  • PROGNOSTİC VALUE OF Kİ67 İN BORDERLİNE MUCİNOUS
           OVARİAN TUMORS

      Abstract: ObjectiveOvarian borderline tumors (OBTs) are an intermediatetype of ovarian neoplasm. Ovarian mucinous borderlinetumors (MBT) are the second most common subtypeof OBT. Because of their uncertain pathogenesisand biological behaviour they cannot be classifiedclearly into benign and malign categories. There is aneed to identify a parameter that provides more prog-nostic information about the clinical course and can beevaluated easily in routine practice. Ki67 is a routinelyused immunohistochemical marker that is strongly associatedwith cellular proliferation, including tumor cells.We aimed in this study to evaluate the prognosticsignificance and to determine prognostic cut-off valueof Ki67 counting in the MBTs and to investigate its relationshipwith clinicopathologic features, recurrence,and long term survival status and also to provide moreinformation to the literature.Material and MethodA total of 20 cases diagnosed with ovarian mucinousborderline tumors were identified. Immunohistochemicalstudies were performed on the most representativesample of the tumor. Positive signal was nuclear and itwas evaluated by two pathologists.ResultsKi67 proliferating index (P.I) value with a median of15%(1-47%) and a mean of 16% Ki67 P.I. reachedstatistically significance at 20% cut-off value withprogression and at 30% cut-off value with 5-year OS.(p=0.021 and p=0.032, respectively). Although theKi67 P.I. tended to increase with age, there was nostatistical association between Kİ67 P.I. and other clinicopathologicalparameters including tumor size, laterality,capsule integrity, intraepithelial carcinoma andfocal atypia.ConclusionIn our study, recurrence was more frequent when Ki67was greater than 20%, and 5-year survival was lowerwhen Ki67 was greater than 30%, suggesting thatthe Ki67 index was associated with a more aggressivecourse and death. Therefore, we predicted thatKi 67 would be useful in determining the frequency offollow-up of patients and predicting their prognosis.However, we think that this result should be supportedby larger series.

  • PRESENCE OF POSTOPERATIVE HISTOPATHOLOGICAL MALIGNANCY IN PATIENTS WITH
           ATYPIA OF UNDETERMINED SIGNIFICANCE

      Abstract: ObjectiveThyroid cancer is the most common endocrineMalignancy, and nodules characterize its clinicalpresentation. 'Atypia of undetermined significance'(AUS) in biopsies performed for thyroid pathologiesis an essential concern for clinicians due to differentmalignancy rates in the literature. In this study, weaimed to discuss the malignancy rates in patients whounderwent thyroid surgery for AUS.Material and MethodThe demographic and histopathologic data of 174patients diagnosed with AUS on fine needle aspirationbiopsy (FNAB) and who underwent thyroid surgery inour clinic between 2016 and 2021 were retrospectivelyanalyzed. According to histopathologic features,patients were divided into two groups, malignantand benign, and the relationship with age, gender,and nodule diameter was investigated. Statisticalsignificance between groups was determined usingChi-square and Student t-tests. P value 0.05).ConclusionAccording to the Bethesda classification, themalignancy rate for AUS is 15-30%. In our study, themalignancy rate was 43.5%, similar to studies publishedin our country. Because of the high malignancyrate, total thyroidectomy should be considered analternative to lobectomy in cases planned for surgerywith a diagnosis of AUS.

