  • Evaluation of the anti-proliferative and cytotoxic potentials of fractions
           of methanol leaf extract of Cnidoscolus aconitifolius

      Abstract: The partitioned aqueous and chloroform fractions obtained from the methanol extract of the leaf of Cnidoscolus aconitifolius were examined for anti-proliferative (1-30 mg/mL) and cytotoxic activities (20-400 μg/mL) using the seed radicle inhibition and tadpole mortality assays over a period of 24 and 96 h respectively. The vacuum liquid  chromatographic sub-fractions of the active chloroform fraction weresimilarly evaluated at 1-10 mg/mL and 5-20 μg/mL concentrations respectively. At 100 μg/mL the chloroform fraction produced 86.67 % mortality which increases to 100 % at 400 μg/mL where the aqueous fraction gave 36.67 %. The potency was further increased as the VLC sub-fractions C4-C6, gave 33 % cytotoxicity at 5 μg/mL and which later increased to 100 % at 10 μg/mL. At the end of 96 h, the control seed had average length of 32.32 ± 2.73 mm compared to 1.92 ± 0.36 and 0.52 ± 0.36 mm observed in the seeds treated with 20 and 30 μg/mL of the chloroform fraction indicating 94.01 and 98.39 % inhibition where as there was total radicle growth inhibition (100 %) of seeds treated with 5 mg/mL of the active VLC sub-fractions (C1-3, C4-C6 and C7). The results showed a significant cytotoxic and growth inhibitory effects of the plant extract, particularly the chloroform fraction and its sub-fractions which further corroborate the folkloric use of the plant in treating cancer.   • Quantitative determination of chromium in some vegetables in Tudun Wada
           Makera- Kaduna, Nigeria

      Abstract: Chromium has been known to be a micronutrient for mammals for more than four decades. Deficiency in the body results to diabetes, infertility and cardiovascular diseases. However, progress in elucidating the role of chromium has proceeded slowly. Recent studies have shown a potential role of chromium in maintaining proper carbohydrate and lipid  metabolism at the molecular level. This study quantitatively determined the concentration of chromium in Moringa oleifera, Vernonia  amygdalina, , Adansonia digitata, Teiferia occidentalis, Lactuca serriola, Amaranthus tricolor and Brassica olaracea. Samples were purchased from Tudun Wada-Makera and environs for the study. The samples were digested using a tri-acid mixture (HNO3: HClO4: H2SO4 = 25:4:2). The Cr3+ content of the digests obtained was determined using TAS 990 Intec Roma model of atomic absorption spectrophotometer (AAS). The results showed significant (p<0.05) concentrations of Cr3+ ranging from 0.014 ± 0.032 to 0.008 ± 0.012 mg/g (Mean ±S.E.M) with Moringa oleifera, recording the highest concentration and Teiferia occidentalis recording the least. Chromium was detected in all the samples except Adansonia digitata.

  • Evaluating the ethyl-acetate fraction of crude methanol leaf extract of
           Ocimum gratissimum formulated as ointments for wound healing properties
           using the excision wound model

      Abstract: Ointment formulations of the ethyl acetate fraction of the crude methanol leaf extract of <i>Ocimum gratissimum</i> was in this study evaluated for wound healing activities in rat using the excision wound model. The air-dried and pulversied leaves were extracted with methanol in a Soxhlet extraction apparatus to obtain the crude methanol extract. This was  subsequently shaken with ethyl acetate and to obtain the ethyl acetate fraction, which was later dried and used in two different concentrations ( 1% w/w and 2 % w/w to formulate simple ointments. The ointments were evaluated for wound healing properties using male albino rats (65 – 180 g) aged 3 – 4 months. Prior to this, the crude methanol extract and ethyl acetate fraction were subjected to phytochemical studies to ascertain their phyto-constituents .The phytochemical studies showed the presence of alkaloids, tannins, flavonoids and resins and absence of glycosides and saponins. The ointment formulations had wound healing activities which were time-dependent, but concentration independent. The ointment formulations containing the extracts showed more effective wound healing than the positive control, Cicatrin® a standard wound healing agent, with the ointment containing 1 % w/w extract having superior effects than the batch containing 2 % w/w, implying that the extract, at 1 % w/w concentration, can be exploited in ointment form, as a potential wound healing remedy.

  • Development and validation of RP-HPLC method for identification and
           quantification of quercetin in chloroform extract of Guiera senegalensis
           leaf

      Abstract: Guiera senegalensis (Sabara in Hausa) is a shrub with numerous  medicinal values attributed to its secondary metabolites e.g. quercetin. The aim of this work is to develop and validate an RP-HPLC method for the identification and quantification quercetin in Guiera senegalensis. After several trials, the optimized condition comprises of a C18 column (250 x 4.6 mm), ternary mobile phase (Methanol: Water: acetonitrile in 55:5:45) containing 0.1 % orthophosphoric acid as additive, isocratic elution mode, 255 nm detection wavelength, flow rate of 1.2 mL/min, 10 μl injection volume, a runtime of 7 minutes and at ambient temperature. Quercetin was found to be eluted at 2.2 minute. The calibration curve was linear within the range of 10-50 μg/mL (r2 = 0.999) and was sensitive as indicated by a lower LOQ (3.15 μg/mL). The method was found to be precise, accurate and robust as indicated by their respective values which are within the acceptable limits.

  • Wound healing properties of ointment formulations of Ocimum gratissimum
           crude n-hexane bark extracts using the excision wound model

      Abstract: present work evaluated the phyto-constituents and wound healing  properties of ointments formulated with the n-hexane crude bark extract of a plant used folklorically in wound healing, Ocimum gratissimum. The excision wound model was employed in the wound healing studies. The air-dried, size-reduced barks were subjected to Soxhlet extraction using n-hexane. The resulting n-hexane extract was subjected to phytochemical screening to identify the phytochemical constituents present.  Subsequently, the extract was employed at two concentrations (1 % w/w and 2 % w/w) in the formulation of simple ointments which were applied to excision wounds inflicted on male albino rats. The wound healings were evaluated in the animals over a period of 21 days. Results obtained showed that the ointments formulated with n-hexane bark extract of Ocimum gratissimum showed superior wound healing properties  compared to those exhibited by Cicatrin®, a standard wound healing agent, thus predisposing the extract as a potential wound healing agent when formulated as ointments.
