  • Survey of Antimicrobial Residue in Table Eggs among Layer Poultry Farmers
           in Maiduguri Metropolis, Borno State

    • Abstract: Objective: The objectives of this study were to survey for antimicrobial usage and to assess the antibiotic used pattern among poultry layer farmers in Maiduguri Metropolis Borno state Nigeria. Methodology: Multistage sampling technique was used together with structured interviewer questionnaire. Coordinates of each farm selected was recorded and map showing spatial distribution of randomly selected farm was constructed. Data were analyzed using Statistical Package for social science. Results: The result indicated that the majority of the farmers 30 (85.7%) admitted the use of antibiotic for disease prevention, 29 (82.9%) of the farmers were not aware of antibiotic residue in table eggs and 34 (97.1%) of the farmers were not aware of public health effect of antibiotic residue. Conclusion: It was deduced that there is indiscriminate use of most antibiotics in layer farms in Maiduguri Metropolis, There is poor knowledge of drug withdrawal period by farmers and poor awareness on passage of antibiotics from hen to eggs and its public health significance. Prohibited antibiotics for use in food producing animals such as nitrofurans, chloramphenicol and unapproved antibiotic for use in laying hens such as sulfonamide, doxycycline, streptomycin, gentamycin and ampicillin are very much in use in the study area.

  • Effects of Honey Supplementation into the Extender on the Motility,
           Abnormality and Viability of Frozen Thawed Of Bali Bull Spermatozoa

    • Abstract: Background and Objective: Honey is comprise high amount of variety of simple sugars which might serve nutrition to sperm cells. The objective of this study was to evaluate the effects of honey addition into the extender on the quality of frozen thawed in Bali bull spermatozoa. Materials and Methods: A total of four Bali cattle bulls were used in this study. Honey solution was added at the concentration of 0.1, 0.2, 0.3 and 0.4% to bovine semen cryoprotective medium. The cryoprotective extender (skim milk-egg yolk) for the control group was the same as that for the treatment groups except that it was not supplemented with honey solution. Sperm parameters were assessed including motility, abnormality and viability. The data were statistically analyzed pre and post-thawing. Results: The results indicated that percentage of the sperm motility before freezing was significantly lower (p>0.05) among control and treatment groups. Furthermore, the percentage of the abnormality and viability were no significantly different (p>0.05) among control and treatment groups. The sperm abnormality frozen thawed was significantly higher (p0.05) among control and treatment groups. Conclusion: It is concluded that honey supplementation into the extender was significantly effect on the sperm motility before freezing and sperm abnormality on the frozen thawed.

  • A Serological Survey of Four Abortifacient Infectious Agents among Small
           Ruminant in Egypt

    • Abstract: Background and Objective: Abortions in small ruminants constitute a key problem not only in Egypt but also in other European countries which have developed the livestock sector. Many pathogens causes abortion, sever economic losses and have ability to transmit to human. Materials and Methods: A serological investigation was performed to estimate the seroprevalence of four abortive infectious pathogens in small ruminants in five governorates of Egypt. A total of 284 serum samples were collected from sheep and goats and examined for Brucella sp., Chlamydophila abortus (C. abortus), Coxiella burnetii (C. burnetii) and Toxoplasma gondii (T. gondii). Results: The seroprevalence confirmed the presence of four investigated pathogens with non-significant difference (p>0.05) among sheep and goats in different localities. The most prevalence pathogen was Brucella sp. 27 (16.8%), 29 (23.6%) followed by C. abortus 24 (14.9%), 17 (13.8%) and C. burnetii 25 (15.5%), 16 (13%) in sheep and goats, respectively. Toxoplasma gondii showed the low detection rate in sheep 10 (6.2%) and goats 8 (6.5%). The results revealed that the single infection with Brucella sp., C. abortus and C. burnetii or mixed infection with Brucella sp. and C. burnetii had a highly significant effect on the occurrence of abortion in small ruminants. Conclusion: The seroprevalence of Brucella sp. is high and other pathogens (C. abortus and C. burnetii) can be found and should be diagnosed to develop an applicable control program.