  • PROTECTIVE ROLE OF ANKAFERD BLOOD STOPPER ON CADMIUM-INDUCED ACUTE
           NEPHROTOXICITY

      Abstract: ObjectiveCadmium (Cd) is a very toxic and carcinogenic heavymetal that can cause harmful effects on human health.Toxicity may develop due to Cd exposure, especially inthe kidneys. Ankaferd blood stopper (ABS) is a herbalmix that is used for its hemostatic properties in surgery.Also, ABS enhances wound and tissue healing. In thisstudy, we aimed to evaluate the possible ameliorativeeffects of ABS in Cd-induced renal damage.Material and MethodThirty-two male rats were randomly divided into 4groups: control, Cd (cadmium chloride, 2.5 mg/kgsingle dose, ip), ABS (ABS, 1.5 ml/kg single dose ip),and Cd+ABS (cadmium chloride, 2.5 mg/kg single doseip- ABS, 1.5 ml/kg single dose ip). At the end of theexperiment, urea and creatinine levels were analyzedfrom the rats’ serum. In addition, total oxidant status(TOS), total antioxidant status (TAS) levels, superoxidedismutase (SOD), and glutathione peroxidase (GPx)activity were measured spectrophotometricallyfrom renal tissues. The oxidative stress index (OSI)was calculated from TOS and TAS levels. Also, weevaluated alterations in the mRNA expression of Bcl-2-associated X protein (Bax), B-cell-lymphoma-2 (Bcl-2), silenced information regulator 1 (SIRT1), and p53in kidney tissue of rats by using the qRT-PCR method.ResultsIn the Cd group, serum urea, creatinine levels, andtissue oxidative stress markers, TOS and OSI weresignificantly higher while Gpx activity was significantlylower than in the control group (p

  • ALVARDO SCORE AND COMPUTED TOMOGRAPHY FOR PREVENT FROM NEGATIVE
           APPENDECTOMY

      Abstract: ObjectiveThe functionality of computed tomography andAlvarado score were investigated to prevent negativeappendectomy.Material and MethodPatients who had appendectomy surgery at NiğdeÖmer Halisdemir University Training and ResearchHospital between June 2020 and March 2021 wereretrospectively scanned. Correlations with pathologyresults were made.ResultsA positive correlation was found between the highAlvarado score and the diagnosis of appendicitis incomputed tomography. The lower alvarado scoreof those without appendicitis than those with acuteappendicitis; It was found statistically significant thatthose with acute appendicitis were lower than thosewith perforated-gangrene-plastrone appendicitis.ConclusionAlvarado scoring system is less specific than computedtomography and it is possible to reduce the negativeappendectomy rate with radiological examinations.

      Abstract: ObjectiveTo investigate the success of imaging methods andthe effect of preoperative findings on postoperativeresults in patients who underwent surgery for primaryhyperparathyroidism (p-HPT).Material and MethodIn this study, the data of 50 consecutive patients whounderwent p-HPT surgery at the General SurgeryClinic of Yuzuncu Yil University Faculty of Medicinebetween January 2008 and December 2010 wereprospectively analyzed. During the study period,the patients’ demographic data, clinical findings,preoperative and postoperative laboratory results,findings of ultrasonography, scintigraphy, andcomputed tomography performed for localization inthe preoperative period, and surgery and pathologyreports were evaluated and recorded.   • EXCESSIVE USE OF FLAXSEED MAY POSE A THREAT TO KIDNEY TISSUE: AN
           EXPERIMENTAL STUDY

      Abstract: ObjectiveWe aimed to investigate the effects of flaxseed,which is widely used due to its healing effects, on thekidney tissue when the daily recommended dose isexceeded.Material and MethodsWe divided the male Wistar Albino rats into fourgroups of eight, namely the control, OD-1 (1.4 mg/kg/d), OD-2 (2.8 mg/kg/d), and OD-3 (5.6 mg/kg/d)groups. Ground flaxseed was administered to eachoverdose group by oral gavage for 7 days. At theend of the experiment, blood and kidney sampleswere collected. Blood samples were centrifuged andserum was separated for renal function analyses.The half of the kidney samples were stored at -20°C until biochemical analyses and we fixed theremaining tissues with 10% buffered formalin forhistopathological analysis.ResultsSerum urea and BUN levels were significantlyincreased in all overdose groups compared to thecontrol group (for all p˂0.05). TAS levels significantlydecreased in the OD-3 group compared to the control(p=0.044). There was an increase renal TOS levelsin all overdose groups compared to the controland also, OSI level of the OD-3 group significantlyincreased compared to the control (p=0.016). Whilehistopathological changes were not observed in OD-1and OD-2 groups compared to the control group,there were non-significant histopathological changesin OD-3 group.ConclusionIt was concluded that the use of flaxseed above therecommended daily dose can pose a threat to kidneytissue. So, while being used for its benefit, in order notto be exposed to its toxic effects, overuse and longtermuse should be avoided.

