  • The effect of natural substances on the radicle development of certain
           plant seeds during the early germination stage

    • Abstract: Successful seed germination is crucial in many plants. Natural substances and chemicals have been used as growth enhancement tools. In the meantime, natural substances are considered environmentally friendly and cost-effective. Hence, the present study was focused on the effects of natural substances such as red rice, white rice washed water, orange coconut, green coconut, and matured coconut water on radicle development of black gram, pumpkin, rice, and maize. Twenty-five healthy sterilized seeds (85%germination rate) of the above plants were pre-soaked in 30 ml of the natural suspension for 24 hours separately at room temperature and repeated twice. Then seeds were transferred to the sterilized petri dish with moistened filter paper and mean values of length of the radicle were measured. Data were subjected to one-way analysis of variance (ANOVA) (P<0.05) followed by Tukey test. Natural substances have not affected radicle development of the tested seeds, except pumpkin, and maize after 24hours whereas in the control radicle length of these two species was 5.48 and 0.68mm, respectively. However, interestingly after 48hours, radicle formation of most tested seeds was highly accelerated by red rice and white rice washed water, except for black gram. Hence, orange coconut, green coconut, and matured coconut water have showed an inhibitory effect on root length development, except for maize at 48 hours. On the other hand, among the selected natural substances red rice and white rice washed water revealed better effects on radicle development. So, they can be used as an environmentally friendly method to stimulate the radicle development. Published on 2021-08-05 00:00:00
       
  • Prime labeling of special graphs

    • Abstract: Prime labeling is the most interesting category of graph labeling with various applications. A graph G= (V(G), E(G)) with v(G) vertices are said to have prime labeling if its vertices are labeled with distinct positive integers 1,2,3,……, v such that for each edge uv e E(G) the labels assigned to u and v are relatively prime, where V(G) and E(G) are vertex set and edge set of G, respectively. Therefore, the graph G has a prime labeling whenever any of two adjacent vertices can be labeled as two relative prime numbers and is called a prime graph. In our work, we focus on the prime labeling method for newly constructed graphs obtained by replacing each edge of a star graph K1,n by a complete tripartite graph K1,m,1 for m = 2,3,4, and 5, which are prime graphs. In addition to that, investigate another type of simple undirected finite graphs generalized by using circular ladder graphs. These new graphs obtained by attaching K1,2 at each external vertex of the circular ladder graph CLn and proved that the constructed graphs are prime graphs when n ≥ 3 and n ≠ 1 (mod3) . Finally, focus on another particular type of simple undirected finite graph called a scorpion graph, denoted by S(2p,2q,r) . The Scorpion graph gets its name from shape, which resembles a scorpion, having 2p + 2q + r vertices p ≥ 1, q ≥ 2, r ≥ 2)are placed in the head, body, and tail respectively.   • Effect of soil applied organic amendments and microbial inoculant on the
           improvement of growth and yield of soybean (Glycine max L.)

    • Abstract: The usage of organic amendments and microbial inoculant together can be a reliable solution to avoid soil heavy metal pollution and to increase crop production in sustainable farming. This study was done to determine the effectiveness of organic amendments and Bradyrhizobium sp. in improving soybean (Glycine max L.) growth and yield. Control (T1) and the other seven treatments were comprised as, field soil + compost (15% by weight) (T2), field soil + wood biochar (15% by weight) (T3), field soil + synthetic chemical fertilizer [0.01 kg m–2 urea, 0.01 kg m–2 P2O5 (triple super phosphate) and 0.008 kg m–2 K2O (muriate of potash)] (T4), field soil + Bradyrhizobium inoculum. (108 CFU ml-1) (T5), field soil + compost (15% by weight) + Bradyrhizobium inoculum (108 CFU ml-1) (T6), field soil + wood biochar (15% by weight) + Bradyrhizobium inoculum. (108 CFU ml-1) (T7), field soil + synthetic chemical fertilizer (111.5 kg N ha–1, 111.5 kg P ha–1, and 82.9 kg K ha–1) + Bradyrhizobium inoculum. (108 CFU ml-1) (T8), The experiment was arranged in a completely randomized block design with ten replicates. In wood biochar amendment together with Bradyrhizobium sp. inoculation significantly (p< 0.05) enhanced plant height and the soybean yield compared to all the other treatments. All the organic amendments added treatments were positively influenced by the inoculation of Bradyrhizobium sp. The present study concludes that soybean yield can be significantly enhanced by the application of Bradyrhizobium sp. and wood biochar amendment. Published on 2020-12-18 00:00:00
       
