      Tuberculosis, caused by Mycobacterium tuberculosis, is the leading cause of death due to a single infectious agent. New therapeutic options are needed, and repurposing clinically approved drugs to destroy biofilms is an attractive approach, as these microbial communities are often less susceptible to antibiotics. A team lead by Seyed Hasnain at the Indian Institute of Technology Delhi identified an enzyme, PpiB, from M. tuberculosis that promoted biofilm formation and showed that PpiB interacts with several drugs that are currently used to treat diabetes, immunological diseases and cancer. These drugs destabilise M. tuberculosis biofilms in culture and enhanced the potency of two current anti-tuberculosis antibiotics.       Pseudomonas aeruginosa bacteria are highly adaptable and dominate different environments—notably biofilms—yet the biological and ecological factors that support this dominance are still poorly understood. Here, Yingying Cheng and Liang Yang at the Nanyang Technological University, Singapore, used multiplexed gene expression analysis of mixed-species microbial communities to monitor transcriptional changes in P. aeruginosa, and reveal the upregulation of the H1 type VI secretion system and the expression of the type IV pili and Psl exopolysaccharide as central events in establishing the dominance of P. aeruginosa over other bacterial species growing in biofilms.       Repeatedly sampling oral bacterial species in 26 volunteers over one month revealed considerable personal differences in the bacteria associated with oral disease, and some significant variation in individuals at different times. The findings come from an international team of researchers led by Mária Džunková at the Foundation for the Promotion of Health and Biomedical Research of Valencia Region, Spain. They emphasize the importance of repeated sampling to understand the significance of changing and varying oral bacterial populations in both health and disease. The study used chemical markers of ‘oxidative stress’ due to inflammation as an indication of the presence of disease-causing bacteria. Different bacterial combinations seem associated with differing oxidative stress levels in different people.       A rapid test for the presence of bacterial cellulose in urine allows urinary tract infections due to bacteria in the biofilm state to be distinguished from less problematic ‘planktonic’ infections caused by free-living bacteria. Agneta Richter-Dahlfors and colleagues at the Karolinska Institute in Stockholm, Sweden, developed the test based on the fluorescence of oligothiophene molecules when they bind to biofilm cellulose in urine. The researchers tested the procedure using ‘uropathogenic Escherichia coli‘ (UPEC) infections in patients. The results also confirmed that UPEC infections can indeed form biofilms in the urinary tract. Previously, there has been no way to readily distinguish between urinary tract infections caused by bacteria in the free-living and biofilm states.       Biofilms in drinking water premise plumbing systems can be disrupted and their microorganisms deactivated by exposure to jets of ionized gas known as plasma. Researchers at the University of Illinois, USA, led by Thanh (Helen) Nguyen and J. Gary Eden, explored the potential of low temperature plasma jets in disrupting & removing drinking water biofilms. The plasma was directed through fabricated microchannels and onto samples that the simulated biofilms. The interaction of the plasma with air and water generated reactive chemical species and ultraviolet radiation that disrupted the biofilms and deactivated the microorganisms within them. The biofilms studied vanished within 20 min. of plasma exposure.
