      A member of a specific group of gene-regulating proteins promotes biofilm formation by a bacterium associated with aggressive forms of gum disease. Forming biofilms helps the bacterium to cause persistent infections. Researchers at Karolinska Institutet and Umeå University (Sweden), and University of Zürich (Switzerland), led by Jan Oscarsson at Umeå University, investigated the role of the “histone-like” protein H-NS in Aggregatibacter actinomycetemcomitans infections. These proteins are known to suppress the activity of specific genes in many bacteria, a property confirmed in this research. By studying mutant bacterial strains deficient in H-NS protein, the researchers demonstrated that this protein promotes the formation of biofilms by the bacteria. The results suggest that H-NS plays a significant role in allowing Aggregatibacter actinomycetemcomitans to thrive in biofilms containing mixed populations of bacteria. This effect appears to involve activating production of hair-like appendages called pili on the bacterial surface.       Mixed populations in microbial mats can have cooperative cycles of gene expression like daily circadian rhythms in individual organisms. Henk Bolhuis and colleagues at the Royal Netherlands Institute for Sea Research in the Netherlands studied variations in gene expression in a coastal microbial mat by sampling every six hours in a 24-h period. More than 1000 genes from a variety of microorganisms displayed circadian-like variation, indicative of coherent “molecular clock” processes governed by the 24-h day. The results suggest that microbial mats can be compared to more complex organisms that have different systems working in a cooperative and synchronized manner for the overall advantage of the constituent parts.       One type of the hair-like appendages called pili on the surface of Vibrio vulnificus bacteria promote the formation of resistant biofilms. Vibrio vulnificus inhabits estuaries and warm coastal waters. It typically infects humans by contact with open wounds or ingestion, most notably by eating infected oysters. Infection can cause gastroenteritis, wounds that will not heal, sepsis and death. Dean Rowe-Magnus and Meng Pu at Indiana University, USA, used gene deletion experiments to reveal the importance of a specific gene required to create one category of pili. In the absence of the gene the ability of the bacterial cells to explore, attach to and colonize surfaces was significantly diminished.       While bathing typically means good hygiene, bath toys can serve as incubators for microbial growth. Microbes colonize nearly every natural and human-made surface, sometimes living within complex communities called biofilms. A team led by Frederik Hammes at the Swiss Federal Institute of Aquatic Science and Technology found that tap water bacteria and fungi readily formed biofilms inside bath toys, suggesting that bathing provides food for microbes. These nutrients may come from bath toys’ polymeric material, from care products like soap and from human secretions like sweat. While 16S rRNA sequence analysis found that some of the microbes were related to disease-causing strains, future work is needed to assess the disease risk from these bath toy-associated biofilms.       Computer simulations yield useful insights into strategies bacteria in biofilms use to switch into antibiotic-tolerant “persister” states. A diverse range of mechanisms are available to promote the transition into persister forms. The populations of cells in persister states are proving to be more varied than previously thought. Researchers in France, led by Gabriel Carvalho at the Complex Systems Engineering Laboratory in Aubière, modelled different switching strategies that allow biofilms to survive, recover and grow despite antibiotic treatments. Their procedure examines the effect on switching rates of a greater range of variable factors than previous simulation methods. These variables include the concentration and flow patterns of relevant chemical compounds, changing environmental conditions, and the challenge presented by different antibiotics.       A method to model and assess the effect of interactions and processes that inhibit biofilm growth closes a gap in understanding. Biofilm growth is often inhibited by chemicals used or produced by microorganisms, or by other components and characteristics of the surroundings. This has been a neglected aspect of the procedures used to analyze and predict biofilm behavior. Stefan Wuertz at Nanyang Technological University in Singapore, with colleagues in Singapore and Argentina, mathematically modeled these often-overlooked inhibitory effects. They tested the accuracy of their procedure on complex biofilms under a variety of growing conditions. The insights gained into growth inhibition should help with the design of bioreactors, including those used to degrade pollutants.