  • EVALUATION OF IMMUNOHISTOCHEMICAL NEOVASCULAR PSMA AND CD34 EXPRESSIONS IN
           THYROID TUMORS

      Abstract: ObjectiveThe aim of this study was to investigate the placeof Prostate Specific Membrane Antigen (PSMA)expression in differential diagnosis and its effect onprognosis in thyroid tumors.Material and MethodImmunohistochemical PSMA and CD34 were appliedto paraffin blocks of 95 patients diagnosed withdifferent histological types of thyroid cancer. Thepercentage of vascular expression in the tumoral andextratumoral stroma and the density score in the tumorepithelium were evaluated and grouped according toPSMA expression level. Final PSMA expression wasdetermined by multiplying intensity and percentagepoints.An increase in expression was observed with PSMAin areas with neovascularization in thyroid tumors. Novascular staining was detected with PSMA in nonneoplasticthyroid tissue. No increase in expressionwas seen in Non-invasive folicular thyroid neoplasmwith papillary like nuclear features (NIFTP) casescompared to aggressive tumorsConclusionIt was determined that increased PSMA expressionwas associated with poor prognosis, and it was foundthat it can be used to differentiate NIFTP cases fromother tumors.

  • EVALUATION OF LIVER ELASTICITY USING pSWE AND 2D-SWE TECHNIQUES IN HEALTHY
           CHILDREN

      Abstract: ObjectiveShear wave elastography (SWE) is a newultrasonography technique that can non-invasivelyevaluate liver stiffness. The efficacy of SWE in adultshas been demonstrated by numerous studies, butthere are few SWE studies of pediatric groups. Sincethe liver stiffness values (LSV) vary according to thedevice and the SWE method used, standard referencevalues have not been established as yet. The aim ofthis study was to establish the reference values ofnormal liver stiffness in healthy children according tothe two SWE methods.Materials and MethodLiver stiffness values were measured using the twodifferent SWE methods of point SWE (pSWE) and2-dimensional SWE (2D-SWE) in 107 healthy childrenaged 8-18 years with no liver disease. The pSWE(EPQ) and 2D-SWE (EQI) measurements were takenin one session on the same device (Philips Epiq Elite)and after at least 4 hours of fasting. The effects onliver elasticity values were investigated of gender,age, body mass index (BMI), some biochemicalmarkers (such as fasting blood glucose, aspartateaminotransferase (AST), alanine aminotransferase(ALT), albumin, platelet, INR) and liver size.ResultsThe mean LSV was 4.04±0.84kPa ((95% confidenceinterval (CI):3.67-4.42)) on pSWE and 4.41±0.71kPa(95% CI:4.09-4.72) on 2D-SWE. The upper limit LSVswere found to be 4.42kPa and 4.72kPa, respectively.The LSVs measured on 2D-SWE were found to behigher than on pSWE ((median; min-max, 4.34kPa;2.82-6kPa and 3.86kPa; 2.55–5.78, respectively,(r=0.59, p

  • EVALUATION OF BURNOUT IN OPERATING ROOM WORKERS DURING THE PANDEMIC PERIOD

      Abstract: ObjectiveWith the coronavirus pandemic, all healthcare professionalsin general, and operating room personnel inparticular, had to work under difficult conditions. Theaim of this study is to examine burnout levels in operatingroom workers.Material and MethodIn February 2021, the Copenhagen burnout scale wasadministered to 162 healthcare workers working in theoperating room. The burnout levels of the participantswere measured under three sub-headings: personal,work and pandemic-related. Scores above 50 pointswere considered high burnout.ResultsPersonal burnout was 66.9 ±19.5, work-related burnoutwas 66.8 ± 16.1, pandemic-related burnout was74.3±12.1. It was found that females had higherscores for all subscales.ConclusionThe burnout levels of the operating room workersduring the pandemic period were determined at highlevels. Actions to reduce or prevent burnout are neededin this regard.

