  • Therapeutics effects of aqueous extract Cynodon Dactylon on the mice model
           ulcerative colitis

      Abstract: Background and Aim: Cynodon dactylon (C. dactylon) has long been examined for its anti-inflammatory properties. A type of Inflammatory Bowel Disease (IBD) is Ulcerative Colitis (UC). This study investigated the impacts of C. dactylon on the inflammatory mediators of UC induced by acetic acid in Wistar rats.Materials & Methods: The experiment was conducted on 50 male Wistar rats grouped into five equal individuals. Excluding Group I (negative control), acetic acid was used to induce UC in the rats. The treatments received different doses of C. dactylon extract, namely Group II (1 mL of 200 mg/kg orally per day, Group III (1 mL of 200 mg/kg as intra-colonic), Group IV (1 mL of intra-colonic 10 mg/kg Mesalazine), and Group V (1 mL of normal saline) as positive control. The whole treatments lasted for 10 successive days. At completion of the study, intestinal tissue and blood samples were taken from sacrificed rats. Measured parameters were inflammatory mediators, myeloperoxidase (MPO), superoxide dismutase (SOD), glutathione peroxidase (GPx), and activities of cytokines TNF-α and IL-6.Results: A more remedial advantage of C. dactylon extract in two forms of trans-rectal and oral administration was observed on damaged colonic tissue induced by acetic acid with reduced activities of MPO, SOD and GPx. Amounts of TNF-α and IL-6All were lowered by all treatments. Mesalazine and C. dactylon extract were not significantly different in terms of gross damages. Conclusions: Our observations indicate that C. dactylon extract with anti-colitis property can be regarded as an appropriate candidate and a natural source of current medications.

  • Effects of Ginger supplementation on lipid profile: A systematic review
           and meta-analysis of randomized clinical trials

      Abstract: AbstractBackground: Some, but not all, experimental and clinical studies have suggested that ginger supplementation has beneficial effects on serum lipid profiles. Therefore, we conducted a meta-analysis to evaluate the ginger supplementation on human serum lipids.Methods: Systematic search for English published randomized controlled trials (RCTs) was performed up to April 2016 through electronic databases: Medline, EMBASE and Google Scholar. A total of seven RCTs were met inclusion criteria. The pooled weighted mean difference (MD) and its 95% confidence interval (CI) were calculated and pooled using using a random-effects model.Results: Ginger supplementation was associated with a significant reduction in mean serum total cholesterol (-13.31 mg/dL, 95% CI, -20.29 to -6.33 mg/dL, P=0.000), low-density lipoprotein cholesterol (-11.22 mg/dL; 95% CI, -18.37 to -4.06 mg/dL, p = 0.002), triglycerides ( -14.96 mg/dL; 95% CI, -22.13 to -7.79 mg/dL, p = 0.000); Increasing in high-density lipoprotein cholesterol level was not statistically significant (WMD = 0.12 mg/dL; 95% CI, -1.01 to 1.24 mg/dL, p =0.839.Conclusion: Ginger supplementation significantly improved lipid profile (LDL, TG and TC) and it may be an effective intervention for managing serum lipid status. Further, high-quality RCTs are required to identifying the ginger supplementation effects on HDL.

  • Effects of Hibiscus Sabdariffa aqueous extract on spermatogenesis and
           sperm parameters of mice

