  • Latrophilin-2 is a novel receptor of LRG1 that rescues vascular and
           neurological abnormalities and restores diabetic erectile function

      A protein that initiates multiple tissue repair processes could offer effective treatment for erectile dysfunction arising from diabetes mellitus. High blood sugar levels can lead to serious damage to nerves and blood vessels, with impaired male sexual function one potential consequence. Guo Nan Yinof the Inha University School of Medicine, Incheon, South Korea, and colleagues have identified a molecular pathway that can be targeted to reverse such damage. They determined that a protein called LRG1 acts on a receptor called LPHN2 to stimulate blood vessel and nerve growth. LPHN2 is highly expressed in the penile tissue of diabetic humans and rodents, and direct injection of LRG1 effectively restored erectile function in mice.   • Understanding the contextual functions of C1q and LAIR-1 and their
           applications

      Research into the activities of the protein C1q, involved in a cascade of molecular interactions of the immune response called complement activation, is revealing new details of the protein’s role and opening up possible new therapeutic opportunities. Myoungsun Son at Feinstein Institutes for Medical Research in Manhasset, USA, reviews the involvement of C1q in infection, autoimmunity, inflammatory diseases and tumors. The interaction of C1q with a receptor protein called LAIR-1 seems to be particularly significant. LAIR-1 is present in the membrane of most blood-forming cells and is involved in maintaining the healthy balance of cellular activities referred to as homeostasis.   • The optimized core peptide derived from CABIN1 efficiently inhibits
           calcineurin-mediated T-cell activation

      A peptide with therapeutic potential binds strongly to the cellular enzyme calcineurin and may prove valuable in anti-cancer and autoimmune disease treatments. Many cancers and autoimmune diseases are linked with overactivation of a key calcineurin-related pathway which is heavily involved in T cell activation. This pathway has long been a therapeutic target, but existing drugs show problems with stability and delivery, and can cause serious side effects. One known inhibitor of calcineurin is the protein CABIN1, but precisely how well it binds and how useful it may be is unclear. Now, Hong-Duk Youn at Seoul National University College of Medicine, South Korea, and co-workers have identified how one specific peptide from CABIN1 binds strongly to calcineurin.   • Detoxified pneumolysin derivative ΔA146Ply inhibits autophagy and induces
           apoptosis in acute myeloid leukemia cells by activating mTOR signaling

      A mutated form of the bacterial protein pneumolysin offers a new approach to treating acute myeloid leukemia (AML), due to its ability to stimulate cancer cells to undergo a form of cell suicide called apoptosis. Researchers in China led by Xuemei Zhang at Chongquing Medical University studied the effects of a pneumolysin derivative on cultured human and mouse AML cells. They identified the mechanism by which this derivative activates a known molecular signaling system to inhibit the process of autophagy, in which cells routinely ‘clean up’ degraded or unnecessary components during normal maintenance. This inhibition of autophagy then induced the apoptosis that killed cancer cells. The effect became more pronounced when the pneumolysin derivative was combined with the existing autophagy-inhibiting drug chloroquine.   • Comparative characterization of 3D chromatin organization in
           triple-negative breast cancers

      The 3D architecture of the genome is dramatically altered in an aggressive form of breast cancer, leading to changes in the regulation of gene expression that can fuel tumor growth. A team from South Korea, led by Hyeong-Gon Moon of Seoul National University College of Medicine and Daeyoup Lee of the Korea Advanced Institute of Science and Technology, Daejeon, detailed how chromosomes are positioned and folded within the nucleus of cell liness from five different subtypes of breast cancer. They found that triple-negative breast cancers displayed the most extreme reorganization of their genomes, a pattern also observed in biopsy tissues taken from patients with this subtype of cancer.   • Upregulation of the serine palmitoyltransferase subunit SPTLC2 by
           endoplasmic reticulum stress inhibits the hepatic insulin response

      A lipid molecule called ceramide is key to regulating the body’s insulin response, which controls blood sugar, and thus may hold keys to new treatments for metabolic diseases such as diabetes. Although ceramide levels were known to be raised in obesity and diabetes, the mechanism remained unclear. Tae-Sik Park at Gachon University, Sungnam, South Korea, and Sang-Ho Kwon at Augusta University, USA, and co-workers investigated how excess ceramide production is triggered and the blood sugar regulation consequences. They found that the liver-specific SPTLC2 transgenic mice fed a high-fat diet had increased levels of an enzyme activity of serine palmitoyltransferase which led to synthesis of high levels of ceramide in the liver. The high ceramide levels suppressed insulin signaling, imbalancing blood sugar levels and causing liver toxicity.   • Functional and pathologic association of aminoacyl-tRNA synthetases with
           cancer

      Enzymes called aminoacyl-tRNA synthetases (ARSs), which play a central role in all life, are becoming implicated in several aspects of cancer in ways that may lead to new approaches for prevention, detection and treatment. ARS enzymes catalyse the ligation of amino acids to transfer RNA molecules to allow amino acids to combine in the correct sequences to form proteins. Jung Min Han, Sunghoon Kim and colleagues at Yonsei University, Incheon, South Korea, review researches implicating ARS enzymes and the genes that code for them in a variety of cancers.   • Suberoylanilide hydroxamic acid suppresses axonal damage and neurological

      Analysis of the molecular processes involved in the progression of hemorrhage-induced brain damage has yielded a potential therapeutic target. Fei Liu and Aihua Liu at the Central South University in Changsha, China, and co-workers examined mechanisms behind the extensive brain damage that can be caused by subarachnoid hemorrhage, bleeding in a space between the brain and its surrounding membranes. In experiments on human samples and rat models, the researchers showed that the TDP-43 protein was over-expressed in cerebrospinal fluid after hemorrhage, and that the build-up of this protein aggravated nerve cell damage and dysfunction. TDP-43 is regulated by a pathway involving a potent enzyme involved in regulation of gene expression called HDAC1.
