    • Abstract: Title: Systematic review on nine hallmarks of neurodegenerative disease
      Authors : Veeman, Dhinakaran; Dhamodharan, Duraisami; Surendhar, GJ; Natrayan, L; Stalin, B; Ramaswamy, Shanmugam; Jule, Leta Tesfaye; Krishnaraj, R: Alzheimer's disease (AD) and Parkinson's disease (PD) are the primary diseases in neurodegenerative diseases. Nowadays, AD is common in one of the ten individuals whose age is more than 65, and its prevalence is kept on increasing with aging. Very few treatments and no effective treatments are available for curing neurodegenerative diseases. Pathogenesis of neurodegenerative diseases, Alzheimer's disease, Parkinson's disease, and their association with the nine hallmarks of aging were clearly described in this review. Instability in genomic, attrition in telomere, alterations in epigenetics, proteostasis loss, dysfunction in mitochondria, senescence in cells, sensing of deregulated nutrition, exhaustion of stem cells, and alterations in intercellular communication are the nine biological hallmarks of Aging. Improving the medical facilities for neurodegenerative diseases is very much essential. Doctors and researchers are doing surplus research to overcome the unavailability of proper treatments for such neurodegenerative diseases. Reason and the causes behind the diseases and their effects are explained in this review to enhance the further research to help the society.Page(s): 249-257

  • Identification of hub genes in airway epithelial cells of asthma patients
           by WGCNA and PPI network

    • Abstract: Title: Identification of hub genes in airway epithelial cells of asthma patients by WGCNA and PPI network
      Authors : Liu, Zhigang; Dai, Na; Chen, Dehong; Gao, Jiyong; Zheng, Junqing; Wang, Jin: Bronchial asthma is a common chronic disease of airway inflammation, high mucus secretion and airway hyper responsiveness. The pathogenetic mechanisms of asthma remain unclear. In this study, we aimed at identifying genes playing an import role in disease-related pathways in airway epithelial cells of asthma patients. Microarray data GSE41861 of asthma airway epithelial cells was used to screen differentially expressed genes (DEGs) through GEO2R analysis. The weighted gene co-expression network analysis (WGCNA) was performed to identify gene co-expression network modules in bronchial asthma. The DAVID database was then used to perform functional and pathway enrichment analysis of these DEGs. In addition, we have conducted protein-protein interaction (PPI) network of DEGs by STRING, and eventually found key genes and significant modules. A total of 315 DEGs (111 up-regulated and 204 down-regulated) were identified between severe asthma and healthy individual, which were mainly involved in pathways of cilium assembly, cilium morphogenesis, axon guidance, positive regulation of fat cell differentiation, and positive regulation of cell substrate adhesion. A total of 60 genes in the black module and green module were considered to be correlated with the severity of asthma. Combining PPI network, several key genes were identified, such as BP2RY14, PTGS1, SLC18A2, SIGLEC6, RGS13, CPA3, and HPGDS. Our findings revealed several genes that may be involved in the process of development of bronchial asthma and potentially be candidate targets for diagnosis or therapy of bronchial asthma.Page(s): 258-267

      Authors : Kotrikadze, Nanuli; Tuphinashvili, Tamar; Alibegashvili, Manana; Ramishvili, Liana; Nakashidze, Irina; Artmeladze, Mukhamed; Gordeziani, Manana; Zibzibadze, Maia; Khazaradze, Ana; Sepiashvili, Bela; Ahmad, Sarfraz: The indicators for structural analysis of blood formed elements are prominent in the assessment of pathologies, diagnostics and the degree. Therefore, we aimed to evaluate the ongoing alterations that reflect on the structural characteristics of blood formed elements based on the hormonal imbalance among menopausal women with uterine tumors. Blood samples from the women with benign (n=20), malignant (n=20) uterine tumors, and healthy menopausal women (control, n=20) were used. Enzyme-linked Immunosorbent assay (ELISA) kits were used for the quantitative determination of hormones. The blood formed elements ultrastructure observations were conducted using transmission electron microscope. Compared to control (33.8±0.7 pg/mL), estradiol level was higher in benign (45.7±0.9 pg/mL) and malignant (70.7±3.7 pg/mL) cases (P< 0.001). Similar pattern was noted in testosterone levels [control=0.38±0.03 ng/mL, benign=0.55±0.04 ng/mL (P< 0.01), malignant=1.56±0.14 ng/mL (P< 0.001)] was higher in malignant cases. In contrast, progesterone levels were decreased in the disease cases [control=0.93±0.05 ng/mL, benign=0.44±0.003 ng/mL, malignant=0.31±0.02 ng/ml (P< 0.001)]. Assessments of the morphologic structure of erythrocytes revealed pathological forms of erythrocytes (poikilocytosis) in case of benign, as well as in malignant tumors. particularly target cells (codocytes), hamlet cells, teardrop cells (dacrocytes), sickle cell (drepanocytes) erythrocytes. Using ELISA and transmission electron microscopy our results demonstrate that in case of malignant uterine tumor quantitative/structural changes occur in blood formed elements indicating ongoing alterations in hormonal imbalance. Assessing these changes in structural characteristics would be useful in examining uterine pathologies and subsequent treatment plans.Page(s): 268-275

  • Computational studies on the structural variations of MAO-A and MAO-B

    • Abstract: Title: Computational studies on the structural variations of MAO-A and MAO-B inhibitors - An in silico docking approach
      Authors : Nambiar, Megha P; Jayadevan, Sarayu; Babu, BK; Biju, AR: The neurological disorder is a concerning problem in the present social scenario. The malfunction of the monoamine oxidase (MAO) enzyme is the responsible factor behind this disorder because this enzyme regulates the metabolism of monoamine neurotransmitters. This work aimed to design and propose the best MAO inhibitors through extensive computational analysis so that the favourable drug-like molecules could be identified for future synthesis. The drugs selected in this study were three MAO-A inhibitors namely Moclobemide, Tolxatone and Brofaromine and two MAO-B inhibitors namely Selegiline and Rasagiline. By substituting hydrophilic and hydrophobic groups at the specified positions, structural variations were designed for each drug. The designed variations and their parent drugs were optimized (basis set is B3LYP/6-311G(d, p)) and the optimized structures were docked to the target using PyRx software. The binding energy of each variation was compared to that of parent drug. The drug-likeness, physicochemical properties (solubility, polarity, flexibility, gastrointestinal absorption, saturation etc.) and toxicity of the lower binding energy variations were analysed using the swissADME, Osiris property explorer and ProTox-II servers. The interacting residues of the enzymes were obtained from the LigPlot+ program. The safe and low binding energy variations with favourable drug properties are suggested for further drug research.Page(s): 276-295