  • Anti-cancer activity of Fluggea leucopyrus willd (Katupila) against human
           ovarian carcinoma and characterization of active compounds

    • Abstract: Fluggea leucopyrus Willd has been used in the treatment for cancers in the traditional system of medicine in Sri Lanka. This study was aimed on evaluation of anti-cancer activity of F. leucopyrus against Human Ovarian carcinoma and identification of active compounds. Anti-cancer activity of ethyl acetate (EtoAc) extract of leaves and two compounds isolated from EtoAc extract were evaluated using MTS and hTERT assays on Human Ovarian carcinoma (A 2780). MTS assay showed a significant anti-proliferation activity giving IC50 (concentration of extract/compound that inhibits 50% of the activity) values of 36.35±0.17μg/mL, 12.36±1.0 μg/mL and 48.53±1.43 μg/mL for EtoAc extract and two compounds (compound-1 & compound-2) respectively. A rapid depletion of hTERT content in Human Ovarian cancer cells was observed for the compound-2 with the concentration range 50-200 μg/mL. The compounds-1 was identified as bergenin, a known compound, whereas the compound-2 was identified as a diastereoisomer of bergenin (at C-9 and C-14,) with the help of NMR and Mass spectral data, and with the comparison of reported data. Brine Shrimp Lethality assay for EtoAc extract showed LC50 (Lethal concentration) values of 2779.63 μg/mL, implying that no toxicity of plant extract towards brine shrimp. As conclusion, anti-cancer activity of leaves of F. leucopyrus against Human Ovarian cancers is confirmed in this study, and bergenin and its isomer were identified as the responsible compounds for the anti-cancer activity of this plant.   • Impact of salinity on seed germination and seedling performance of tomato
           (Solanum lycopersicum L.) cv KC-1

    • Abstract: An experiment was conducted to evaluate the effect of salt stress on seed germination and seedling performance of Solanum lycopersicum L. (tomato) cv KC-1 under controlled environment and also to select salt tolerant seedlings for plant regeneration. This experiment was laid out in a completely randomized design with three replications. Uniform seeds of tomato cv KC-1 were surface sterilized and placed on sterilized filter paper in the petridishes. Subsequently they were wetted with 5 ml of distilled water as a control or one of the NaCl (sodium chlorite) salt solutions (0, 20, 40, 60 and 80 mM NaCl. Seed germination percentage were calculated and seedling growth attributes were recoreded after seeding. Results showed that germination percentage decreased with increasing salinity levels (0 mM, 20 mM, 40 mM, 60 mM, 80 mM). Germination percentage was less than 50% for seed treated with NaCl above the level of 40 mM. Minimum value (18.3%) was recorded in 80 mM of salinity. Shoot and root lengths, dry and fresh weights of shoots and roots, relative water content and total dissolved salt content of first leaves (cotyledons) were significantly affected (P<0.001) by salt stress after 12 days of seeding. Salinity levels had a considerable effect on all parameters tested in this study. Salt content of first leaves increased with increasing NaCl concentrations. Germination and early seedling growth of Tomato cv. KC-1 was greatly affected in salt concentrations above 40 mM NaCl levels under controlled environmental conditions.   • A case study on the outbreak of paederus dermatitis caused by rove beetles
           (coleoptera: Staphylinidae) in women´s hostels of eastern university, Sri
           Lanka

    • Abstract: Disreputable outbreaks of Paederus dermatitis (PD) has been predominant in women´s hostels of Eastern University and its surroundings. This study was undertaken with the objectives of recommending precautions and treatment for students and local residents around the university from 2015-2019. Sampling was done by light traps and hand collection. Photographs of beetles and skin lesions were taken. Hostel surrounding was inspected to find out the breeding sites of beetles. Case review among affected students and staffs was carried out through face to face interview. Approximately, 145 students were principally affected on the exposed body parts of which 95% of students developed specific symptoms including erythema, edematous plaques with vesicles, micropustules and about 5 % of students reported no specific symptoms. PD was more prevalent during rainy seasons than drought months. The students in women´s hostels were more frequently affected than in men’s hostels. Population dynamics, seasonal variations, life history and behavior pattern of rove beetles should be thoroughly investigated prior to their control. Published on 2020-07-27 00:00:00
       
