  • Physiological characterization of two native yeasts in pure and mixed
           culture using fermentations of agave juice

    • Abstract: Yeast cells are subjected to diverse environmental conditions during the alcoholic fermentation of agave juice, causing different kinetic behaviors. Agave juice was used as culture medium to evaluate the kinetic behavior of Saccharomyces cerevisiae ITD-00185 and Torulaspora delbrueckii ITD-00014a, as pure and mixed cultures, under different inoculum sizes (1×105, 1×106, 1×107 and 1×108 cells mL-1), and combined pH levels (3.5, 4.0 and 4.5) and temperatures (18 °C, 28 °C and 38 °C). Saccharomyces cerevisiae displayed high fermentation capacities at all inoculum concentrations assayed. However, T. delbrueckii required a high inoculum concentration (≥1×107 cells mL-1) to perform at fermentation levels similar to S. cerevisiae. Low temperatures (18 °C) slowed fermentation, while high temperatures (38 °C) adversely affected the development of the yeast strains, especially T. delbrueckii. The best temperature was 28 °C in all fermentations. The pH level had a strong effect on the performance of the coculture, since the fermentation kinetics suggested a synergistic effect at pH 4.5, while an antagonistic effect was postulated at pH 3.5. In all of the mixed culture cases, a positive effect at 28 °C, especially at pH 4.0 and 4.5, was demonstrated by greater levels of sugar consumption and ethanol production (~20%, p<0.05) compared to fermentations of the S. cerevisiae monoculture. The coculture results allow us to postulate that a complex interaction exists between the two yeasts, which could be synergistic or antagonistic, as the environmental conditions change.Resumen Las levaduras se someten a diversas condiciones ambientales durante la fermentación alcohólica de jugo de agave, causando diferentes comportamientos cinéticos. Se utilizó jugo de agave como medio de cultivo para evaluar el comportamiento cinético de Saccharomyces cerevisiae ITD-00185 y Torulaspora delbrueckii ITD-00014a, en cultivos puros y mezclados, en diferentes condiciones de tamaño de inóculo (1×105, 1×106, 1×107 y 1×108 células ml-1) y combinaciones de pH (3.5, 4.0 y 4.5) y temperatura (18 °C, 28 °C y 38 °C). Saccharomyces cerevisiae mostró alta capacidad fermentativa en todas las concentraciones de inóculo. Torulaspora delbrueckii requirió alta concentración de inóculo (≥1×107 células mL-1) para fermentar como S. cerevisiae. A 18 °C se retardó la fermentación, mientras que a 38 °C se afectó negativamente al desarrollo de las levaduras, especialmente T. delbrueckii. La fermentación fue vigorosa para ambas levaduras a 28 °C. El pH tuvo un fuerte efecto sobre el rendimiento del co-cultivo, sugiriendo un efecto sinérgico a pH 4.5 y efecto antagónico a pH 3.5. Para cultivo mixto se observó un efecto positivo a 28 °C, especialmente a pH 4.0 y 4.5, demostrando mejor consumo de azúcar y producción de etanol (~ 20%, p <0,05) comparado con el monocultivo de S. cerevisiae. Los resultados globales permiten postular una interacción compleja entre las dos levaduras, que puede ser sinérgica o antagónica, según las condiciones ambientales.
       
  • Propagation and bulblet enhancement of Rhodophiala pratensis from seeds
           germinated in vitro