  • Parasitological and Molecular Based Detection of Cerebral Babesiosis in
           Kankrej Bullock and its Successful Therapeutic Management

    • Abstract: Background and Objective: Bovine babesiosis is an economically important tick borne disease of tropical and subtropical parts of the world including India. The disease accounts for significant morbidity and mortality in cattle and buffaloes. The current study investigates the rare occurrence of cerebral form babesiosis in a Kankrej bullock by microscopy and polymerase chain reaction (PCR) based assay and its successful therapeutic management. Materials and Methods: A 4-years-old Kankrej bullock was brought to Teaching Veterinary Clinical Complex (TVCC), College of Veterinary Science and A.H., Junagadh, Gujarat with symptoms of high temperature (108°F), dullness, dyspnoea, pale mucous membrane, icterus, anaemia, hyperexcitability and convulsions. Blood sample was collected and subjected to various haematological parameters estimation, microscopic examination and PCR for detection of haemoparasites. Results: Optical microscopy based thin blood smear revealed the presence of Babesia piroplasm in the circulating erythrocytes. It was further confirmed by PCR that amplified an approximately 278 bp specific for Babesia bigemina (B. bigemina). The haemogram revealed suppressed haematological indices. The clinical and haematological findings coupled with detection of piroplasm by conventional and molecular techniques in circulating erythrocytes made us infer that the bullock was suffering from cerebral form of babesiosis. The bullock was treated with Diminazene aceturate along with the supportive therapy. The bullock promptly responded to therapy within 72 h and effectively restored the normal haematological indices and erythrocytes were found to be free from the piroplasms. Conclusion: The current study highlights the importance of microscopy complemented with PCR for accurate detection and could be used for diagnosis of latent infection in carrier animals. This seems to be the first report of cerebral form of babesiosis in a Kankrej bullock and placed on the record.

  • Effect of Fermentation Using Different Microorganisms on Nutritive Values
           of Fresh and Dry Cassava Root

    • Abstract: Background and Objective: Cassava (Manihot esculenta Crantz) is widely grown in sub-tropical and tropical areas, producing roots as an energy source containing high soluble carbohydrate but low in crude protein. The process of protein enrichment of animal feed using microorganisms in a semi-solid culture to improve the nutritional value of ruminants feed has been considered. This study aimed to investigate the effect of microorganism fermentation on nutritional values of cassava products and in vitro rumen fermentation and digestibility. Materials and Methods: The experimental design was a 2×4 factorial arrangement in a completely randomized design. Factor A was two types of cassava root (fresh cassava root (FC) and cassava chip (CC)) and factor B was four sources of microorganism inclusion [no microorganism (No), Yeast (Y), effective microorganism (EM) and Yeast+EM (EMY)), respectively.   • Productive Performance and Histological Evaluation of Delayed Post Hatch
           Feed Access Broilers Fed Threonine Supplemented Diet

    • Abstract: Background and Objective: Chicks may expose to delayed post hatch feed access due to several causes which may have consequences for poultry production. The present study investigated the impacts of different delayed time's access of feed on the production performance, histological examination and carcass traits in broiler chicks and how to substitute the detrimental effect of this delay by supplementation of threonine. Methodology: A total of 300 one-day old broiler chicks "Cubb 500" were used. They were alienated into 5 experimental groups, each group was further alienated into 6 replicates of 10 birds. Several productive traits as body weight gain, feed intake, feed efficiency, mortality and carcass traits were measured. In addition, villus height, villus width, crypt depth and villus surface area were also measured. Results: Results showed that group IV which fastened 24 h after hatch and fed threonine supplemented diet showed the highest performance traits. Control group showed the least yolk residual and the highest feed intake. Group of bird fastened either 24 or 48 h showed the least weight gain compared to other groups. Regarding to carcass traits, group of bird fastened for 48 h showed the least giblets weight and dressing percentage, while group IV showed the highest values of carcass traits. Conclusion: The productive performance of chicks was improved following early access to diet after hatch through skip all obstacles that prevent early chicks feed access.

  • Quality Indicator Hypoxanthine Compared with Other Volatile Amine
           Indicators of Sea Foods Stored in Refrigerator

