  • Qualitative Tests of Secondary Metabolite Compounds in Ethanol Extract of
           Spirulina platensis from Karimun Jawa Sea, Indonesia

      Abstract: Spirulina platensis is a spiral green-blue algae that is abundant in the subtropical and tropical water bodies. S. platensis has several potentials to be used in the medical fields due to its wound healing and antitumor potentials. This research aimed to evaluate the secondary metabolite compounds contained in the ethanolic extract of S. platensis from Karimun Jawa Sea, Indonesia. The dried S. platensis was macerated and extracted using ethanol, then was filtered using Whatman filter paper. A series of tests has been done to determine the secondary metabolites in the ozonated oil using several chemistry tests and reagents in the ethanol extract of S. platensis. The tests found that S. platensis microalgae harvested from Karimun Jawa sea contains alkaloid, saponin, flavonoid, and quinone. As S. platensis from different region contains different substances, this research shows the substances contained in S. platensis from Karimun Jawa sea. With this research, we knew that S. platensis from Karimun Jawa islands contains alkaloid, saponin, flavonoid, and quinone which can be used as a medicinal ingredient.

  • Nutritional Content of Bone Flour Made from Plecos Fish Pterygoplichthys
           pardalis from the Ciliwung River, Indonesia

      Abstract: Pterygoplichthys pardalis bones that are thrown into the river is one source of pollutants in the Ciliwung River. P. pardalis bones have a high calcium content, they can be used as an alternative raw material for flour. The purpose of this study was to analyze the nutritional content of P. pardalis bone flour from the Ciliwung River. The research method used was a proximate analysis consisting of moisture, ash, fat, protein, and carbohydrates content analyses. Proximate analysis results showed that the P. pardalis bone plus head flour contained 3% moisture, 35% ash, 34% protein, 23% fat, and 5% carbohydrate. P. pardalis body bone flour contained 4% water, 37% ash, 37% protein, 19% fat, and 3% carbohydrate. Based on the proximate analysis results, P. pardalis bone flours did not met the national standard of SNI 2013 fish flour for quality I, II, and III. Until now, there has been no research that explain the nutritional content of bone flour of P. pardalis from Ciliwung River. This research provides information on the nutritional content of P. pardalis flour. Data obtained from the results of this study can be the basis for consideration in utilizing and processing P.pardalis from the Jakarta Ciliwung River.

  • Potential of Ecology based Agrotourism for Agricultural Education in Besur
           Village, Lamongan Regency

      Abstract: Currently, agriculture has prospective potential to be developed as a tourist attraction, one of which is agricultural tourism with an ecological perspective. The objective of the research was to examine the potential of ecology-based agrotourism as a means for agricultural education in Besur Village, Lamongan Regency. The research applied a quantitative descriptive approach in the form of scoring, weighting, and ranking of the categories being assessed. The research data consisted of primary and secondary data. The data was collected using direct observation and in-depth interviews. The data analysis was carried out using for the feasibility of  agrotourism area. The results showed that the area of Besur Village was classified as suitable (potential) with zone I that had the highest KKA value (2.6) as the object of the agrotourism. Potential ecological aspects obtained analysis 36 species of food producing plants. The highest Importance Value Index (IVI) of plant species was  recorded for Oryza sativa spp. (83.76%) and Zea mays L. (27.11%). Analysis of the physico-chemical properties of soil resulted in moderate or sufficient fertility. Analysis of public perceptions obtained was 4.08 and classified as good for the development of agrotourism in Besur Village. The novelty of this research was to obtain of the potential for agrotourism management in Besur Village through an ecological approach based on the concept of Healthy Plant Management.

  • Stability and Antibacterial Property of Polyherbal Mouthwash Formulated
           Using Local Ingredients

      Abstract: The oral cavity is a home to more than 500 bacterial species. Although some of the oral bacteria are harmless, there are certain species that may cause oral plaques, bad breath, and mouth disease. Thus, maintaining a good oral hygiene is essential for a healthy mouth and body. The present study aimed to formulate a polyherbal mouthwash that may have antibacterial properties. Mouthwash formulations were prepared containing varying percentages of herbal extracts, with each formulation stored at 12 °C and 25 °C. Over the course of 12 weeks, the appearance and pH of the formulated mouthwash were measured. The mouthwash formulations maintained good homogeneity and color when stored at 25 °C, displaying lower pH level ranging between 3.71 and 4.85. Although the mouthwash stored at 12 °C maintained good homogeneity, a change in color in the formulation was evident and a more unstable pH readings were recorded. Antibacterial assay showed that mouthwash formulations stored at 25 °C had better inhibitory activity compared to those stored at 12 °C. Furthermore, mouthwash formulation containing (30% v/v) aleppo oak extract as the major ingredient conferred the greatest inhibition zone diameter (IZD = 10-18 mm) against salivary bacteria compared to formulations with (30% v/v) clove and (30% v/v) turmeric extracts as major ingredients. The best polyherbal mouthwash formulation in terms of inhibiting bacterial growth followed the 3:1:2 ratio for aleppo oak extract, clove extract, and turmeric, respectively. Therefore, the polyherbal mouthwash formulated in this study has the potential to be optimized and commercialized to antagonize growth of pathogenic oral bacteria.