      Abstract: Background and Aim: Some medicinal plants are conventionally used for treatment of different aspects of male infertility. This study was conducted to evaluate the effects of Hibiscus Sabdariffa aqueous extract on spermatogenesis and sperm parameters of mice.materials and methods: Forty-eight were divided into 4 groups with 3 replications and 3 animals in per replication and received doses of 0, 100, 200 and 300 mg/kg of Hibiscus Sabdariffa aqueous extract for 28 days. Levels of testosterone, LH, FSH, TAS, SOD, GPx, blood variables, testis weight, sperm abnormality and viability, motility and sperm concentration were evaluated.Results: Oral administration of Hibiscus Sabdariffa aqueous extract lowered levels of FSH and LH in comparison to control group (P<0.05). The level of TAS was significantly lower in level of 300 mg/kg of the extract in comparison to control group (P<0.05).  The levels of SOD and MDA were significantly higher and lower in comparison to control group, respectively. The levels of triglycerides and cholesterol were significantly lower in control group in comparison to other groups (P<0.05). Left and right weights were significantly lower in 100 and 200 mg/kg extract groups in comparison to 300 mg/kg extract group (P<0.05). Normal sperm count was significantly higher in mice treated with 100 mg/kg of the extract. Defected sperms were significantly lower in mice treated with extract.Conclusion: It cannot be stated to use of clear dose of Hibiscus Sabdariffa aqueous extract in order to improve spermatogenesis and sperm parameters in mice.

  • Phenolic Composition, in vitro Antioxidant and Anticancer Activities of
           Hypericum Japonicum Thunb and Scoparia Dulcis L

      Abstract: Background and Aim: Hypericum Japonicum Thunb (HT) and Scoparia Dulcis L (SL) have received considerable attention as natural products for their applications with health benefits. This study aimed at investigating the in vitro antioxidant and HepG2 (human liver cancer cell line) and A549 (adenocarcinomic human alveolar basal epithelial cell line) growth inhibitory effect of the ethanol extracts from the HT and SL.Materials and Methods: Total phenolic and flavonoid contents were identified. Antioxidant activity was determined by measuring hydroxyl radical scavenging activity, 2,2’-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS+) and 2,2’-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities. Phenolic compounds were identified by high performance liquid chromatography (HPLC) method. The activity of the apoptotic factors Bcl-2, Bad, Bax, Caspase-3 and Caspase-9 were measured by Western blotting.Results: The chlorogenic acid and rosmarinic acid were the compounds that had been identified in the HT. The rutin and rosmarinic acid were newly identified compounds from the SL. Moreover, it was found that the higher levels of ABTS+, DPPH and hydroxyl radical scavenging activities were detected from the HT extract with IC50 of 45.13±0.89, 89.29±0.78 and 38.32±0.17µg/mL, respectively. The HT and SL extracts could induce HepG2 growth inhibition with IC50: respectively 245.755 ± 2.23b µg/mL and 553.32 ± 14.46a µg/mL and also induce A549 growth inhibition with IC50 of the HT and SL: 314.15± 5.96b µg/mL and 446.35± 0.9a µg/mL at 48 hr, respectively. The HT and SL extracts stimulated the apoptotic factors including bcl-2, bad, caspase-3, caspase-9 in HepG2; p53, caspase-3, bax and bad in A549.Conclusion: The findings strongly suggest that the HT and SL extracts could be an excellent sources of antioxidants as functional food ingredients in the prevention and treatment of cancer.

  • The Determination of Flavonoid Components, Total Phenolic Content, and
           Antioxidant Capacity in Dog Rose (Rosa canina L.) in Lorestan Province

      Abstract: Background and Aim: Dog rose (Rosa canina L.) is a valuable deciduous shrub, which has long been used for food and medicinal purposes in various parts of the world. This study was laid out for the quantitative determination of the flavonoid components such as kampferol and quercetin, total phenol content and antioxidant capacity in petals and the fruit of the dog rose (Rosa canina L.) ecotype collected in Lorestan province.Materials and Methods: Petals and fruit samples of wild dog rose were harvested in June and September 2014 respectively. The samples were kept in cold box during their transportation to laboratory. Subsequently, the pericarp and seeds were separated and dried at room temperature. The antioxidant capacities of petals and fruits were determined using DPPH method. The flavonoid components, including kampferol and quercetin, were analyzed by high performance liquid chromatography (HPLC). The total phenol and flavonoids were measured using a spectrophotometric device. The results indicate that the amount of total phenol in petals (1957 mg.ml-1) was higher than that of fruits (937 mg.ml-1). The amount of total flavonoid in dog rose petals and fruits were 776 and 450 mg.ml-1 respectively.Results: The antioxidant capacity of dog rose was measured and found to be 9.6% in fruits and 66.7% in petals. However, the fruits contained 141 µg.g-1 of kampferol and petals contained 108 µg.g-1. The measured quercetin in petals (127µg.g-1) was higher than that of fruits (66µg.g-1). In this study, a direct relation was observed between the total phenol and flavonoid contents with antioxidant capacity.Conclusion: The results of this research showed that there are high amounts of total phenol and flavonoid contents, antioxidant capacity and also kampferol and quercetin in petals and fruits of dog rose grown in Lorestan. These results indicated the significance of dog rose in dietary and medical purposes.