    • Abstract: Rhodophiala pratensis is an endemic geophyte from Chile with great importance to the national market of cut flowers and potted and garden plants. However, many geophytes have conservation issues and are considered endangered species. Efficient and controlled propagation techniques have been useful for the commercial production of this species and for its conservation. A micropropagation protocol using seeds was developed to determine an optimum method to increase bulb production rates and plant production. The effect of sucrose levels in the medium on the growth of bulbs was tested in vitro (30, 60 and 90 g L-1). Then, the effects on shoot and bulb induction of the cytokinin BAP at concentrations of 1.0, 2.0 and 4.0 mg L-1 with the auxin NAA at concentrations of 0.1, 1.0 and 2.0 mg L-1 were analyzed. The germination rate after eight weeks increased to 86% with very low contamination (7%). Bulb growth increased when high levels of sucrose were added to the medium; the optimum sucrose concentration was 90 g L-1. Acclimatization was successful, with a plant survival rate of 63% and an average bulb growth of 0.5 cm. This is the first study to demonstrate the possibility of propagating R. pratensis seeds in vitro.Resumen Rhodophiala pratensis es un planta geófita endémica de Chile con gran importancia para el mercado nacional de flores, macetas y plantas de jardín. Sin embargo, muchas de estas geófitas tienen problemas de conservación, siendo consideradas especies en peligro de extinción. Para la conservación y producción comercial de esta especie, las técnicas de propagación eficiente y controlada han sido muy útiles. Por esto, se desarrolló un protocolo de micropropagación utilizando semillas para determinar un método óptimo que permita aumentar las tasas de producción de bulbos y la producción de plantas. Se probó el efecto de distintas concentraciones de sacarosa en el medio de cultivo sobre el crecimiento in vitro de los bulbos (30, 60 y 90 g L-1). A continuación, se analizaron los efectos de la citoquinina BAP en concentraciones de 1.0, 2.0 y 4.0 mg L-1 con auxina NAA en concentraciones de 0.1, 1.0 y 2.0 mg L-1 en la inducción de brotes y bulbos. La tasa de germinación después de ocho semanas aumentó a 86% con una contaminación muy baja (7%). El crecimiento del bulbo aumentó cuando se añadieron altos niveles de sacarosa al medio, con una concentración óptima de 90 g L-1. La aclimatación tuvo éxito con una supervivencia de plantas del 63%, con un promedio de crecimiento de bulbo de 0.5 cm. Esta es la primera investigación que demuestra la posibilidad de propagar semillas de R. pratensis in vitro.
       
  • Evaluation of the growth, yield and nutritional quality of pepper fruit
           with the application of Quitomax®

    • Abstract: The consumption of vegetables as a source of vital nutrients is very important for human health. In the present work, the effects of the application of several concentrations of Quitomax® and a control treatment on the growth, yield and nutritional value of bell pepper (Capsicum annuum L. variety Lical) fruit were evaluated. The treatments consisted of the foliar application of Quitomax® at 200, 300 and 400 mg ha-1 20 days after transplantation. A randomized block design with four replicates per treatment was applied. The number of fruit harvested as well as their length and width, the fresh mass of the fruit, the thicknesses of the mesocarps, the mass of the peduncles with seeds, the yield, the nutritional quality including the soluble solid, vitamin C, carotenoid, and polyphenol content and the antioxidant activity of the fruit were evaluated. Quitomax at a concentration of 300 mg ha-1 produced significantly more pepper fruit with the largest dimensions and highest fruit mass, yield and antioxidant activity than all other treatments. Therefore, Quitomax can be used as a promissory stimulant for bell pepper crops.Resumen El consumo de vegetales como fuente de vital de nutrientes es muy importante para la salud humana. En el presente trabajo se evaluaron la aplicación de varias concentraciones de Quitomax® y un tratamiento control sobre variables del crecimiento, rendimiento y valor nutricional de frutos de pimiento variedad Lical. Las plantas se obtuvieron de semillas sembradas en charolas de polietilenos y a los 25 días de edad se trasplantaron a bolsas de 1 kg conteniendo como sustrato Sogemix y arena. Los tratamientos consistieron en la aplicación foliar de Quitomax® a 200, 300 y 400 mg·ha-1 a los 20 días después del trasplante. Se aplicó un diseño de bloques al azar con cuatro réplicas por tratamientos. Las evaluaciones realizadas fueron: el número de frutos o cosechados así como su largo y ancho, también masa fresca de los frutos, grosores de los mesocarpios y masas de los pedúnculos con semillas además del rendimiento y los variables de calidad nutricional contenidos de sólidos solubles, vitamina C, carotenoides, polifenoles y actividad antioxidante de los frutos. Los resultados mostraron que la concentración de Quitomax a 300 mg ha-1 produjo significativamente el mayor número de frutos de pimiento con las mayores dimensiones, mayores masas de frutos, rendimiento y actividad antioxidante de todos los tratamientos. Por lo tanto, Quitomax pudiera ser usado como un estimulante promisorio en el cultivo del pimiento
       