  • The Antioxidant Content and Activity of Various Plant Organs of Kitolod
           (Isotoma longiflora)

      Abstract: Antioxidants play a crucial role in human health owing to their ability to neutralize oxidative reactions. Kitolod (Isotomalongiflora) is one of the medicinal plants that can be used as a source of antioxidant. The aim of study wasto compare the content of antioxidant compounds (flavonoids, phenolics, chlorophylls, carotenoids, and ascorbic acid) in various organs of the kitolod plant. The plant organs studied includingroots, stems, leaves, flowers, and fruits. The extraction of each plant organ wasconducted by maceration technique using ethanol as a solvent at the room temperature for 48 hours. The analysis of total flavonoids, phenolics, and ascorbic acid was performed by colorimetry method using AlCl3, Folin, and sulfosalicyclic acid reagent respectively, while chlorophylls and carotenoids was determined by direct colorimetry method with methanol as a solvent. The antioxidant activity of various plant organs of kitolod was analyzed using the DPPH method. The highest total flavonoid, phenolic, chlorophyll, and carotenoid compounds werefound in leaves, respectively by 10.48, 1.46, 7.25, and 56.98 ppm. The highest ascorbic acid content and antioxidant activity were obtained fromfruits.The research findings provide new and important information aboutthe contents and antioxidant activity of the secondary metabolites (flavonoid, phenolic, chlorophyll, carotenoid, and ascorbic acid) in each organ of kitolod plant. The information from the results of this study can be used to increase the medicinal value ofkitolod plants.

  • Identification of Gamma-Amino Butyric Acid Isolates Lactic Acid Bacteria
           Results from The Isolation of Rusip

      Abstract: Gamma-aminobutyric acid (GABA) is a non-protein amino acid produced by glutamate decarboxylation by the enzyme glutamate decarboxylase and it is widely distributed in plants, animals and microorganisms. GABA-producing microorganisms including Lactic Acid Bacteria (LAB) which play a role in the fermentation process of food such as rusip. Rusip is a preserved food of marine fish made from raw anchovy of Bangka-Belitung. The purpose of this study was to obtain LAB isolates through the process of isolation from rusip and identify the presence of GABA. Isolation of LAB was carried out by the pour method on MRS agar and NA medium. The growing colonies were then characterized based on observations of colony morphology, Gram staining, catalase test, motility test, and fermentation type test. LAB isolates obtained were selected to obtain isolates which are capable to produce GABA using the Thin Layer Chromatography (TLC) method. The results of isolation from rusip and inoculation on MRS agar medium obtained, three LAB isolates namely RSP-A1, RSP-A2, and RSP-A3. Their characteristics are Gram positive bacteria, negative catalase test, negative motility test, and belong to homofermentative and heterofermentative bacteria groups. They were also able to produce GABA. Research on GABA from isolates isolated from rusip is a new thing in the world of GABA research.   • Selection of Soybean (Glycine max) Germplasm Against Biotrophic Fungi
           Disease Based on Anatomical Resistance

      Abstract: The obstacle to increasing the soybean production is an infection of rust disease caused by the biotrophic fungus, Phakopsora pachyrhizi. The research objectives were to determine the anatomical resistance and the level of resistance of soybean cultivars against rust disease. The embedding method for observed leaf structural anatomy. The disease severity based on the method of International Working Group on the Soybean Rust (IWGSR) rating system.  The experiment was arranged as a Completely Randomized Design (CRD) with Factorial Pattern and five times repetition. The first factor was soybean cultivars, namely Gepak Kuning, Slamet, Tanggamus, and Wilis. The second factor was P. pachyrhizi inoculation with 0 uredospores/mL (uninoculated) and 104 uredospores/mL (inoculated). The results showed that the soybean cultivars that have thicker cuticle and epidermis, high trichomes and low stomatal density, and low of stomatal conductance have better anatomical resistance to leaf rust disease. Wilis and Slamet cultivars are resistant cultivars, indicated by disease intensity of 20% and 24.6%, respectively. While the Tanggamus is moderately resistant cultivar and Gepak Kuning is a susceptible cultivar, indicated by disease intensity of 56.5% and 85.3%, respectively. The novelty of selection soybean germplasm against biotrophic fungal disease are important and effectiveness in order to increase the crop productivity. These three soybean cultivars potentially serve as genetic sources to develop high yielding soybean cultivars and resistant to rust disease.

