  • IGS-Fingerprinting and Virulence Properties of Vibrio alginolyticus
           Isolated from Farmed Fishes in Saudi Coast of Arabian Gulf

    • Abstract: Background and Objective: Vibrio sp. are a crucial constituent of marine ecosystems worldwide. Vibrio alginolyticus is extremely significant and major pathogenic agent facing wild and farmed fishes. The main objective of this study was to record the molecular characterization (IGS-Typing pattern) by single PCR step and virulence genes in V. alginolyticus isolates. Materials and Methods: For this study 360 fishes were collected, including gilt head seabream (Sparus auratus), two bar seabream (Acanthopagrus bifasciatus), black bream (Acanthopagrus latus), red sea seabream (Diplodus noct), brown-spotted grouper (Epinephilus coioides) and rabbitfish (Siganus canaliculatus) between July, 2015-June, 2016. Primary identification of isolates was done conventionally using API 20NE systems (Biomerieux), while confirmation was done by sequencing of 16S rDNA and V. alginolyticus specific gene targets, construction of phylogenetic tree using Molecular Evolutionary Genetics Analysis (MEGA) 6.0 software. IGS-Typing and antibiogram test screening were also carried out for isolates. Results: Two hundred and eighty isolates of V. alginolyticus were recovered from farmed fishes. Clinical signs expressed as general septicaemia, ulcers and corneal opacity. Conventional methods identified isolates as V. alginolyticus (ID% 96-99.9), sequencing and phylogenetic tree confirmed this identification. The PCR results for IGS-Typing were expressed as four typical band patterns ≈ 420, 580, 650 and 820 bp, while those for virulence genes were positive amplification for collagenase, toxR and ompK, while tlh and VPI virulence genes were not recognized. Conclusion: Isolated V. alginolyticus were highly pathogenic to farmed fishes as determined by virulence genes identified. Results of IGS-Typing facilitate further identification of isolates by single PCR step as it records the pattern for V. alginolyticus.

  • Fish Processing and Exports on the Zambezi/Chobe Floodplain, Zambezi
           Region, Namibia

    • Abstract: Background and Objectives: In the Zambezi Region, a large number of people, many of whom live below the poverty line, find employment in fish marketing and processing. A survey on fish processing and exports was conducted on the Zambezi/Chobe floodplain between June 2015 and December 2016. The main objective of the study was to assess the fish processing techniques employed by fish processors herein referred as fish vendors to preserve fish products on the Zambezi/Chobe floodplain and to quantify the volume of fish exported from the area. Materials and Methods: Survey questionnaires were used to determine fish vendor’s characteristics and their methods of fish processing and preservation. Daily fish products destined for exports were weighed and recorded using a hanging scale at Wenela border. Results: The study found that women are the key players in the fish processing and preservation on the Zambezi/Chobe floodplain. The major processing techniques employed by the fish vendors were sun drying and smoking. A total of 2515 bags of fish were recorded in 122 days from June, 2015 to December, 2016. Approximately 1575.8 t of fish products worth N$36 million were exported to foreign markets in Zambia. Conclusion: The active involvement of women in the fish processing and export suggested that this sector has the potential to contribute immensely to improving the economic and livelihood of the Zambezi region. However, the fish vendors faced challenges of inadequate cold storage facilities, poor weather and packaging.

  • Morphometric and Meristic Characteristics of Protopterus annectens (Owen,
           1839) in River Niger at Agenebode, Edo State, Nigeria

    • Abstract: Background and Objective: The West African Lung Fish are common to the waters of West Africa. It is the only species in the genus Protopterus of the family Protopteridae found in the West African Swamps, studies have shown that the distribution and abundance of P. annectens are often affected by the construction of dams causing shift in salinity, flood, a significant rise in pollution. Investigation into the morphometric characteristics of P. annectens may provide steps into understanding the phylogeny, growth pattern and morphometric features responsible for survival of this economically important fish species in Nigeria. Materials and Methods: Ten morphometric and meristic characteristics of West African lung fish Protopterus annectens collected from river Niger at Agenebode between the periods of November, 2016 to October, 2017 using different fishing gears were analyzed. Out of the 345 specimens collected, 45 were analyzed to obtain the desired characteristics using graduated ruler and a digital scale.   • Effect of Different Freezing Processes on the Quality and Histological
           Changes of Red Tilapia (Oreochromis niloticus × Tilapia mosambicus)

    • Abstract: Background and Objective: Tilapia fish has been a favorable food and has a long history of farming. The present study was designed to investigate the effects of freezing speed and time on the chemical quality indices as well as histological changes. Materials and Methods: The fillets were frozen and packaged using too slow and quick-freezing methods and stored at -18°C for 6 months. Then fillet changes including drip, total volatile basic nitrogen (TVB-N), peroxide value (PV), thiobarbituric acid value (TBA) were evaluated on a monthly basis. Finally, histological changes were studied every two months by a scanning electron microscope (SEM). Results: All studied qualities changed in the course of different months in both quick and slow-freezing methods, though changes were more noticeable in slow-freezing. Drip percentage (4.8-11.4), PV (0.02-0.93 mEq kg–1), TBA (0.03-1.2 mEq kg–1) and TVB-N (12.63-21.93 mg/100 g) levels all showed greater variations with slow freezing. On the other hand, it detected less degradation in fillets by the freezing method. Conclusion: The changes in Drip, TVB-N, PV and TBA as well as histological were less dramatic in the samples treated by quick freezing, compared to those of slow-freezing samples.