  • PERMANENT ATRIAL FIBRILLATION IS RELATED WITH AN INCREASE IN DEPRESSION
           AND ANXIETY DISORDER IN TURKISH POPULATION

      Abstract: ObjectiveThe aim of this study was to explore the relationshipwith Permanent Atrial Fibrillation (AF) and depression/anxiety disorder in Turkish population. Wethought that more data exploring needs of AF patientsand the associated factors as well as the impact ofanxiety/depression on needs are limited.Material and MethodForty-five consecutive patients with permanent AFand forty-five control group (sinus rhythm) were enrolledin this study. All participants completed BeckDepression Scale (BDS) and Hamilton Anxiety Scale(HAS). BDS was used to evaluate mood disordersand depression. HAS was used to evaluate severityof anxiety level and symptom distribution. BDS andHAS were compared between two groups.ResultsThere was no difference between two study groupin terms of clinical and demographic characteristics.Mean scores of BDS and HAS were significantlyhigher in patients with permanent AF (p< 001).ConclusionOur study demonstrates that patients with PermanentAF had higher BDS and HAS scores compared tocontrol group. In our knowledge our study is the firstresearch that examining the relationship betweenpermanent AF and depression/anxiety disorder inTurkish population.

  • EFFECT OF WHEATGRASS ON HUMAN LYMPHOCYTE CELLS

      Abstract: ObjectiveWhile chemotherapeutic drugs are effective at eliminatingcancer cells, they can also damage healthycells. The aim of this study was to analyze the phenoliccomponent content of wheatgrass extract (Triticumaestivum L.) and to determine the effect of thisextract against DNA damage caused by cisplatin andetoposide used in chemotherapeutic treatment inhealthy cells will.Material and MethodSprouted wheat methanol extract was prepared andanalysis of phenolic components was performed byHPLC (high performance liquid chromatography).IC50 (half maximal inhibitory concentration) and LD50(median lethal dose) values were calculated by applyinga cell viability assay based on wheat extractconcentration. Cells were incubated at these determinedconcentration levels and the presence of DNAdamage was assessed by the Comet method.ResultsAs a result of the phenol component analysis, p-hydroxybenzoicacid was determined in the highestamount and o-coumaric acid in the lowest amount.The values for wheatgrass extract, etoposide andcisplatin at different concentrations applied to lymphocytecells were found to be IC50=204.6 μg/mL,LD50=15.84 μg/mL and LD50=24.51 μg/mL, respectively.As a result of comet analysis, it was foundthat etoposide LD50 and etoposide LD50+wheatgrassextract IC50 group were statistically significant(p0.05). Similar to this result, cisplatin LD50 andcisplatin LD50+wheatgrass extract IC50 group werefound to be statistically significant compared to thecontrol group (p 0.05).ConclusionAlthough wheatgrass was observed in our study tocause a reduction in DNA damage caused by etoposideand cisplatin, no statistical significance wasfound.

Therefore, in this study,we compared the radiologic, and clinicopathologicalfeatures of breast cancer patient’s < 40 age and ≥55age.Material and MethodThe files of a total of 759 patients, including 92patients under 40 aged, and 322 patients 55 aged andover who were operated on for breast cancer over a10-year period in three centres were retrospectivelyreviewed and Estrojen Reseptor (ER), ProgesteronReseptor (PR), Human epidermal growth factorreceptor 2 (HER2), Lymphovascular invasion (LVI)status, presence of axillary lymph node metastasis(ALNM), multifocality, presence of Ductal Carsinomaİnsitu (DCIS) or Lobular Carsinoma İnsitu (LCIS),tumor size, tumor histopathological type, grade, andscore were recorded.ResultsIn patients under the age of 40, the tumor is lesslocalized in the upper-inner and lower-inner quadrantsof the breast, multifocality is more common, most ofthem have dense breast structure, the histologicalgrade of the tumor is higher, LVI and LNM are morecommon.   • DETERMINATION OF FACTORS ASSOCIATED WITH FEAR OF HYPOGLYCEMIA AND
           PERCEPTION OF SELF-EFFICACY IN DIABETES PATIENTS USING INSULIN

