction tolerates a variety of functional groups on the starting cyclohex-2-enones and primary amines. A mechanistic pathway involving nucleophilic addition/cyclization/aromatization is proposed.

  • Synthesis of novel pyrimido[4,5-d]pyrimidine derivatives from
           5-acetyl-4-aminopyrimidines

      Abstract: Two routes were proposed for the synthesis of pyrimido[4,5-d]pyrimidine derivatives from 5-acetyl-4-aminopyrimidines. Acylation of 5-acetyl-4-aminopyrimidines followed by cyclization by the action of NH4OAc made it possible to access pyrimido[4,5-d]pyrimidines with alkyl, aryl, and hetaryl substituents in positions 2 and 7. Reductive amination of the acetyl group in 5-acetyl-4-aminopyrimidines and subsequent cyclization with DMF–DMA or HC(OEt)3 resulted in the formation of 6-alkyl-3,4-dihydropyrimido[4,5-d]pyrimidines.

      Abstract: Chiral 2-trichloromethyloxazolines 4,5-cis-annulated to a carbohydrate fragment were synthesized on the basis of levoglucosenone. The synthesized compounds were converted by acid hydrolysis into 4-substituted 3-amino-1,6-anhydro-3-deoxy-β-D-mannopyranoses. Intramolecular amination of allyl trichloroacetimidates by electrophilic reagents was used to construct the oxazoline ring. It was established that trichloroacetimidates containing an epoxide ring in α,β-positions spontaneously undergo recyclization with the formation of an oxazoline ring.

      Abstract: The computational wb97xd/6-311+G(d)(PCM) study confirms without any doubt the polar, stepwise mechanism of hetero-Diels–Alder reactions as illustrated on the example of 5,6-diphenyl-1,2,4-triazine and 2-cyclopropylidene-1,3-dimethylimidazoline. The first reaction stage is the formation of a prereaction molecular complex, which can next convert to two possible zwitterionic intermediates characterized by “cyclic” and “extended” conformation. The first one cyclizes in the second reaction step to the expected six-membered cycloadduct.

      Abstract: The review presents the general methods for the formation of the framework of 1,4-dihydro- 1,2,4-benzotriazin-4-yl radicals (Blatter radicals) and their polycyclic derivatives published over the past 7 years. The target heterocyclic structures are formed by a number of approaches such as oxidative cyclization of amidrazones, addition of organolithium reagents to 1,2,4-benzotriazines, intramolecular cyclizations of 1,2,4-benzotriazine derivatives, and other methods.

      Abstract: 3-{[(1-Methyl-1H-tetrazol-5-yl)imino]methyl}quinoline-2-thiol and 3-{[(2-methyl-2H-tetrazol-5-yl)imino]methyl}quinoline-2-thiol were synthesized. The sequence of the thiol-Michael reaction and the (aza)-Morita–Baylis–Hillman reaction yielded 4-[(1-methyl-1H-tetrazol-5-yl)amino]-2-phenyl-4H-thiopyrano[2,3-b]quinoline-3-carbaldehyde, 4-[(2-methyl-2H-tetrazol-5-yl)amino]-2-phenyl-4H-thiopyrano[2,3-b]-quinoline-3-carbaldehyde, and 4-hydroxy-2-phenyl-4H-thiopyrano[2,3-b]quinoline-3-carbaldehyde. Cytotoxicity and antiviral activity against the A/Puerto Rico/8/34 (H1N1) influenza virus strain in MDCK cell culture were determined for the obtained compounds. The study showed that the replacement of the hydroxyl group in 4-hydroxy-2-phenyl-4H-thiopyrano[2,3-b]quinoline-3-carbaldehyde with a 1-methyl- or 5-amino-2-methyltetrazolyl fragment decreased antiviral activity. At the same time, 3-{[(1-methyl-1H-tetrazol-5-yl)imino]-methyl}quinoline-2-thiol has a higher activity than 3-{[(2-methyl-2H-tetrazol-5-yl)imino]methyl}quinoline-2-thiol. This fact indicates a possible relationship between the arrangement of substituents in the tetrazole ring and the antiviral activity of the tested heterocyclic system.

