In vitro Apical Shoot Culture and Callus Formation of Durian (Durio
zibethinus Murray)

  Abstract: Backgroud: Durian (Durio zibethinus Murray), A tropical fruit with high nutritional and commercial value, is widely cultivated in Southeast Asia. Unfortunately, growing durian is fraught with difficulties. Propagation is the traditional method for conserving plants. This method does have some drawbacks, one of which is that the original tree may suffer as a result. Therefore, a contemporary method for growing durian plants is plant tissue culture. Aim: The objective of this research is creating a premise for the development of durian high-value varieties by biotechnology, specifically setting up the first step closer to develop durian somatic cell formation protocol in vitro. Methodology: Effects of media MS on shoot culture and Kinetin and 2.4D on callus formation and growth. Experiments were designed by randomized complete block (RCB). Results: There are not significant difference between two treatments of 0.1 mg/L BAP (6-benzylaminopurine) added to 1/3xMS and free hormone medium, in an effort to preserve high-value durian varieties. There is research on formating callus from young leaves of durian affected by Kinetin and 2.4D. The outcomes demonstrated that MS medium supplemented with 3 mg/L 2.4D had an impact on the formation of callus on leaves. The combination of 1.0 mg/L Kinetin + 4.0 mg/L 2.4D make a significant to induce callus growth and callus area. Callus was white color, fresh and soft. Conclusion: the combination of Kinetin + 2.4D was finding the best combination to maintain the survival of callus induction and growth; coconut water treatment was highly effective in sustaining fresh callus.

Profiling Bioactive Compounds in the Bark of Cinchona officinalis L. and
Anti-arthritic Properties Thereof

  Abstract: The bark samples of C. officinalis were collected from The Nilgiris district of Tamil Nadu. The collected samples were processed and extracted using methanol solvent. The extracts showed high yield of secondary metabolites especially alkaloids, phenols, flavonoids, steroids, terpenoids, sterols, quinones and tannins. Alkaloid content was very high in C.officinalis followed by phenol, flavonoid, steroid and Tannin. The secondary metabolites present in C. officinalis (Bark) were isolated, purified, identified and characterized using differential Chromatographic techniques such as TLC, HPLC and GC-MS/ MS. The compounds identified using High Performance Liquid Chromatography (HPLC) technique in bark of C. officinalis in the retention times of 1.750, 2.250, 4.016 and 7.266 minutes were quinine, isoquinidine and cinchonidine. The compounds were then characterized using GC- MS/ MS technique with reference to NIST library. The compounds were eluted from the bark of C. officinalis in the retention time of 9.569, 10.488, 11.814, 13.593, 14.282, 16.542, 16.967, 23.494, 24.954, 25.151, 25.988, 26.752, 26.982, 27.906, 28.277, 31.454, 33.604, and 35.744 minutes. By correlating the retention times with (NIST) mass spectral library the eluted compounds were confirmed as Phthalic acid, Hexadecanoic acid,1,2,3,5-Cyclohexanetetrol, Cinchonab-9-one, 9,12-Octadecadienoic acid (Z,Z)-, Heptadecanoic acid, Quinine, Cinchonine, Cinchonidine, Phenol, Quinine, and gamma.-Sitosterol. The phytochemicals present in the C. officinalis (Bark) samples were evaluated for their biological properties viz., anti-inflammatory activity and anti-arthritic activity in in-vitro conditions. Anti- inflammatory properties of plant samples were tested by protein denaturation method using diclofenac (75 mg/ ml concentration) as positive control. The methanol extract of C.officinalis (Bark) showed anti- inflammatory properties of range between 17.0 and 57.3%. Anti- arthritic activity of bark extract of C. officinalis (75 mg/ ml concentration) was evaluated by albumin denaturation method using diclofenac (75 mg/ ml concentration) as positive control. The methanol extract of C.officinalis (Bark) showed anti- arthritic properties of range between 18.4 and 58.5%. From the study it is understood that the alkaloids and polyphenols present in the methanolic extract of bark of C. officinalis were responsible for their biological properties. Hence, C. officinalis can serve as an alternative to synthetic drugs available in the market to treat arthritis subject to safety (toxicity) studies in humans. Based on the In vitro results, it was confirmed that the bark of C.officinalis possesses comparatively high biological values. Thus an herbal aerosol for arthritis may be formulated using the bark extract of C.officinalis as key ingredient and the therapeutic product will be of cost effective, easy to use and pocket friendly.

Study of Medicinal Plants: Ocimum Sanctum & Tinospora Cordifolia,
Bioresources to Characterize Bioactive Compounds Using Soxhlet Apparatus

  Abstract: This study researched the phytochemical organization of two significant restorative plants - Ocimum sanctum (Tulsi) and Tinospora cordifolia (giloy). Leaf tests of O. sanctum and stem tests of T. cordifolia were gathered, cleaned, dried and ground into powder. Extractions were performed involving ethanol and hexane solvents in a Soxhlet device. Qualitative phytochemical screening tests for a variety of bioactive compounds, including proteins, carbohydrates, terpenoids, flavonoids, saponins, tannins, steroids, alkaloids, glycosides, and phenolics, were performed on the extracts. The majority of the phytochemicals tested were found in O. sanctum extracts, while fewer compounds were found in T. cordifolia extracts. In particular, flavonoids, tannins, alkaloids, sugars, terpenoids and proteins were distinguished in O. sanctum but not in T. cordifolia separates. The two plants showed the presence of saponins, steroids, heart glycosides and phenolics. The study sheds light on the bioactive compound profiles of these significant medicinal plants and highlights their potential for further research in the fields of drug discovery and herbal formulation development. Future possibilities for research on these plants in regions like pharmacology, nutraceuticals and maintainable assets the executives are additionally talked about.
