  • Self-Association of the Macromolecules of Aldrich Humic Acid in Aqueous
           Solutions Using Ultraviolet–Visible (UV–Vis) Absorption Spectroscopy

      The ultraviolet–visible (UV–Vis) spectra of the aqueous solutions of humic acids (HA) Aldrich were obtained within the concentration range 1–20 mg/L (0.1–2ċ10–6 mol/L). A conclusion on the existence of the self-association (dimer formation) of HA macromolecules is based on the deviation of the concentration dependence of optical density (OD) from the linear one at [HA] > 10 mg/L (>10–6 mol/L). An original, mathematical algorithm is proposed to determine a dimerization constant K and a molar coefficient of dimer absorption [math] The value of K was (2.56 ± 0.07) ċ 106 L/mol. The calculated molar coefficients of HA macromolecule dimer absorption, [math], indicate an increase in the dimer extinction coefficient [math] compared to the double value of the molar monomer absorption 2[math]. It was established that the spectral dependence of the value β = [math]/[math] exhibits a minimum within a wavelength range of 300–450 nm, which is due to the difference in the efficiency of the interaction of various sites upon HA macromolecule dimerization. Thus, an approach of studying the processes of self-association is proposed using the method of UV–Vis absorption spectroscopy. This method is implemented for molecules that do not have characteristic absorption bands. The proposed method can also be successfully applied to molecules with characteristic absorption bands.

      A burial and a rich offering were found under Room 2 in the Murals Building, Bonampak, a Mayan archaeological site situated in Chiapas, Mexico. This burial may be related with the creation of the famous mural paintings. A rich set of jewelry made of green stones was among the different objects found. Green stones have great importance in Mesoamerican cultures, those composed of jadeite being the most appreciated. To characterize the green stones, different spectroscopic techniques were used in a complementary way: Raman and infrared spectroscopies (FT-IR) were used for global mineralogical analysis, while X-ray diffraction (XRD) and X-ray fluorescence (XRF) were applied simultaneously in situ on the artifacts that were not successfully identified by these molecular techniques. In addition, XRF was used to contrasts the elemental information from pieces composed of pyroxenes that may be related to the raw sources of jade in Guatemala. The main minerals identified within the beads and earrings were jadeite with omphacite and jadeite with albite; to a minor extent, quartz, and serpentine. In this paper, the main features of the molecular and X-ray techniques are compared in order to determine the advantages and limitations of these spectroscopies for mineral identification. With this combination of techniques, it was possible to undertake a suitable characterization of the analyzed objects. This paper focuses on the XRD–XRF combined analysis for in situ noninvasive characterization.

  • Analytical Solution to the Depth-of-Origin Profile of Transmission Raman
           Spectroscopy in Turbid Media Based on the Kubelka–Munk Model

      An analytical formula to the depth-of-origin profile of transmission Raman spectroscopy in turbid media was derived from the one-dimensional (1D) Kubelka–Munk model. The depth-of-origin profile of the transmitted Raman is proportional to the excitation intensity profile and the transmittance profile, which are two functions of similar forms. The effect of scattering, absorption, and signal-enhancing reflectors are incorporated into the formula. Depth-of-origin profile of a model sample was measured at better than 0.2 mm resolution and fits the formula reasonably well. Conical reflective cavities placed at the front and/or back of the sample enhanced the signal significantly; the relationship among the enhancement functions is verified by the formula. Optical parameters derived from the fitting are compared to theoretical value predicted by optical ray tracing and direct measurements; discrepancies are related to deficiency of the 1D Kubelka–Munk model.

      Brain ischemia represents a leading cause of death and disability in industrialized countries. To date, therapeutic intervention is largely unsatisfactory and novel strategies are required for getting better protection of neurons injured by cerebral blood flow restriction. Recent evidence suggests that brain insulin leads to protection of neuronal population undergoing apoptotic cell death via modulation of oxidative stress and mitochondrial cytochrome c (CytC), an effect to be better clarified. In this work, we investigate on the effect of insulin given intracerebroventricular (ICV) before inducing a transient global ischemia by bilateral occlusion of the common carotid arteries (BCCO) in Mongolian gerbils (MG). The transient (3 min) global ischemia in MG is observed to produce neurodegenerative effect mainly into CA3 hippocampal region, 72 h after cerebral blood restriction. Intracerebroventricular microinfusion of insulin significantly prevents the apoptosis of CA3 hippocampal neurons. Histological observation, after hematoxylin and eosin staining, puts in evidence the neuroprotective role of insulin, but Raman microimaging provides a clearer insight in the CytC mechanism underlying the apoptotic process. Above all, CytC has been revealed to be an outstanding, innate Raman marker for monitoring the cells status, thanks to its resonant scattering at 530 nm of incident wavelength and to its crucial role in the early stages of cells apoptosis. These data support the hypothesis of an insulin-dependent neuroprotection and antiapoptotic mechanism occurring in the brain of MG undergoing transient brain ischemia. The observed effects occurred without any peripheral change on serum glucose levels, suggesting an alternative mechanism of insulin-induced neuroprotection.

