  • Effect of Prolonged Administration of Artemether-Lumefantrine on
Testicular Biomarkers of Oxidative Stress: Ameliorative Effect of Vitamin
           E

    • Abstract: Publication year: 2018Source: Basic Sciences of Medicine , Volume 7, Number 1Onuche A. Daikwo, Mohammed U. Kawu, Rabiu A. Magaji, Ejike D. EzeRecurrence of malaria infection in Nigeria encourages frequent use of artemisinin-based combination therapies (ACTs). The ACTs function via generation of free radicals. Reactive oxygen species have been implicated in the etiology of male infertility. The present study evaluated prolonged administration of artemether-lumefantrine (AL) and the ameliorative effect of vitamin E (VE) on testicular biomarkers of oxidative stress. Adult male Wistar rats (n = 35) were divided into five groups of seven rats each and treated twice daily for a period of 2 weeks via oral route as follows; group I: 1ml/kg distilled water; group II: 1ml/kg olive oil; group III: 2.29 mg/13.74 mg/kg AL; group IV: 2.29 mg/13.74 mg/kg AL + 100 mg/kg of VE; group V: 2.29 mg/13.74 mg/kg AL + two weeks recovery (allowed to recover from AL treatment for 2 weeks before sampling). At the end of the experiment, animals were sacrificed and testes harvested for analysis of testicular biomarkers of oxidative stress. Testicular concentrations of antioxidant enzymes glutathione peroxidase (GPx) and catalase (CAT) were significantly (P < 0.05) higher in groups IV and V rats when compared with other groups. Testicular malondialdehyde (MDA) level was increased, while superoxide dismutase (SOD) was decreased in group III as compared to other groups, though these changes were not significant. It is concluded that, prolonged administration of artemether-lumefantrine induces significant alterations in testicular antioxidant enzymes activity suggestive of oxidative stress and that vitamin E and cessation of treatment with artemether-lumefantrine appreciably ameliorates the alterations in testicular antioxidant enzymes activity.

Thirty five (35) adult female rats were used for this study. At parturition, the animals were randomly divided into seven (7) groups of five (5) rats each (n=5) and treated as follows: Group I: (Normal control) was given normal feed and distilled water, orally (1 ml/kg bw), Group II: metoclopramide (5 mg/kg bw), Group III: 100 mg/kg bw of Vitamin C, Groups IV, V and VI were given soya bean supplement thus; 10%, 20% and 40%, respectively. Group VII was co-administered with 20% soya bean supplement and Vitamin C (100 mg/kg bw). Treatment was done for the period of ten (10) days at 06:00 pm daily. The result of serum level of Growth hormone showed significant increase (P < 0.05) in the groups treated with vitamin C (VIT C), Soya Bean (SB) 10%, SB 20% and SB 20% + VIT C. compared to the control group. Progesterone level was significantly high (P

  • Study of Glutathione-S-Transferase, Cancer Embryonic Antigen and
Carbohydrate Antigen 19-9 in Ulcerative Colitis and Colorectal Cancer

    • Abstract: Publication year: 2015Source: Basic Sciences of Medicine , Volume 4, Number 4Fatma Saffeyeldin Mohamed, Mirhan M. Elkady, Eman Fekry Mohamed, Boshra E. Hussein, Walid S. H. ElsaiedBackground: Ulcerative colitis (UC) is a chronic inflammatory bowel disease (IBD) with a high risk of colorectal cancer (CRC), which is a major cause of cancer morbidity and mortality worldwide. Glutathione-S-transferase (GST) has an essential role within the cells, including the conjugation and detoxification of toxic or carcinogenic compounds, such as reactive oxygen species (ROS). Cancer embryonic antigen (CEA) and carbohydrate cancer antigen (CA19-9) are tumor markers that are used for diagnosis of colorectal cancer. Aim: The aim of the study was to assess the total plasma GST activity, CEA and CA 19-9 in ulcerative colitis and colorectal cancer. Subjects and methods: The plasma GST activity, CEA and CA 19-9 were assessed in 40 patients with CRC (group I), 40 patients with UC (group II) and 20 apparently healthy individuals as controls (group III). Results: The mean GST activity, CEA and CA19-9 were significantly higher in CRC and UC patients than controls with no significant difference between CRC & UC patients in CEA or CA19-9. A significant difference was found between Gs I & II as regards GST activity. Conclusion: Measurement of the total plasma GST activity may be helpful as a tumor marker of colorectal cancer.