    • Abstract: Background and Objective: Study evaluates the suitable chemical method for the determination of the freshness of refrigerated seafood. Hypoxanthine content (Hx) of the refrigerated seafood was analysed and it was compared with the level of volatile amines production due to bacterial spoilage. Materials and Methods: Fresh seafood such as Stolephorus commersonii, Scomberomorus koreanus and Portunus sanguinolentus were collected from the fishing harbour of Tuticorin. The fishes were analyzed initially for TMA-N, TVB-N, hypoxanthine and pH content and it was stored at -4°C for 30 days. Sampling was done for every 10 days interval and it was checked for all the above quality indicators. Results: The results showed that volatile amines (TVB-N) and (TMA-N) were not found in stored seafood till the end of 10 days storage. Changes in pH and hypoxanthine content were noticed at the initial storage period itself. Refrigeration of sea foods are common at homes and at this condition even the superior quality stored fish has hypoxanthine formation and it gradually increase during the storage period. Hx assay reflects ATP break down and ATP-related compounds and the degradation of ATP to Hx has been attributed to muscle endogenous autolytic enzymes. The enzymatic reaction occurs in the initial stage of storage at low temperature but the contents of trimethylamine-nitrogen and total volatile base-nitrogen increase progressively during the later period of spoilage. Thus, hypoxanthine is regarded as the major catabolite of adenosine triphosphate while it was formed in refrigerated situation and it is a valuable freshness marker in stored fish. Conclusion: Thus, the study reveals that Hx measurements have some advantage over TMA-N and TVB-N analyses and Hx could be used as a superior spoilage indicator in the refrigerated sea foods. This study proves that the Hx could be used as a spoilage indicator and the sea foods stored at -4°C, should be consumed within 10 days of storage period to avoid Hx accumulation.

  • PCR-RFLP and Sequence Analysis of Hair Cytochrome b Gene for
           Identification of Animal Species

    • Abstract: Background and Objective: Examination of animal hair by conventional methods could not reach a conclusive parameter that can differentiate between animal species. In this study, PCR- RFLP and sequencing of cytochrome b gene were used to differentiate between DNA samples extracted from human and animal hair. Materials and Methods: Hair samples were collected from human, cattle, sheep, dog, cat and mice, DNA was extracted from 5 samples of each species and subjected to PCR amplification of a mitochondrial cytochrome b (cyt b) gene segment (358 bp) using universal primers (forward primer: L14816: 5'-CCA TCC AAC ATC TCA GCA TGA TGA AA-3' and reverse primer: H15173: 5'-CCC CTC AGA ATG ATA TTT GTC CTC A-3'). Subsequent digestion with 2 restriction endonuclease (HaeIII and HinfI) and sequencing of pure amplified DNA and matching with data base of GenBank. Results: Digestion of PCR products with HaeIII and HinfI restriction endonucleases showed species-specific pattern on agarose gel. HaeIII could differentiate between all the studied species except human and mice, which showed the same pattern of bands in the form of three cleavage sites while HinfI produced negative reaction with human hair and cut the mice hair at three sites. HinfI endonuclease produced different pattern of bands in all studied species except cattle and dog showed similar fragments. Sequence analysis gave 93-100% identity between laboratory results and GenBank data. Conclusion: The results suggest that the method of PCR-RFLP using HaeIII and HinfI restriction enzymes can reliably identify the studied species. Furthermore, sequence analysis of cyt b is very sensitive and powerful for animal species identification.

  • Impact of Dietary Supplementation of Whole Flaxseed and Flaxseed Meal to
           Infected Broiler Chickens with Eimeria tenella

    • Abstract: Background and Objective: Coccidiosis is the major parasitic disease affecting poultry industry. Drugs and live vaccines are the two main control measures for the disease. However, due to the prolonged drug usage and the high cost of vaccines, alternative strategies are needed for more effective and safer control of coccidiosis in chickens. This experiment was conducted to determine the effects of supplementation broiler diets with source of n-3 fatty acids as 10% grinding whole flaxseed (WF) or flaxseed meal (FM) during coccidian infection. Materials and Methods: A total of 121 days old Cobb chicks were randomly allocated to 4 treatments, 3 replicates containing 10 chicks in each replicate. The chickens in groups 1 and 2 were fed corn-soybean based diet (negative and positive control), chickens in group 3 were fed control diet supplemented with 10% WF and chickens in group 4 were fed 10% FM. Diets were fed from 8 days of age throughout 33 days of age. Chickens were inoculated by gavage with 40,000 sporulated oocysts of Eimeria tenella (E. tenella) at 21th day of age. Growth performance, oocysts count, cytokines and cecal histopathological changes were determined at the days 28 and 33 of age.   • Performance and Quality in Old Laying Hens Fed Dietary Silicon Extracted
           from Rice Hull Ash