  • Decrease of flavonol synthase enzymatic activity in Ugni molinae Turcz due
           to the domestication process

    • Abstract: Flavonoid biosynthesis may be affected by plant domestication, with flavonoid production being reduced in proportion to the degree of domestication. In this context, kaempferol (3,4´,5,7-tetrahydroxyflavone) has been identified in the leaves of wild and cultivated Ugni molinae, a berry endemic to Chile. The biosynthetic pathway of kaempferol production begins with naringenin (4´,5,7-trihydroxyflavanone), which is converted to dihydrokaempferol (3,4´,5,7-tetrahydroxyflavanone), catalyzed by flavanone 3ß-hydroxylase (FHT) and then converted to kaempferol by a bifunctional enzyme called flavonol synthase (FLS). Therefore, our study aims to evaluate how FLS activity is affected in murtilla plants that are subjected to the domestication process. Kaempferol was quantified from methanolic extracts of leaf samples collected from both cultivated and wild U. molinae plants using high-performance liquid chromatography, and enzyme extraction was performed to determine FLS activity. The results showed that kaempferol concentration in wild plants from the Soloyo (0.14 μg g-1), Mehuín (0.18 μg g-1) and Queule (0.25 μg g-1) sampling areas was higher than in their cultivated counterparts. Our data are consistent with the FLS activity detected in samples obtained from Manzanal Alto (134.79 pKatal, Soloyo (96.48 pKatal), and Mehuín (119.97 pKatal). These samples also exhibited higher enzymatic activity than their cultivated counterparts. Together, these data suggest that FLS activity is negatively affected by the domestication process.Resumen La biosíntesis de flavonoides puede verse afectada por la domesticación vegetal, reduciéndose la producción en relación al grado de domesticación. En este contexto, ha sido identificado el kaempferol (3,4´, 5,7-tetrahidroxiflavona) en hojas de Ugni molinae silvestre y cultivada. La ruta biosintética del kaempferol comienza con naringenina (4´, 5,7-trihidroxiflavanona), que se convierte en dihidrokaempferol (3,4´, 5,7-tetrahidroxiflavanona), catalizada por la flavanona ٣ß-hidroxilasa (FHT). Finalmente, se convierte en kaempferol por una enzima bifuncional llamada flavonol sintasa (FLS). Por lo tanto, nuestro estudio tiene como objetivo evaluar cómo se ve afectada la actividad de FLS en plantas de murtilla que están sujetas a un proceso de domesticación. El kaempferol fue cuantificado a partir de los extractos metanólicos de hojas recogidas en plantas cultivadas y silvestres de U. molinae mediante cromatografía líquida de alta resolución. Además, se realizó una extracción de enzimas para determinar la actividad de FLS. Los resultados mostraron que la concentración de kaempferol en las áreas de muestreo silvestre de Soloyo (0.14 μg g-1), Mehuín (0.18 μg g-1) y Queule (0.25 μg g-1) fue mayor que en sus contrapartes cultivadas. Estos datos fueron consistentes con la actividad de FLS determinada en muestras obtenidas de Manzanal Alto (134,79 pKatal), Soloyo (96,48 pKatal) y Mehuín (119,97 pKatal). Estos también exhibieron una mayor actividad enzimática que sus contrapartes cultivadas. Por lo tanto, nuestros datos sugieren que la actividad de FLS se ve reducida por la domesticación vegetal.
       