      Abstract: ObjectiveThe aim of this study is to determine the factors associatedwith fear of hypoglycemia and perception ofself-efficacy and related factors in diabetic patientsusing insulin.Material and MethodThis descriptive and correlational study was conductedwith a total of 136 patients followed by a diabeteseducation nurse in a State Hospital betweenDecember 2016 and February 2017. "Patient InformationForm", "Self-Efficacy Perception Sub-Scale ofMultidimensional Diabetes Questionnaire" and "HypoglycemiaFear Scale (HFS)" were used to collectdata. In order to determine whether the data showeda normal distribution, Kolmogorov-Smirnov test, kurtosisand skewness values were calculated and thetests to be used for the analysis of the data were determined.The t test was used to compare the databetween two independent groups, and the one-wayanalysis of variance was used to compare the databetween more than two groups. Pearson Correlationanalysis was performed to determine the relationshipbetween scale scores. Multiple Linear Regressionanalysis was performed to determine whetherthe independent variables significantly predicted theSelf-Efficacy Perception Scale and HFS scores.ResultsThe mean age of the individuals participating in theresearch is 58.12 ± 12.4. In addition, 66.9% of theparticipants are women and 75% of them are fiftyyears old and over. The total mean score of theparticipants on the HFS was 52.5 ± 11.2; Behaviorsub-dimension mean score was 28.12 ± 4.76 andWorry sub-dimension mean score was 24.46 ± 8.11.The mean score of the Self-Efficacy Perception Scaleof the patients was 364.2 ± 18.46. It was determinedthat there was a statistically significant difference betweenthe patients' DM duration, DM type, DM drugtreatment type, regular follow-up visits, hypoglycemiaexperience and knowing the symptoms of hypoglycemia,and the scores of HFS and Self-Efficacy PerceptionScale (p < 0.05).   • IN SILICO ANALYSIS OF THE POTENTIAL ROLE OF SRY GENE IN PARKINSON’S
           DISEASE

      Abstract: ObjectiveParkinson’s disease (PD) is the most common movementdisorder and is caused by the loss of dopaminergicneurons in the substantia nigra. The firstmotor symptoms of PD begin to appear when morethan 60% of dopaminergic neurons are already lost.Although the etiology of PD is not fully understood,PD is believed to result from the complex interactionsbetween genetic and environmental factors. Previousstudies demonstrated that men are at least 1.5 timesmore likely to develop PD than women. The currentkulstudyaimed to investigate whether SRY, a chromosomegene, plays a role in the pathogenesis of PDusing in silico methods.Material and MethodThe mRNA expression of SRY in different human tissueswas analyzed using the Human Protein Atlas(HPA). The interactions between SRY and PD-relatedgenes were analyzed using the STRING andGeneMANIA databases. The JASPAR database wasused to determine the putative binding sites of SRY inthe promoter regions of some PD-related genes. Finally,the BioGRID and GeneMANIA databases wereused to identify the key genes that interact with SRY.ResultsIn silico analyses revealed that there are SRY bindingsites in the promoter regions of some PD-associatedgenes and that SRY can regulate androgen receptor(AR) signaling.ConclusionThese findings suggest that SRY may play a role inthe pathogenesis of PD and may thus serve as a moleculartarget for developing new therapeutic strategiesagainst PD.

Sele-nium (SEL) is a trace element that takes part in somechemical and enzymatic reactions in both animalsand human organisms and can cause some disordersin many systems such as the cardiovascularsystem in case of deficiency. The aim of this study isto investigate whether SEL has a protective effect oncardiac tissue in experimentally induced ACR toxicityin rats.Material and MethodA total of 28 rats were randomly and equally dividedinto four groups: Control, SEL, ACR, SEL + ACR. Atthe end of the study, cardiac markers such as creatinekinase MB (CKMB), lactate dehydrogenase (LDH)and ischemia-modified albumin (IMA) from bloodsamples were measured. Oxidative stress markerssuch as total oxidant status (TOS), total antioxidantstatus (TAS) and oxidative stress index (OSI) in hearttissue were analyzed. In addition, inducible nitric oxidesynthase iNOS activities were determined in heartsections using immunohistochemical methods.ResultsThe ACR + SEL group showed a significant decreasein TOS, OSI, CKMB, LDH, IMA levels and significantincrease in TAS level compared to the ACR group.In histopathological and immunohistochemical evaluationof the heart sections; Disorganization in cardiacmuscle fibers, increased connective tissue betweenmuscle fibers and irregular eosinophilia in musclefibers were detected in ACR group. A decrease inhistopathological findings was observed in the SELtreated groups compared to the ACR group. iNOSimmunoreactivities; moderate marking was observedin the ACR group sections, while poor marking wasobserved in the ACR + SEL group.ConclusionWe determined that ACR caused damage to the hearttissue of rats through various metabolic pathways andthat these damages were reversed in the groups giventhe SEL.

