  • Evolution, Function, and Deconstructing Histories: A New Generation of
           Anthropological Genetics
    • Abstract: It has been an amazing decade for anthropological genetics. Our questions have remained the same—profound inquiries about human nature and diversity—but thanks to an array of new technologies, our answers become infinitely more insightful and accurate. A click away, we now have access to thousands of whole modern human genomes and hundreds of ancient individuals with genome-wide data. Our comparisons of these genomes become even more meaningful because we now know much more about what a variant at a given location of the genome may do to the expression of genes, functioning of proteins, cellular phenotypes, or processes in our organs. These new data sets build on existing concepts. A DNA sequence is still a DNA ... Read More

  • The Genomic Health of Ancient Hominins
    • Abstract: Ancient human remains yield valuable information about the health and disease of individuals from the past. Skeletal remains provide details about medical practices (Andrushko and Verano 2008) and certain diseases, such as cancers (Binder et al. 2014; Odes et al. 2016) and rheumatic diseases (Entezami et al. 2011), that visibly alter or affect bone. However, many diseases do not leave their mark on bone; instead, they cause soft tissue damage, requiring mummified remains for diagnosis. One of the best-studied sets of mummified human remains is Ötzi, the Tyrolean Iceman, a well-preserved 5,300-year-old Neolithic man discovered in the Ötztal Alps. Examination of his remains shows hardening of the arteries, suggesting ... Read More

  • Admixture and Ancestry Inference from Ancient and Modern Samples through
           Measures of Population Genetic Drift
    • Abstract: The wealth of genome-wide polymorphism data from diverse human populations around the world (Cann et al. 2002; Gibbs et al. 2003; Altshuler et al. 2010; McVean et al. 2012) has allowed researchers to access a record of human history unprecedented in its breadth, spanning thousands of years (Patterson et al. 2012; Jones et al. 2015; Mendez et al. 2015; Lazaridis et al. 2016). With these data, new insights into human migration (Rasmussen et al. 2014; Lazaridis et al. 2014; Skoglund and Reich 2016) and interbreeding (Hellenthal et al. 2014) during the peopling of the world have emerged. Specifically, the recent publication of genomes from ancient human remains (Rasmussen et al. 2010; Olalde et al. 2014; Fu et al. ... Read More

  • Infectious Disease and the Diversification of the Human Genome
    • Abstract: The human immune system is a complex and highly physiologically infiltrative organization of proteins, cells, tissues, and organs that serves as the main interface between ourselves and the outside world. As such, it is under tremendous selective pressure mediated by pathogens and other microorganisms (Barreiro and Quintana-Murci 2010; Filip and Mundy 2004; Fumagalli et al. 2011; Nakajima et al. 2008; Nedelec et al. 2016; Novembre and Han 2012; Sawyer et al. 2005). Genome-wide scans for evidence of natural selection in humans and other species have repeatedly found that immune genes are overrepresented in groups of genes associated with signatures of selection (Andersen et al. 2012; Cagan et al. 2016; Kosiol et al. ... • Introgression Makes Waves in Inferred Histories of Effective Population
           Size
    • Abstract: The origins of today’s modern human populations included introgression or gene flow from genetically divergent ancestral populations of archaic humans. Direct evidence of this ancient introgression comes from the ancient DNA of Neandertals, which contributed between 1% and 4% of the genetic makeup of today’s populations outside Africa (Green et al. 2010; Prüfer et al. 2014), and from the high-coverage Denisovan genome, which represents an archaic population that contributed up to 5% of the genetic makeup of today’s Melanesian and Australian populations (Reich et al. 2010, 2011; Meyer et al. 2012; Prüfer et al. 2014), as well as a much smaller fraction of introgression within mainland Asian and American populations ... • Leveraging Multiple Populations across Time Helps Define Accurate Models
of Human Evolution: A Reanalysis of the Lactase Persistence Adaptation
    • Abstract: Understanding the evolutionary forces that human populations have been subject to in the past has been a central issue in evolutionary biology. In particular, determining how two principal mechanisms, genetic drift and natural selection, have shaped genetic variation remains a highly studied area of research. One of the difficulties in addressing this issue is that there are many forms of natural selection, and sometimes various types of selection lead to similar genetic patterns. For example, dips in genetic diversity along the genome have been associated with the effects of both negative and positive selection or with a neutral scenario in a region with low mutation rate (Otto 2000). Furthermore, positive ... Read More