  • Arthropods of forestry and medical-veterinary importance in the Limarí
           basin (Coquimbo region, Chile)

    • Abstract: The Limarí River valley, located in the Coquimbo Region of Chile, is an important area for agricultural production that pertains to the transverse valleys ecoregion, known as Norte Chico. In recent decades, the continuous expansion of agriculture towards the dryland areas has favored the introduction and establishment of potential pests, such as arthropods, that may affect forestry crops and transmit zoonotic diseases. We analyzed the arthropod assemblages that were of forestry and medical-veterinary importance in the Limarí basin. After reviewing historical data, specimen collections, and the available literature, a total of 10 families, 39 genera, and 51 species of arthropods of forestry importance were recorded, 43.1% of which were of quarantine importance. Xylophagous groups of quarantine importance to the US included Cryptotermes brevis (Kalotermitidae), Neoterius mystax (Bostrichidae), Tyndaris planata (Buprestidae), Rhyephenes humeralis (Curculionidae) and Phoracantha recurva (Cerambycidae). Other quarantine insects of forestry importance recorded in the area included were Micrapate scabrata and Polycaon chilensis (Bostrichidae). In addition, the groups of medical-veterinary importance were represented by 11 families, 14 genera, and 18 species, 9 of which were arachnids and 9 were insects. Among the identified arachnids, the most important genera were Loxosceles (Sicariidae), Latrodectus, Steatoda (Theridiidae) and Rhipicephalus (Ixodidae), whereas among the identified insects, the most important were Triatoma and Mepraia (Reduviidae), Pediculus (Pediculidae), and Ceratophyllus (Ceratophyllidae). We hope this study serves as a basis for the development of forest pest monitoring and control programs as well as an understanding of the potential risks posed by zoonotic agents and their implications for public health.Resumen El valle del Río Limarí, en la Región de Coquimbo, es un importante foco de producción agrícola en los valles transversales del Norte Chico. En las últimas décadas, la constante expansión de los cultivos agrícolas hacia zonas de secano ha permitido la introducción y establecimiento de potenciales plagas, como los artrópodos, que pueden afectar las producciones forestales y constituir elementos zoonóticos. Se analizó la importancia forestal y médico-veterinaria de los artrópodos de la cuenca del Limarí. Mediante la revisión de bases de datos históricas, colecciones y literatura disponible, se registró un total de 10 familias, 39 géneros y 51 especies de artrópodos de importancia forestal, de las cuales el 43,1% fueron de importancia cuarentenaria. Entre los elementos xilófagos de importancia cuarentenaria para EE.UU. destacaron los registros de Cryptotermes brevis (Kalotermitidae), Neoterius mystax (Bostrichidae), Tyndaris planata (Buprestidae), Rhyephenes humeralis (Curculionidae) and Phoracantha recurva (Cerambycidae). Otros insectos de importancia forestal reportados fueron Micrapate scabrata y Polycaon chilensis (Bostrichidae). Por su parte, los elementos de importancia médico-veterinario estuvieron representados por 11 familias, 14 géneros y 18 especies, de las cuales 9 especies correspondieron a arácnidos y 9 a insectos. Dentro de los arácnidos destacaron los géneros Loxosceles (Sicariidae), Latrodectus, Steatoda (Theridiidae) and Rhipicephalus (Ixodidae), mientras que dentro de los insectos las más importantes fueron Triatoma y Mepraia (Reduviidae), Pediculus (Pediculidae), y Ceratophyllus (Ceratophyllidae). Se espera que la información del presente estudio sirva de base para desarrollar programas de monitoreo y control de potenciales plagas forestales, así como también entender los potenciales riesgos de elementos zoonóticos con implicancias para la salud pública.