  • Carotenoid Production by Rhodosporidium paludigenum Using Orange Peel
           Extract as Substrate

      Abstract: Carotenoids are fat-soluble pigments that have various benefits in health and beauty. The demand and market of carotenoids are increasing; thus, the faster and cheaper biological production of these pigments using microorganisms is desired. In this study, the optimum medium pH and nitrogen concentration for carotenoid production by Rhodosporidium (R.) paludigenum using orange peel extract as substrate were determined. The soluble sugars from orange peels were extracted using distilled water at 100 °C. The yeast inoculum was cultivated in Yeast Peptone Dextrose (YPD) media before fermentation. The independent variables in the fermentation factorial design included initial medium pH (5; 6; and 7) and nitrogen concentration (0; 1; and 1.75 g/L urea in the medium). The decrease in reducing sugar concentration and the increase in biomass dry weight during fermentation demonstrated carbon source consumption by R. paludigenum for growth and carotenoid production. On the sixth day of fermentation (D6), the culture was harvested for the total carotenoid extraction and determination. The highest total carotenoid yield (107.63 µg/g) was achieved by the treatment group with an initial medium pH of 6 without nitrogen supplementation. This result indicated the orange peel extract potential as a substrate for carotenoid production using R. paludigenum as a workhorse. This was the first reported research in natural carotenoid generation using R. paludigenum (strain InaCC Y-236) locally isolated in Indonesia as the producer and orange peel extract as the substrate. Thus, this research would enrich the microbial carotenoid exploration and waste valorization to value-added products at the same time. The study can then be developed further and upscaled for industrial applications.

  • Bifidobacterium longum, a Predominant Bifidobacterium in Early-life Infant
           Potentially Used as Probiotic

      Abstract: In early life, Bifidobacteria are reported as dominant bacteria in the human digestive tract. Bifidobacterium is potential as a probiotic. The probiotic property of Bifidobacterium is strain-specific. This study aimed to identify the Bifidobacterium (isolated from less than one-month-old healthy infant stool that potentially used as probiotic) based on the 16S rRNA gene and determining their similarities among Bifidobacteria. The probiotic-potentially Bifidobacterium was re-characterized by performing a Gram’s staining and catalase test. The DNA extraction process was followed by the 16S rRNA amplification using 27F-1492R primers. Sequence similarity was checked by using the BLAST program in the GenBank. The phylogenetic tree was constructed by using a neighbor-joining (NJ) method within the MEGA version 7.0 package. The 16S rRNA gene was presented at 1,500 bp length. Bifidobacterium strains have a 91.14-94.26 % sequence similarity to B. longum subsp. longum strain CCUG30698 which is considered as insufficient for species and genus identifications. However, those isolates could be assigned in a phylogenetic position. This present study suggested the B. longum as the dominant strain of Bifidobacterium in the gut of early-life infants which has potential as a probiotic and is considered as an ideal probiotic for human consumption. This study is useful as basic information for other related research, as well as its application in industrial or community service fields.

  • Histopathological of White Rats Aorta Induced by High-Fat Feed After
           Administered by Neem Leaf Ethanolic Extract

      Abstract: Neem (Azadirachta indica A. Juss) is one of the traditional medicines used by local people as antioxidants, anti-hyperlipidemia, anti-inflammatory, and treatment for other diseases such as heart disease. This study aimed to analyze the effect of neem leaf ethanolic extract on aortic wall thickness and aortic lumen diameter of white rats induced by high-fat feed. The male Wistar rats (Rattus norvegicus L.) with 2 months of age were used. This research was an experimental study with a Randomized Complete Block Design consisted of 6 treatments (P0: commercial feed, P1: high-fat feed and duck egg yolk 2.5 ml/200 g BW per oral, P2: P1 + 8 mg/200 g BW simvastatin, P3-P5: P1+ neem leaf extract of 75, 100, and 125 mg/200 g BW respectively) and 4 replications. Aortic preparations were made by the paraffin method and Hematoxylin-Eosin staining. Data were analyzed using ANOVA followed by Duncan Multiple Range Test with 95% confidence level. The result showed that the decrease in wall thickening and lumen narrowing was getting higher along with the increasing doses of ethanol extract of neem leaves. Based on the result of this research, it was found that the high doses of neem leaf ethanolic extract has the same ability with simvastatin to reduce aortic wall thickning and aortic lumen narrowing of white rats. This study is give a new information that the neem leaves can be used as an alternative medicine for cardiovascular disease.