  • The Preventive Effects of Avocado Fruit and Seed Extracts on
           Cardio-nephrotoxicity Induced by Diethylnitrosamine/2-acetylaminoflurine
           in Wistar Rats

    • Abstract: Publication year: 2017Source: Basic Sciences of Medicine , Volume 6, Number 1Adel Abdel-Moneim A., Osama M. Ahmed, Hanaa I. Fahim, Eman E. MohamedThe current study was designed to evaluate the possible preventive effects of avocado (Perseaamericana) fruit and seed hydroethanolic extracts on diethylnitrosamine/2-acetylaminoflurine (DEN/2AAF)-induced cardio-nephrotoxicity in Wistar rats. The Wister rats used in this study were allocated into 4 groups. Group I was kept as a normal group, while the other 3 groups were administered a twice intraperitoneal dose (150 mg/kg bw./week) of DEN, followed by daily 2AAF-administration by oral gavage four days per week for three weeks. Group II was DEN/2AAF-administered control group and groups III and IV were administered DEN/2AAF and were orally given avocado fruit and seed hydroethanolic extract for 20 weeks. DEN/2AAF-administration induced kidney and heart injury evidenced by histological alterations as well as significant increase of serum indicators of impaired kidney function including creatinine, urea and uric acid levels and serum marker of impaired heart function including CK, CK-MB, LDH and AST activities. On the other hand, the renal and cardiac lipid peroxidation was elevated while renal and cardiac glutathione content and activities of superoxide dismutase, glutathione peroxidase and glutathione-s-transferase were significantly declined as a result of DEN/2AAF-administrations. Concomitant supplementation with avocado fruit and seed hydroethanolic extract potentially prevented DEN/2AAF-induced biochemical and histopathological deleterious alternation. In conclusion, the results suggest that avocado fruit and seed hydroethanolic extracts successfully have ameliorative effects against DEN/2AAF-induced cardio-nephrotoxicity via the enhancement of the antioxidant defense system and suppression anti-inflammatory actions.

  • Arcanobacterium Hemolyticum Chorioamnionitis Leading to Preterm Labour
           – A Case Report

    • Abstract: Publication year: 2017Source: Basic Sciences of Medicine , Volume 6, Number 1Deepthy BalakrishnanArcanobacterium hemolyticum has been identified as a cause of upper respiratory tract infection and pyoderma. The presence of this organism in amniotic fluid is very rare. We report a case of subclinical chorioamnionitis due to Arcanobacterium hemolyticum. Mother did not have the typical features of chorioamnionitis except for foul smelling amniotic fluid but developed preterm labour with intact membranes. The diagnosis is usually difficult as they may be viewed as contaminants or normal flora.

  • Aberrant Configuration and Branching Pattern of Circumflex and Profunda
           Femoris Arteries

    • Abstract: Publication year: 2016Source: Basic Sciences of Medicine , Volume 5, Number 1Rajani Singh, Munish WadhwanDuring dissection of femoral triangles of female cadaver aged 70 years fixed in 10% formalin for teaching purpose in the department of Anatomy, AIIMS Rishikesh, variant configuration of profunda femoris and its branches were observed bilaterally. On right side, common trunk arose from femoral artery 4 cm distal to inguinal ligament. Instantly the common trunk gave medial circumflex femoral artery. The common trunk descended further for 2 cm and trifurcated into lateral circumflex femoral, profunda femoris arteries and muscular branches. Such branching pattern of profunda femoris artery is unique. On the left side, femoral artery gave common trunk on medial aspect at a distance of 4 cm from inguinal ligament. This common trunk bifurcated into medial and lateral circumflex femoral arteries. Then femoral artery gave origin to profunda femoris artery at a distance of 6 cm below the inguinal ligament. Such branching pattern of profunda femoris and femoral artery is also rare. Variations in the branching pattern of femoral and profunda femoris arteries have clinical implications while performing various therapeutic and diagnostic procedures around femoral triangle. Hence the case has been reported.

  • Anomalous Configuration of Medial and Lateral Circumflex Femoral Arteries

    • Abstract: Publication year: 2016Source: Basic Sciences of Medicine , Volume 5, Number 1Rajani SinghNormally profunda femoris artery arises from femoral artery below the inguinal ligament. Profunda femoris artery gives rise to lateral circumflex artery laterally and medial circumflex artery medially. But abnormal configuration of profunda femoris artery and its branches was observed in two cases in present study during dissection of lower limbs for teaching purpose in the department of Anatomy AIIMS Rishikesh, India. In one case, femoral artery trifurcated into medial circumflex, lateral circumflex and profunda femoris arteries. This finding is rare. In another case, common trunk arose from femoral artery which descended for 2.5 cm and gave medial circumflex artery. This common trunk descended further for 1 cm and bifurcated into lateral circumflex and profunda femoris arteries. This finding is new and unique and hence the case is reported. Knowledge of these variations will be of utmost importance to surgeons carrying out surgical procedures around the femoral triangle, to radiologists to avoid misinterpretation of radiographs and to anatomists for new and rare variants.