  • Coherent Anti-Stokes–Stokes Raman Cross-Correlation Spectroscopy:
           Asymmetric Frequency Shifts in Hydrogen-Bonded Pyridine-Water Complexes

      Hydrogen bonding is a vital molecular interaction for bio-molecular systems, yet deep understanding of its ways of creating various complexes requires extensive empirical testing. A hybrid femtosecond/picosecond coherent Raman spectroscopic technique is applied to study pyridine-water complexes. Both the coherent Stokes and anti-Stokes Raman spectra are recorded simultaneously as the concentration of water in pyridine varied. A 3 ps and 10 cm−1 narrowband probe pulse enables us to observe well-resolved Raman spectra. The hydrogen bonding between pyridine and water forms the complexes that have altered vibrational frequencies. These red and blue shifts were observed to be uneven. This asymmetry was result of the generated background nonlinear optical processes of pyridine-water complexes. This asymmetry tends to disappear as probe pulse further delayed attaining background-free coherent Raman spectra. For better visualization, spectral analyses both traditional two-dimensional correlation spectroscopy and recent second-order correlation functions defined in frequency domain are employed. Recognized as a label-free and background-free technique, the coherent Raman spectroscopy, complemented with a known high-resolution spectroscopic correlation analysis, has potential in studying the hydrogen-bonded pyridine-water complexes. These complexes are of great biological importance both due to the ubiquitous nature of hydrogen bonds and due to the close resemblance to chemical bases in macro-biomolecules.

  • Orientational Analysis of Monolayers at Low Surface Concentrations Due to
           an Increased Surface-to-Noise Ratio (S/N) Using Broadband Sum Frequency
           Generation Vibrational Spectroscopy

      Sum frequency generation (SFG) *Equal contributors.spectroscopy was used to deduce the orientation of the terminal methyl (CH3) group of self-assembled monolayers (SAMs) at the air–solid and air–liquid interfaces at surface concentrations as low as 1% protonated molecules in the presence of 99% deuterated molecules. The SFG spectra of octadecanethiol (ODT) and deuterated octadecanethiol (d37 ODT) SAMs on gold were used for analysis at the air–solid interface. However, the eicosanoic acid (EA) and deuterated EA (d39 EA) SAMs on the water were analyzed at the air–liquid interface. The tilt angle of the terminal CH3 group was estimated to be ∼39 ° for a SAM of 1% ODT : 99% d37 ODT, whereas the tilt angle of the terminal CH3 group of the 1% EA : 99% d39 EA monolayer was estimated to be ∼32 °. The reliability of the orientational analysis at low concentrations was validated by testing the sensitivity of the SFG spectroscopy. A signal-to-noise (S/N) ratio of ∼60 and ∼45 was obtained for the CH3 symmetric stretch (SS) of 1% ODT : 99% d37 ODT and 1% EA : 99% d39 EA, respectively. The estimated increase in S/N ratio values, as a measure of the sensitivity of the SFG spectroscopy, verified the capacity to acquire the SFG spectra at low concentrations of interfacial molecules under ambient conditions. Overall, the orientational analysis of CH3 SS vibrational mode was feasible at low concentrations of protonated molecules due to increased S/N ratio. In support, the improved S/N ratio on varying incident power density of the visible beam was also experimentally demonstrated.

  • Homogenization of Plasma Emission Collection for Multichannel

      Laser-induced breakdown spectroscopy (LIBS) has recently demonstrated its unrivaled performance for broadband elemental imaging of surfaces. The dimensions of the laser sampling spot still being potentially larger than the interfaces of chemical domains, the plasma created at each location can be largely varying and inhomogeneous with contributions from the different sides of the interface. This variation can become problematic when imaging it on fiber bundles connected to multiple spectrometers. A spatially heterogeneous signal would lead to spatially dependent image on the fiber bundle causing inconsistent readings and loss of efficiency. Köhler illumination is used in this study to create a homogenous illumination, regardless of the source homogeneity, thus improving light collection efficiency. The performance of this approach was demonstrated with inhomogeneous spectral sources and applied to the LIBS analysis of a metallic interface, showing up to a sixfold improvement of the homogeneity of the plasma collection.