  • Exponential smoothing on forecasting dengue cases in Colombo, Sri Lanka

    • Abstract: Prediction of number of people to be infected is an essential component in studying any leading diseases. Particularly it is important in dengue disease as it is the most critical mosquito-borne viral disease in the world. The number of reported dengue cases gradually increased all over the world as well as in Sri Lanka. In Sri Lanka, the majority of dengue cases reported in the Colombo district. The authors applied exponential smoothing technique in order to model and forecast dengue cases in Colombo, Sri Lanka. Data consist of monthly reported dengue cases in Colombo district from January 2010 to May 2019. January 2010 to February 2019 data used for model building and rest of the data used for model validation. Both original cases and log transformed cases considered for modelling and Holt Winters smoothing suits well with both cases. Best model in each case and finally the most parsimonious model within these two best models were selected by considering AIC, BIC, MAE, RMSE and MAPE measures. The most parsimonious model fits on log transformed dengue cases. Using the most parsimonious model predictions were made for June to August 2019. It can be concluded that the best model able to fit on the data in an adequate level and reported dengue will increase slowly during the prediction period. Published on 2020-07-27 00:00:00
       
  • Small scale biodiesel production from Sargassum sp and optimization of
           conditions for yield enhancement

    • Abstract: Production of biofuel from excessive natural resources and underutilized plant materials has drawn global attention due to its cost effectiveness, eco friendliness and sustainability compared to the conventional fossil fuels. This work was aimed to select the best marine flora available in the Northern Sri Lankan sea to produce biodiesel and to optimize the conditions to enhance the yield. Sargassum sp., Ulva fasciata, Turbinaria ornata, Gelidium sp. and Thalassia sp. were collected from the coastal region of the Jaffna peninsula and washed thoroughly sun-dried. The organic solvents used to extract oil from these algal and sea grass species were n-hexane and di-ethyl ether, while alkaline catalysts were used to convert the extracted oil into biodiesel via trans-esterification reaction. When the oil extraction was done separately from the five dried marine plant species using n-Hexane and Di-ethyl ether as solvents, significantly higher quantity of oil was obtained from Sargassum sp. than the other tested algal and plant species, hence Sargassum sp. was selected for further studies. When the conditions for the extraction of oil from Sargassum sp. such as Sargassum to solvent(n-Hexane: Di-ethylether) ratio (0:200), amount of algal biomass (100g), longer contact time (24h), molar ratio (4:1), catalyst amount (0.6%), temperature (600C), reaction time (25min) were optimized before the conversion of oil into biodiesel via trans-esterification reaction, there was a significant increase in the quantity of extracted oil by 11.7 times than the non-optimized conditions. Since the density of the biodiesel (867.3 kg/m3) meets the international standards, the quality of the biodiesel produced from Sargassum sp. could be recommended as satisfactory.   • Screening and biochemical identification of Tef (Eragrostis tef Zucc.)
           Trotter) endophytic bacterial species with plant-growth-promoting, biotic
           and abiotic stress tolerance properties

    • Abstract: Endophytic bacteria play many important beneficial roles in the metabolism and physiology of the host plants, including fixing atmospheric nitrogen, solubilizing phosphates, synthesizing phytohormones, sequestrating iron, suppression of the ethylene production, and inhibiting pathogens. The present study was conducted to screen and identify endophytic bacterial species with plant growth-promotion, biotic and abiotic stress tolerance properties to develop bio inoculants. For this purpose, root and seed samples of different cultivated tef varieties were collected. A total of 195 pure endophytic bacterial colonies were isolated and screened for PGP traits, biotic and abiotic stress tolerance factors. Fourteen endophytic bacterial species or strains were identified using the Biolog microbial identification system. Among the identified endophytes, the majority of them utilized different carbon sources tagged on microplates. Three potential endophytic bacterial species or strains were evaluated for seed germination and seedling growth ability using tef seeds. Tef seeds inoculated with potential endophytes showed 100 % germination performance on 3 to 4 days after inoculation. An increase in mean shoot and roots length of the inoculated seeds were observed up to 3 and 2.6 cm respectively. Vigor index of seedling was measured from 460 to 520 on the last day of the experiment. These promising identified tef endophytic bacterial species or strains having PGP, biotic and abiotic stress tolerance properties could be candidate organisms for the development of microbial inoculants as single strain or consortia. This strategy would work towards solving the problem of over-utilizing chemical fertilizers, and reducing environmental pollution while increasing the yield and productivity of tef crops and other agricultural plants.