    • Abstract: Background and Objective: Normally, old laying hens must be provided with proper diet supplementation to maintain their optimum egg production and egg quality. As a silicon is an essential trace mineral which plays an important role in normal bone and associates with calcium in the early stages of biocalcification. Therefore, the current study was conducted to investigate the effects of dietary silicon extracted from rice hull ash (SERA) on the egg performance and egg quality of old laying hens. Materials and Methods: At 75 weeks of age, a total of 80 laying hens (Lohmann Brown Classic) were divided into 4 dietary groups of 20 birds each as follows: Each group was fed the basal diet (CP, 18%; ME, 2,850 kcal kg–1) supplemented with 0.00, 0.50, 0.75 or 1.00% SERA, respectively.   • Assessment of Modified Rice Husk and Sawdust as Bio-adsorbent for Heavy
           Metals Removal Using Nano Particles in Fish Farm

    • Abstract: Background and Objective: Improving water quality in fish farms is essential key factor for maintaining the fish health and its productivity. This study was designed to assess the quality of pond’s water and the effectiveness of bio-adsorbent materials (sawdust and rice husk) and modify these materials using silver nanoparticles (AgNPs) to enhance the adsorption capacity for removal of lead, cadmium and improving physicochemical parameters in different water sources used in fish farm. Materials and Methods: A total number of 150 representative water samples were collected from five pond’s water to assess hygienic water quality. The physicochemical parameters [pH, alkalinity (mg L–1), electrical conductivity (μS cm–1), ammonia (NH3) (mg L–1), nitrite (NO2) (mg L–1), nitrates (NO3) (mg L–1) and total hardness (mg L–1)] were determined before and after using bio-adsorbent materials and some heavy metals (Pb and Cd) in pond’s water were determined using atomic absorption spectrometer.   • Effect of Additives on the Forage Quality of Pelleted Hazelnut Husks

    • Abstract: Background and Objective: Harvest wastes and some forages are pelleted in order to be used in ruminant nutrition to improve forage quality. This study was conducted to determine nutrient contents and in vitro true digestibilities (IVTD) of hazelnut husks (HH) pelleted with the addition of molasses, urea+molasses, corn and sepiolite. Methodology: In the study, there were total 8 treatment groups (4 sepiolite added groups and 4 non- sepiolite groups for HH). Daisy incubatorD220 was used to determine the IVTDs of the feeds. The data obtained from the experiments were analysed in accordance with the completely randomized design. Results: The crude protein (CP) contents of HH were 8.79-16.89%. Sepiolite addition did not affect the CP content of HH. The HH had highest IVTD value. The sepiolite addition decreased the IVTD value of HH. All of the feeds were listed in the lowest quality class. But the addition of urea and molasses to the ration was found to have a significant effect on the CP content of HH. The highest IVTD values were found from the addition of corn to rations of HH groups. Conclusion: The addition of sepiolite and corn must be given special attention in pelleting process of hazelnut post-harvest wastes.

  • Live Bacteria in Clots from Bovine Clinical Mastitis Milk with No Growth
           in Conventional Culturing

    • Abstract: Background and Objective: Although screening of clinical mastitic milk for bacteria is necessary for evidence-based antibiotic treatment and prevention of mastitis, these milk samples are frequently culture negative. The purpose of the present study was to evaluate clots from culture-negative mastitic milk for the presence of viable bacteria in an attempt to unravel the reason for the no-growth (NG) status of mastitic milk. Methodology: A total of 449 clinical mastitic milk samples were enrolled, visual inspections were performed to confirm the presence of clots in milk samples and specimens were subjected to bacterial culturing by conventional methods. Milk samples that were NG by primary culture were re-cultured. Furthermore, clots in NG milk samples were recovered aseptically and cultured. Bacteria isolated from milk or clots were identified using biological methods. Results: In primary culture, NG was detected in 96 out of the 449 samples tested (21.4%). The proportion of mastitic milk with clots was significantly higher in NG milk (45.1%, 78/173) than in growth (G) milk (6.5%, 18/276). In the secondary culture of NG milk and its clots, the culture-positive rate of clots from NG milk (53.8%, 42/78) was significantly higher than that from NG milk (34.4%, 33/96). Although no differences were found in other pathogens, the isolation rate of coliforms from clots (20.8%, 20/78) was more than two times that from milk (8.3%, 8/96). Conclusion: These findings suggested that elimination of mastitic bacteria by the host immune system may be a cause of culture-negative mastitic milk and that this pattern is often observed upon infection with coliforms.