  • Antiplasmodial and Cytotoxic Properties of the Ethanolic Extract of the
           Leaves of Sarcocephalous Latifolius

      Abstract: Background and Aim: Sarcocephalus latifolius serves multiple purposes in the treatment of different ailments in Nigeria. In this study, antiplasmodial and cytological activities of the ethanolic extract of leaves of S. latifolius were assessed using albino mice and Allium cepa models.Materials and Methods: For the antiplasmodial test, a total of 25 mice were divided into five groups of five mice. The groups were administered plant extract (25, 50 and 100mg/kg), chloroquine (10mg/kg) and distilled water respectively. This test involved two phases. In the suppressive phase, the extract was orally administered for four days after inoculation, and blood smear was prepared on the fifth day.  In the curative phase, the mice were inoculated with parasites three days before the administration of the extract to ensure the full development of parasites, and the extract was administered for five days. Blood smears were prepared along the periods of administration and five days after the administration. In cytotoxic study, onion bulbs were exposed to 25, 50 and 100 mg/ ml concentrations of the extract for 24, 48 and 72 hours. Subsequently, the macroscopic and microscopic investigations were evaluated.Results: All doses exhibited significant antiplasmodial activity dose-dependently. Root length significantly decreased in all concentrations compared to control. After 72 hours, the percentage root length and root inhibition at all concentrations were decreased. S. latifolius, therefore, demonstrated significant antiplasmodial activities. Availability of metabolites, such as alkaloids and flavonoids, may elicit endoperoxidation thereby causing death of parasites.Conclusion: This finding supports the traditional use of S. latifolius against malaria. Cytoarchitecture revealed aberrations like sticky telophase, vagrant metaphase and telophase which may indicate the signs of toxicity that could cause cell death.

  • Use of Satureja khuzestanica Essential Oil (SKEO) in the Treatment of
           Diarrhea: Modes of Action on Intestinal Function in Animal Model

      Abstract: Background and Aim: Satureja khuzestanica is an annual herb that is used in Iranian traditional medicine to treat intestinal disorders such as abdominal pain and diarrhea. Satureja khuzestanica essential oil (SKEO) was tested to elucidate the mechanism(s) of its gastrointestinal activity in two animal models.Materials and Methods: The in vitro test of the effect of SKEO on the contractions induced by different spasmogens in isolated rat ileum preparations was carried out. The impacts of SKEO on intestinal transit, diarrhea, and intestinal secretion in mice were investigated with the charcoal meal, castor oil-induced diarrhea, and entropooling assays.Results: Pretreatment with SKEO at doses of (5-20 µg/mL) had a significant and concentration-dependent relaxant effect on contractions induced by acetylcholine, nicotine, histamine and serotonin in isolated rat ileum preparations. Moreover, it had an inhibitory effect on both K+- and Ca2+-induced contractions in the same tissue. At doses of 100 and 200 mg/kg, SKEO decreased intestinal transit, fecal weight and intestinal secretion.Conclusion: SKEO showed anti-diarrheal, anti-motility and anti-secretory activities that might prove beneficial in the treatment of gastrointestinal disorders.