  • The Protective Effect of Pumpkin Seed Oil on Azathioprine – Induced
Hepatic Toxicity in Adult Male Albino Rats: Histological and
           Immunohistochemical Study

    • Abstract: Publication year: 2014Source: Basic Sciences of Medicine , Volume 3, Number 4Fatma Sayed Abdel AalBackground: Pumpkin is an herbal medicine rich in unsaturated fatty acids (UFAs), which represent about 84% of the total fatty acids. It is also rich in minerals and antioxidants as tocopherols and carotenoids. It has many medicinal properties like anti-diabetic, antioxidants, anti-inflammatory and anti-carcinogens. Azathioprine (AZ)(imuran) is an immunosuppressive drug used to treat autoimmune diseases in the skin, blood, and multiple body systems. On the hand it also causes damage to remote organs.MaterialandMethods: Twenty four adult male albino rats were used in the present study. They were divided into four equal groups. GroupI, was untreated and used as a control group. GroupII,groupIIIand groupIV were treated with pumpkin(4ml/day), azathioprine (15mg/day) andpumpkin/azathioprine (4 ml/15mg/day) orally respectively for four weeks. At the end of the experiment blood was collected for biochemical study. Tissues samples from the liver were collected from all rats for histological and immunohistochemical study.Aimofthe work:to evaluate the possible protective role of pumpkin seed oil on hepatic injury induced by AZ treatment in male albino rats. Results: Azathioprine injection elevated plasma level of SGPT, SGOT and ALP compared to control group. Pumpkin seed oil improved these results to some extent.Light microscopic examination showed hepatic tissues with hemorrhage, cellular vacuolation, and dilation of blood vessels, cellular infiltration and intense immune reaction for caspase-3. The ultrastructure showed the cytoplasm of hepatocytes appeared rarified with marked vacuolation and destroyed organelles. The rats treated concomitantly with both AZ and pumpkin seed oil showed significance attenuation of the histological and immunohistochemical alternations. Conclusions:These results suggest that Pumpkin seed oil has a protective effect against Azathioprine induced toxicity, may be via its antioxidant role.

  • Abnormal Cord Formation of Brachial Plexus

    • Abstract: Publication year: 2016Source: Basic Sciences of Medicine , Volume 5, Number 1Rajani Singh, R. Shane Tubbs, Raj KumarBrachial plexus is very complicated nerve network with umpteen numbers of variations. During routine dissection of a 70-year old female cadaver, an anomalous single cord was detected after fusion of all the three trunks without splitting into anterior and posterior divisions. This enhances the complexity of configuration of axons and fascicles making clinical investigation and treatment through nerve cuff electrode, functional electrical stimulation and other micro-neurosurgical interventions more cumbersome. Therefore the case is reported for a rare variant to unfold the complexity theoretically, by glimpses of fascicular anatomy. The study will be useful to anesthetists, surgeons, radiologists and anatomists.

  • Should a Highly Skilled Surgeon be an Advanced Anatomist first' - A
           View Point

    • Abstract: Publication year: 2015Source: Basic Sciences of Medicine , Volume 4, Number 4Rajani Singh, R. Shane TubbsSkilled surgeons treat their patients by repair/removal/cutting/alteration/replacement of the diseased part/organ. So before starting to make an incision on a patient, the surgeon requires not only confidence, a sound knowledge of the organization of macro/micro forms and structures and their shapes, sizes and locations and the correct diagnosis of the disease, but also the anatomical relationship to the disease. The aim of this study is to explore the intimate involvement of anatomy with successful surgery in order to help reduce the failure rate and consequently the incidence of litigious claims. The role of anatomy in surgical procedures from diagnosis to completion is analyzed. The current failure rates in surgery due to lapses in the anatomical knowledge felt by many surgeons are revealed. Remedial measures to ensure safe medical practice have been proposed by senior surgeons and regulators involved in settling the increasing number of litigious claims, such as improving knowledge by inviting educationists to oppose the lowering standards of anatomy teaching among passing-out doctors. A balanced scheduling of medical curricula is required with emphasis on the synergistic vertically and horizontal integration of anatomy, besides mastering subject as it is taught at the outset of the course. Successful surgery and in-depth knowledge of anatomy are complementary to each other. Human anatomy can be practiced by surgeons on cadavers to help improve surgical success.