  • Total Phenol Content of Avicennia marina Leaf and Its Relationship to the
           Environmental Quality

      Abstract: Environmental dynamic in the coastal area is suggested to affect the metabolite concentration in mangrove plants. This research aimed to study the concentration of total phenol in A. marina leaf and to analyze the effect of environmental parameters on total phenol content dynamics. Environmental parameters studied in this research was temperature, pH, DO, and salinity, as well as N, P, and C sediment content. Data analysis was carried out through multiple regression of natural logarithm transformed data. Laboratory analysis resulted the value of total phenol content in A. marina leaf ranging from 0.88 – 1.62% with the average concentration of 1.28 ± 0.28%. Regression analysis resulted the significant effect of temperature, DO, and sediment content P and C expressed in the formula: ln(TP) = 31.229 – 7.224ln(T) – 0.067ln(DO) – 1.054ln(P) – 1.241ln(C). The research implicated that the increasing value of those factors was approaching the suitable condition for A. marina. Thus, instead of increasing the phenol concentration, the parameters negatively effect the secondary metabolite. The result showed that increasing temperature, DO, and content of P and C reduced the stress in A. marina and reduce total phenol content. This suggests that low temperature, DO, P and C concentration provides more potential of phenolic products from A. marina.

  • Larvicidal Activity of Brugmansia candida against Aedes aegypti and Culex
           quinquefasciatus (Diptera: Culicidae)

      Abstract: Mosquitoes are well known as vectors of hazardous diseases for human. Plant extracts can be used as an alternative for larval control due to they are a rich source of bioactive chemicals and safe for the environment. The present study investigated the larvicidal activity of crude extracts derived from leaf and flower of Brugmansia candida against the second larval instar of Aedes aegypti and Culex quinquefasciatus. The larval mortality was observed at 24 and 48 h exposure of both leaf and flower extracts, at the concentration of 100, 250, 500, and 1000 ppm. The 24 h exposure of both extracts at the concentration of 500 and 1000 ppm resulted in larval mortality rates were significantly lower than those of 48 h exposure. However, the mortality rate was not significantly different at the lower concentrations of crude extracts. The results also suggested that there was no significant difference in the larvicidal effect between leaf and flower extracts at 24 and 48 h exposure for all concentrations. The LC50 values at 48 h exposure for leaf extract were 789 and 791 ppm for Ae. aegypti and C. quinquefasciatus, respectively, whereas for flower extract were 772 and 780 ppm for Ae. aegypti and C. quinquefasciatus, respectively. Overall, B. candida showed larvicidal activity against Ae. aegypti and C. quinquefasciatus. This research contributes to new finding regarding the larvicidal activity of B. candida. This finding also supports the next study to develop B. candida as an alternative source for larval control agent.

  • Antioxidant Activity of Jojoba (Simmondsia chinensis) Seed Residue Extract

      Abstract: Jojoba (Simmondsia chinensis (Link)) is a shrub plant that widely used as cosmetic ingredients especially jojoba oil. When making jojoba oil, the residue still reminds and become waste. This study aimed to determine the antioxidant activity of jojoba seed residue (JSR). Jojoba seed residue was collected from Sudan. The JSR extracted by maceration with n-hexane, ethyl acetate, and 70% ethanol solvent. The antioxidant activity was determined using DPPH (2,2-diphenyl-1-picrylhydrazyl) assay. The result showed that 70% ethanol extract of JSR had the highest antioxidant activity with 50% Inhibition Concentration (IC50) value of 219.42 mg/L. The 70% ethanol extract was fractionated by using silica gel column chromatography with gradient elution produced 10 fractions. All fractions showed antioxidant activity (IC50106-447 mg/L). Fraction 3 had the highest antioxidant activity with IC50 value of 106.26 mg/L. Therefore, JSR has a great potential for use as a raw material in cosmetic and pharmaceutical products industry, or as a source of bioactive compounds.