  • The Antioxidant Properties and Chemical Composition of Stachys inflata
           Benth Leaves Grown Wild in Lorestan Province

      Abstract: Background and Aim: In recent years there has been an increasing interest in herbal medicines and natural antioxidant products to be used as medicines and food additives. The present research identified the various antioxidative activities of ethanol and methanol extracts, and chemical composition of Stachys inflate Benth leaves essential oil.Materials and Methods: This experimental study was carried out in 2015 in Lorestan University of Medical Sciences. Ethanol and methanol extracts of Stachys inflata Benth leaves were primarily prepared. The total antioxidant capacity of samples was assessed by phosphomolybdate method. Total phenol content and total flavonoid content were determined by Folin-Ciocalteu and Zhishen methods. The components of Stachys inflate Benth leaves essential oil were analyzed with gas chromatography/mass spectrometry (GC/MS).Results: It was demonstrated that the total antioxidant capacity of ethanol and methanol extracts of Stachys inflata Benth leaves were 4.30±0.17; 3.63±0.23 mg of ascorbic acid equivalents /g of extract. Moreover, the total phenol content ethanol and methanol extracts of Stachys inflata Benth leaves were 830.334±26.41; 384.29±13.37mg of gallic acid equivalents /g of extract, and flavonoid content ethanol and methanol extracts of Stachys inflata Benth leaves were 17.09±0.154; 31.18±1.03 mg of quercetin equivalents /g of extract. GC/MS data and retention indices of Stachys inflate Benth leaves essential oil samples were used to identify 54 constituents. These compounds make up a total of 48.87 percent essential oil. Phenol, 2-methyl-5(1-methyleth; Phenol, 5-methyl-2-(1methyleyth; 2-pentadecanone and hexadecanoic acid are the major compounds of Stachys inflate Benth leaves essential oil. This study indicated that Stachys inflate Benth leaves essential oil has remarkable antioxidant properties. Furthermore,  Stachys inflate Benth leaves essential oil was identified as an easily accessible source of natural antioxidants such as Phenol,2-methyl-5(1-methyleth; Phenol,5-methyl-2-(1methyleyth; 2-pentadecanone,6,10,14-trimet; Veridi florol; 2-cycohexen-1-on,2-methyl-5- and 1H-cycloprop[e]azulen-7-ol,dec. It may also be suitable to be used in food and pharmaceutical applications.Conclusion: This study demonstrated that Stachys inflata Benth leaves are easily accessible sources of natural antioxidants. Therefore, they may be suitable to be used in food and pharmaceutical applications.

           Six Iranian Pomegranates Cultivars Using a New Miniaturized Matrix
           Solid-Phase Dispersion Method

      Abstract: Background and Aim:The peels of six Iranian pomegranates (Punica granatum L.) cultivars, as a traditional medicine, were treated with a new miniaturized matrix solid-phase dispersion (MSPD) method for the HPLC determination of ellagic acid (EA).Materials and Methods:In the proposed method, only 10mg of the sample powder was ground and blended with an equal amount of C18 sorbent in an agate mortar. The use of the agate mortar with smooth surface facilitated the sample transfer into a cartridge and reduced the required amount of sample and sorbent. Micro volumes of dichloromethane, n-hexane and methanol were used as modifier, washing and elution solvents, respectively. The eluate was injected into an HPLC-UV system for the analysis.Results:Several factors such as the type and amount of dispersing sorbent, modifier, washing solvent and eluent were carefully studied and optimized. Six replicated analyses at the optimized conditions resulted in a recovery of 96.7% and a relative standard deviation of 5.87%. The proposed method was successfully applied to the extraction and determination of EA in the peels samples.Conclusion:According to the ultimate results, the MSPD method is an efficient technique for the quantitative extraction of EA from the peels of pomegranate. Malas cultivar has the highest amount (18.1 g kg-1) of ellagic acid content compared to the other studied pomegranate cultivars.