      Abstract: Imidazo[1,2-a]pyridine derivatives have been shown to possess a broad range of biological activity. 2-Amino-1-propargylpyridinium and 2-amino-1-(2-bromoallyl)pyridinium bromides, the structures of which were established on the basis of X-ray structural analysis, reacted with sodium methoxide, leading to the formation of 2-methylimidazo[1,2-a]pyridine. 2-Methylimidazo[1,2-a]pyridine further reacted with bromine and iodine, providing 3-halo-2-methyl-1H-imidazo[1,2-a]pyridinium trihalides, the structure of which was confirmed by X-ray structural analysis. 3-Halo-2-methyl-1H-imidazo[1,2-a]pyridinium halides were obtained from the respective trihalides. 3-Bromo- 2-methyl-1H-imidazo[1,2-a]pyridinium bromide showed antimicrobial properties against Staphylococcus aureus at the concentrations of 2700 and 675 μg/ml.

      Abstract: Upon treatment of 3-nitrobenzofurans with dimethylphenacylsulfonium salts in the presence of a base, the opening of the furan ring to form (E)-1-aryl-2-(dimethylsulfonio)-4-nitro-1-oxobut-3-en-2-ides takes place. The mechanism of nucleophilic dearomatization of 3-nitrobenzofurans involves consecutive carbo- and retro-oxa-Michael reactions. The reaction illustrates the high propensity of 3-nitrobenzofurans for ring opening.

      Abstract: The conditions for bromination of the methyl groups of dimethylglyoxime were found, and the resulting product, a 1,4-dioxane solvate of bis(bromomethyl)glyoxime, was used as a precursor for heterocyclizations involving cyclodehydration or oxidative cyclization of the oxime groups leading to the formation of furazan and furoxan rings, respectively.

  • New 13-vinyl derivatives of berberine: synthesis and characterization

      Abstract: The possibility of obtaining electroneutral substituted 13-vinylberberines was demonstrated experimentally and via quantum-chemical DFT/B3LYP calculations in the 6-31++G(d,p) basis set. The introduction of pharmacophoric fragments conjugated through vinyl moiety opened new possibilities for structural modification of berberine, enabling pronounced changes in tropicity toward supramolecular biological structures. The newly synthesized 13-vinylberberines were stable in their reduced form due to significant intramolecular electron density transfer from berberine ring system to the vinyl moiety bearing electron-withdrawing groups. It was demonstrated that berberine derivatives may exist not only in ion pair form consisting of organic cation and inorganic anion, but also as zwitterionic structures featuring significant intramolecular charge transfer. The obtained 13-vinylberberines exhibited biological activity against the highly pathogenic Vibrio cholerae.

  • Expanding the chemical space of 3(5)-functionalized 1,2,4-triazoles

      Abstract: An efficient approach to the gram-scale synthesis of 3(5)-substituted, 1,3- and 1,5-disubstituted 1,2,4-triazole-derived building blocks is described. The key synthetic precursors – 1,2,4-triazole-3(5)-carboxylates (20 examples, 35–89% yield) were prepared from readily available acyl hydrazides and ethyl 2-ethoxy-2-iminoacetate hydrochloride. Further transformations were performed following the convergent synthetic strategy and allowed the preparation of 1,3- and 1,5-disubstituted 1,2,4-triazole-derived esters (16 examples, 25–75% yield), 3(5)-substituted, 1,3- and 1,5-disubstituted carboxylate salts (18 examples, 78–93% yield), amides (5 examples, 82–93% yield), nitriles (5 examples, 30–85% yield), hydrazides (6 examples, 84–89% yield), and hydroxamic acids (3 examples, 73–78% yield). Considering wide applications of the 1,2,4-triazole motif in medicinal chemistry, these compounds are valuable building blocks for lead-oriented synthesis; they have also great potential for coordination chemistry.

  • Synthesis of Polymethoxyflavonoids from Hesperidin and Naringin and their
           Antiproliferative Activity

      Abstract: A series of polymethoxyflavonoids were synthesized via cleavage of the glycosidic bond, dehydrogenation, selective methylation, bromination, nucleophilic aromatic substitution, O-prenylation, Claisen–Schmidt aldol condensation, cyclization, and oxidation from very abundant and inexpensive natural flavonoids hesperidin and naringin. The in vitro antiproliferative activity of the synthesized compounds was evaluated against a panel of four human cancer cell lines (Aspc-1, HCT-116, HepG-2, and SUN-5) by the CellTiter-Glo assay. The results showed that some of synthesized compounds exhibited moderate to high antiproliferative activity. Among them, (2E)-1-(2-hydroxy-3,4,5,6-tetramethoxyphenyl)-3-(7-methoxy-1,3-benzodioxol-5-yl)prop-2-en-1-one was revealed to be the most active with IC50 values ranging from 10.35 to 13.89 μM against all four cancer cell lines, the IC50 value (10.35 μM) being below positive control cisplatin (12.36 μM) against HepG-2 cells.