  • ANTIPROLIFERATIVE EFFECTS OF VITAMIN K2 IN OSTEOSARCOMA CELLS: COMPARISON
           OF DIFFERENT CYTOTOXICITY ANALYZES

      Abstract: ObjectiveThe aim of this study was to compare four commonlyused cytotoxicity assays (XTT, neutral red uptake, crys-tal violet assay, and propidium iodide staining) whichanalyzed the antiproliferative effects of vitamin K2.Material and MethodSaos-2 cells, an osteosarcoma cell line, wereexposed to vitamin K2 at different concentrations (10μM, 20 μM, 30 μM, 40 μM, 50 μM, 100 μM and 200μM) for 48 and 72 hours. Tetrazolium salt test (XTT),neutral red uptake (NR) assay, crystal violet assay,and propidium iodide (PI) staining were performed todetermine cytotoxic potential of vitamin K2 in termsof the cell viability and IC50 values. The results wereevaluated with one-way analysis of variance (ANOVA)and the Tukey test.ResultsCytotoxic effects of vitamin K2 on osteosarcoma cellswere analyzed with XTT, neutral red, crystal violetassay, and propidium iodide, respectively. IC50 valueswere determined exposure to 61.93; 40.21; 62.11;70.57 μM vitamin K2 for 48 and 75.44; 68.22; 41.66;88.01 μM vitamin K2 for 72 hours.ConclusionStatistical analysis revealed that there is a significantstatistical difference between four tests used in thisstudy. In addition, it was determined that the viabilityrates in propidium iodide staining were higher thanother tests for cytotoxicity analyses. It has beenconcluded that incubations at different concentrationswere required to prevent misinterpretation of results incytotoxicity analyses, and tetrazolium salt-based testsshould be supplemented with other non-metabolictests.

  • THE PREVALANCE OF IRON DEFICIENCY ANEMIA IN ADULTS IN HOSPITAL OF ISPARTA
           SULEYMAN DEMİREL UNIVERSITY AND EVALUATION OF ETIOLOGICAL FACTORS

      Abstract: ObjectiveIron deficiency anemia (IDA) is one of the most commondiseases in our country and effecting lives ofmillions of people all over the world. There are multifoldreasons for its etiology. This is a retrospectivecross-sectional study to address the prevalence andetiology of IDA among patients in the SDU ResearchHospital, Isparta, Turkey.Materials and MethodsA total of 1000 patients ages between 18-65 whowere admitted to the Outpatient Clinic of Departmentof Internal Medicine, SDU School of Medicine, betweenApril 2017 and November 2017. Complete bloodcount including ferritin, vitamin B12 and level of folicacid values were determined. Gender, age, menopausalstatus, additional diseases and etiologic factors ofthe patients were recorded. Hemoglobin, hematocrit,MCV, RDW, leukocyte, platelet, ferritin, vitamin B12and folic acid levels were also documented. Hemoglobinlevels were calculated as below 12 g/dl and 13g/dl in females and males, respectively. Ferritin lessthan 15 ng/ml (patients with rheumatic diseases lessthan 100 ng / ml) were accepted as IDA.To examinethe etiologic factors in patients with IDA; dietaryiron intake, menstrual bleeding in women, hemorrhoid,peptic ulcer, gastric surgery, gastritis, malignancywere investigated. Additional diseases identified atadmission were chronic liver/kidney/lung/rheumatoid/endocrinological, hypertension, diabetes mellitus,malignancies, hemoglobinopathies and inflammatorybowel disease.ResultsIn our study, 25 % of the women and 7,4 % of menhad IDA, and we found that the prevalence of IDA was20,3 % in all patients. The most common cause ofIDA was menorrhagia in women and gastritis in men.However, we could not fully assess the etiologic factorin 55 % of men and 34,4 % of women with IDA.DiscussionProspective, community based studies that also evaluatepika and parasitosis should be performed in patientswith iron deficiency anemia.
